Molecular consequences of Csf1R (M-CSF receptor) inhibition by PLX3397 in the pilocarpine-induced chronic epileptic mouse hippocampus

ABSTRACT: Efficacy of Csf1R inhibition by PLX3397 compound in a mouse model of chronic epilepsy has been assessed by daily administration of either vehicle, 3 mg/kg or 30 mg/kg during 2 weeks monitoring. At the end of the study, RNA was extracted from study animals hippocampi to assess treatment consequences at the gene expression level in view of the in vivo efficacy results and the previously identified consequences of epilepsy in hyppocampus. Naive healthy mice were added separately to determine baseline expression.

Dorsal hippocampal transcriptome analysis after Auditory Fear Conditioning and pharmacological GR modulation

Project description:Aim: We investigated the role of GR in Auditory Fear Conditioning memory consolidation by application of a pharmacological filter using selective glucocorticoid receptor modulators. Methods: Adult male mice were exposed to Auditory Fear Conditioning training and subsequently injected with 3.0 mg/kg corticosterone, 20 mg/kg CORT108297, 80 mg/kg CORT118335, 40 mg/kg RU486 or vehicle. Dorsal hippocampi were dissected 3 hours after injection and snap-frozen. Total RNA was isolated and send for 100bp paired-end sequencing by BGI.

Transcriptome analysis using RNA sequencing of the perilesional cortex in traumatic brain injury mice treated with CSF1R inhibitor or vehicle

Project description:C57BL6 mice were subjected to a controlled cortical impact (CCI) to induce a unilateral traumatic brain injury (TBI). After surgery, mice were treated for 5 days with the CSF1R antagonist, PLX3397 or vehicle by daily i.p. injections. PLX3397 is an inhibitor of Colony Stimulating Factor receptor 1 (CSF1R) that leads to a depletion of microglial cells which was confirmed by immunohistochemistry in a subgroup of mice at day 5. Termination of PLX3397 allows for a repopulation of microglial cells which was near complete at 30 days, as assessed by immunofluorescence studies. Perilesional cortical tissue was obtained at the late time point of 30 days after TBI to assess lasting effects of microglia depletion for cortical reorganization that manifested at the transcriptional level. The RNAseq study was done in male and female mice (PLX3397 n = 5 male, 5 female; vehicle n = 5 male, 5 female). Behavioral scores for well-being and motor functions were obatined repeatedly during the observation time. Histology of brain lesion size, indices of neuronal damage and neuroinflammation were obtained at 5d and 30d after TBI. Microglia depletion (PLX3397 treatment) did not reduce the lesion volume and hindered phagocytosis and removal of the hematoma at 5 days, but reduced structural brain damage and neuron loss at 30 days after the injury. Neuroinflammatory markers tended to be lower in PLX3397 treated male than female mice which was associated with better neurological outcome. Overall, CSF1R inhibition provided only subtle lasting beneficial effects.

SVZ Study: Two-month-old male C57BL/6 (Jax 000664) were maintained on control diet for 19 days (control), treated for 14 days with PLX3397 (600 ppm) diet (microglial elimination), or treated for 14 days with PLX3397 (600 ppm) diet and then placed on control diet for 5 days (5d recovery). Mice were sacrificed and the subventricular zone was micro-dissected for RNA isolation. 28d Alt Repop +/- LPS Study: Two-month-old male C57BL/6 (Jax 000664) were maintained on control diet for 35 days (control) or treated for 14 days with PLX3397 (600 ppm) diet and then placed on control diet for 28 days (28d Recovery). At 6 or 24 hours (hr) prior to sacrifice, lipopolysaccharide (LPS; 0.33 mg/kg body weight) was administered via intraperitoneal injection. Some mice did not receive an LPS injection and serve as controls (0 hr). Mice were sacrificed and myeloid cells were extracted from whole hemispheres. RNA was extracted from single-cell suspensions generated from fluorescence-activated cell sorting (FACS) gating for CD11b+CD45int cells.

CLARITY study: Expression data from RRMS patients treated with Oral Cladribine versus Placebo at week 96

Project description:The CLARITY trial (NCT00213135) was designed as a double-blind, placebo-controlled study to allow the best comparison of absolute efficacy and safety of oral cladribine in RRMS subjects. 1326 patients with relapsing MS were randomized (1:1:1) to receive cladribine tablets 3.5 mg/kg or 5.25 mg/kg bodyweight or placebo. Gene expression data in whole blood samples at 96 weeks were prepared according to standard Affymetrix protocols Gene expression data in whole blood samples at 96 weeks were available from patients randomized to placebo (n=57), cladribine tablets 3.5 mg/kg (n=62), and cladribine tablets 5.25 mg/kg (n=70).

Glioblastoma Genetic Drivers Dictate the Function of Tumor-Associated Macrophages/Microglia and Responses to CSF1R Inhibition

Project description:Tumor-associated macrophages/microglia (TAMs) are prominent microenvironment components in human glioblastoma (GBM) that are potential targets for anti-tumor therapy. However, TAM depletion by CSF1R inhibition showed mixed results in clinical trials. We hypothesized that GBM subtype-specific tumor microenvironment convey distinct sensitivities to TAM targeting.We generated syngeneic PDGFB-driven and RAS-driven GBM models that resemble proneural-like and mesenchymal-like gliomas, and determined the effect of TAM targeting by CSF1R inhibitor PLX3397 on glioma growth. We also investigated the co-targeting of TAMs and angiogenesis on PLX3397-resistant RAS-driven GBM. Using single-cell transcriptomic profiling, we further explored differences in tumor microenvironment cellular compositions and functions in PDGFB- and RAS-driven gliomas. We found that growth of PDGFB-driven tumors was markedly inhibited by PLX3397. In contrast, depletion of TAMs at the early phase accelerated RAS-driven tumor growth and had no effects on other proneural and mesenchymal GBM models. In addition, PLX3397-resistant RAS-driven tumors did not respond to PI3K signaling inhibition. Single-cell transcriptomic profiling revealed that PDGFB-driven gliomas induced expansion and activation of pro-tumor microglia, whereas TAMs in mesenchymal RAS-driven GBM were enriched in pro-inflammatory and angiogenic signaling. Co-targeting of TAMs and angiogenesis decreased cell proliferation and changed the morphology of RAS-driven gliomas.Our work identify functionally distinct TAM subpopulations in the growth of different glioma subtypes. Notably, we uncover a potential responsiveness of resistant mesenchymal-like gliomas to combined anti-angiogenic therapy and CSF1R inhibition. These data highlight the importance of characterization of the microenvironment landscape in order to optimally stratify patients for TAM-targeted therapy.

Utomilumab in Refractory Melanoma and NSCLC (NCT01307267)

Project description:The paper reports clinical and translational results from patients with non-small-cell lung cancer (NSCLC) or melanoma dosed once every 4 weeks with either 0.24 mg/kg or 1.2 mg/kg of single-agent utomilumab, a 4-1BB/CD137 agonist mAb. The dose levels tested were well-tolerated but clinical efficacy was low. Analysis of gene expression in tumor biopsies suggested utomilumab efficacy was associated with immune-active tumor microenvironments, and pharmacodynamic activity may be reduced at higher doses. In vitro models of stimulated human T cells showed loss of pharmacodynamic activity at higher doses of utomilumab.
