Project description:We generated 4 separate subpopulations of selected flies from 1 starting population of a mixture of Canton-S flies. Two of the populations (AggrI and AggrII) were selected for increased aggression by picking males from a population cage that engaged in the most intense fighting (known as escalation behavior). Thirty males were selected every generation (from a total of 120 males per cage) per line and mated with random virgin females from that same generation. Two control populations (NeutrI and NeutrII) were selected by picking random males from the population cage after 15-30 aggressive males were removed. After 21 generations of selection males from the Aggr lines were dramatically more aggressive than males from the Neutr lines. The Neutr lines did not significantly differ from the starting population although a trend of decreased aggression was visible. Experiment Overall Design: Four populations were analyzed. Two were selected for increased aggression and two were reference groups, selected for a decrease but with no significant effect.

Project description:Genes relevant to manifestion of and variation in aggression behavior might be differentially expressed in lines selected for divergent levels of aggression. Experiment Overall Design: Drosophila males were assessed for their aggression levels in a behavioral assay that quantified aggressive encounters. A subset of the sampled population was selected as parents for the next generation, with High, Low, and Control selection groups maintained. This artificial selection was continued for 28 generations, with a variety of other behavioral and life history traits assessed for correlation with response to selection for aggression. At generation 28, male and female flies were collected for RNA extraction and subsequent gene expression analysis.

Project description:In this new study, we analyzed the blood transcriptome of horses housed individually that were first examined for their behavior, health and microbiota. We performed differential and regression analyzes of gene expression to unveil the molecular pathways related to the behavioural changes associated with housing horses in individual boxes. This study shows that aggressiveness towards humans and stereotypies are behavioural indicators that covary with physiological alterations.

Project description:Modulation of action choices according to the environmental context is crucial for animal survival and reproductive success1. Aggression, despite its general importance as a social behavior2, can become detrimental if uncontrolled3,4. However, the genetic and neural basis for aggression control through active suppression remains largely unknown. Here we found nvy, a Drosophila homolog of vertebrate myeloid translocation gene (MTG)5 involved in transcriptional regulation6-8, suppresses aggression through a specific subset of neurons. Mutation of the nvy gene resulted in hyper-aggressiveness, which was rescued by neuronal expression of human MTGs as well as Drosophila nvy. The mutant exhibited persistent aggression under various contexts in which wild-type flies switch their behavioral patterns. Knockdown of nvy in octopaminergic/tyraminergic (OA/TA) neurons increased aggression, phenocopying the nvy deletion. Cell-type specific transcriptomics on OA/TA neurons derived from the nvy mutants revealed aggression-controlling genes that are likely downstream of nvy. Moreover, we found that the nvy-expressing OA/TA neuronal subpopulations specifically suppress aggression. Our results presents a powerful genetic model to unravel not only the neuronal systems that modulate aggression in a social context-dependent manner, but also the conserved molecular mechanism underlying the tumorigenesis caused by the MTG mutations.

Project description:This study compared gene expression profiles of 16 prefrontal cortex (PFC) samples from aggressive and non-aggressive cattle breeds: Lidia, selected for agonistic responses, and Wagyu, specialized on meat production and selected for tameness. RNA sequencing was used to identify and classify 918 up and 278 down-regulated differentiated expressed genes (DEG). The functional interpretation of the up-regulated genes in the aggressive cohort revealed enrichment of pathways such as the Alzheimer disease-presenilin, integrins or the ERK/MAPK signaling cascade, all implicated in the development of abnormal aggressive behaviors and neurophysiological disorders. Moreover, gonadotropins, leading to testosterone release, are also up-regulated as a natural mechanisms enhancing aggression. Concomitantly, heterotrimeric G-protein pathways, associated with low reactivity mental states, and the GAD2 gene, a repressor of agonistic reactions at the PFC, are down-regulated, guaranteeing the development of the adequate responses required by the aggressive Lidia cattle. We also identified six upstream regulators, whose functional activity fits with the etiology of abnormal behavioral responses associated with aggression.

Project description:The identification of biomarkers indicating the level of aggressiveness of prostate cancer (PCa) addresses an urgent clinical need to minimize the general over-treatment of patients with non-aggressive PCa, which account for the majority of PCa cases. In this study we combined N-glycopeptide isolation and SWATH-MS towards the molecular characterization of tumor malignancy and aggressiveness. Here, we isolated formerly N-linked glycopeptides from normal prostate (n=10), non-aggressive (n=22), aggressive (n=16) and metastatic PCa (n=25) tumor tissues and analyzed the samples by SWATH-MS, an emerging data independent mass spectrometric acquisition method that generates a single MS file containing fragment ion spectra of all ionized species of a sample. The resulting datasets were searched using a targeted data analysis strategy where a priori spectral reference library representing known N-glycosites of the human proteome was used to identify groups of signals in the SWATH-MS data. On-average we identified 1430 N-glycosites from each SWATH map of which 1057 were quantified across all samples. The 220 glycoproteins that showed significant quantitative changes associated diverse biological processes with the level of PCa aggressiveness and metastasis and indicated functional relationships with common PCa genomic mutations.

Project description:Epistasis and pleiotropy are important features of the genetic architecture of quantitative traits, but are difficult to assess in outbred populations. We performed a diallel cross among Drosophila P-element mutations previously associated with hyper-aggressive behavior, and demonstrated extensive epistatic and pleiotropic effects on aggression, brain morphology, and genome wide gene expression. Epistatic interactions were often of greater magnitude than homozygous effects, and the topology of the epistatic networks varied among the traits. The transcriptional signature of the homozygous and double heterozygous genotypes derived from the six mutations implies a large mutational target size for aggressive behavior and evolutionarily conserved genetic mechanisms and neural signaling pathways affecting aggressive behavior. We used six mutant lines homozygous for a P-element insertion, the 15 possible transheterozygous combinations of the initial six lines and a control line. All lines were isogenic other than the P-element insert. three replicates were done for a total of 66 arrays

Project description:In humans, mutations in the transcription factor encoding gene, FOXP2, are associated with language and Autism Spectrum (ASD) Disorders, the latter characterized by deficits in social interactions. However, little is known regarding the function of Foxp2 in male or female social behavior. Our previous studies in mice revealed high expression of Foxp2 within the medial subnucleus of the amygdala (MeA), a limbic brain region highly implicated in innate social behaviors such as mating, aggression, and parental care. Here, using a comprehensive panel of behavioral tests in male and female Foxp2+/- heterozygous mice, we investigated the role Foxp2 plays in MeA-linked innate social behaviors. We reveal significant deficits in olfactory processing, social interaction, mating, aggressive and parental behaviors. Interestingly, some of these deficits displayed in a sex-specific manner. To examine the consequences of Foxp2 loss of function specifically in the MeA, we conducted a proteomic analysis of microdissected MeA tissue and found sex differences in a host of proteins implicated in neuronal communication, connectivity and dopamine signaling. Consistent with this, we discovered that MeA Foxp2-lineage cells were responsive to dopamine with differences between males and females. Thus, our findings reveal a central and sex-specific role for Foxp2 in social behavior and MeA function.

Project description:High frequency of physical aggression is the central feature of severe conduct disorder and is associated with a wide range of social, mental and physical health problems. We have previously tested the hypothesis that differential DNA methylation signatures in peripheral T cells are associated with a chronic aggression trajectory in males. Despite the fact that sex differences appear to play a pivotal role in determining the development, magnitude and frequency of aggression, most of previous studies focused on males, so little is known about female chronic physical aggression. We therefore tested here whether or not there is a signature of physical aggression in female DNA methylation and, if there is, how it relates to the signature observed in males. Genome-wide promoter methylation profiles of T cell DNA from adult females on a Chronic Physical Aggression (CPA, n=5) trajectory during childhood and adolescence were compared to those on normative physical aggression trajectories (NPA, n=14). Two technical replicates were generated for each individual.
