A full-length transcriptome dataset of Takifugu obscurus for isoform-resolved analysis of complement C3 expression and splicing

ABSTRACT: The anadromous pufferfish Takifugu obscurus is an economically important aquaculture species, yet comprehensive full-length transcriptome resources remain limited. Here, we generated an integrated transcriptomic dataset using PacBio Iso-Seq long-read sequencing and Illumina RNA-seq from 12 tissues. The PacBio dataset identified 23,116 isoforms corresponding to 12,173 genes, including 846 novel genes. Approximately 60% of genes were represented by a single isoform, while the remaining genes showed multiple isoforms with diverse transcript structures. Functional annotation identified transcripts associated with diverse biological processes and pathways, including immune-related functions. Three complement C3 genes located on different scaffolds were represented by 26, 9, and 6 isoforms, respectively, with most C3 isoforms showing predominant liver-associated expression. The resulting dataset provides full-length transcript sequences, structural and functional annotations, genomic coordinates, and tissue-associated expression information, supporting isoform characterization, alternative splicing studies, and comparative transcriptomic research in T. obscurus and other teleosts.
