Bulk RNA-seq of human iPSC-derived epithelial cysts and trunk morphs

ABSTRACT: This series contains bulk RNA-sequencing data from human induced pluripotent stem cell-derived epithelial cysts and trunk morphs generated on microwell chips with localized delivery of BMP4 from coated beads. Four sets of libraries are included: (1) pluripotent cysts collected 0, 7 and 24 h after exposure to BMP4-coated beads, used to follow induction of endogenous BMP4 expression; (2) cysts after two days of differentiation in the presence of the Activin/Nodal inhibitor A83-01, and cysts differentiated without the inhibitor, used to test whether definitive endoderm is induced under the differentiation conditions used in this study; (3) sorted neuromesodermal progenitor and lateral plate mesoderm populations; and (4) cells carrying a CRISPR interference construct targeting the BMP receptor BMPR1A compared with cells carrying a scrambled guide control. Together these data characterise the transcriptional response to localized BMP signalling and the specification of lateral plate mesoderm in this model.
