RNA sequencing of Cyfip2N/- and Cyfip2N/N mice

ABSTRACT: RNA-seq analysis of 8 Cyfip2N/- and 8 Cyfip2N/N mice. Cyfip2N/- are mice contain one copy of the B6NJ missense mutation and one copy of the nonsense mutation (binge-resistant; N/-), whereas Cyfip2N/N are mice that have two mutated B6NJ allele (binge-prone; N/N), at rs240617401, a marker denoting a missense SNP in Cyfip2. Genotype identity is denoted as either J for binge-resistant; N/-, or N for binge-prone; N/N.

Deficiency of the neurodevelopmental disorder-associated gene Cyfip2 alters retinal ganglion cell properties and visual acuity

Project description:Intellectual disability (ID) is a neurodevelopmental disorder affecting approximately 0.5-3% of the population in the developed world. Individuals with ID exhibit deficits in intelligence and impaired adaptive behavior, frequently as well as visual impairments. Cytoplasmic FMR1-interacting protein 2 (CYFIP2) is an interacting partner of fragile X mental retardation protein, whose loss results in fragile X syndrome, the most common inherited cause of ID. Recently, CYFIP2 variants were found in patients with early-onset epileptic encephalopathy, developmental delay, and ID. Individuals with these CYFIP2 variants often exhibit visual impairments, however, the underlying mechanism is poorly understood. In the current study, we investigated the role of Cyfip2 in retinal and visual function by generating and analyzing mice in which Cyfip2 is depleted in the retina (Cyfip2 CKO mice). We found no major difference in the layer structure and cell composition between control and Cyfip2 CKO retinas, whereas a subset of genes associated with transporter and channel activity was differentially expressed in the Cyfip2 CKO retina. Multi-electrode array recordings showed more sustained and stronger response to positive flashes of ON ganglion cells in the Cyfip2 CKO retina, although we did not observe that Cyfip2 deficiency affects photoreceptor and ON bipolar cell functions by electroretinogram analysis. Furthermore, initial and late phase OKR analysis demonstrated that Cyfip2 deficiency impairs visual function at the organismal level. Together, our results shed light on the molecular mechanism underlying visual impairments observed in individuals with CYFIP2 variants and, more generally, in patients with neurodevelopmental disorders including ID.

Microarray of BAC transgenic Zfhx2 p.R1907K mutant mice (4 genomic copies) versus wild-types

Project description:A novel missense mutation has been identified in a human pain-insensitive family in the gene ZFHX2. We modelled this mutation in mice by changing the orthologous amino acid and carried out pain behaviour tests in mutant BAC transgenic mice. These showed a phenotype of hyposensitivity to noxious thermal stimuli. ZFHX2 has enriched expression in dorsal root ganglia neurons, and so we carried out microarray analyses in mutant vs wild type controls from lumbar DRG to see which genes were deregulated by the mutant ZFHX2 protein. To understand which genes are differentially expressed in lumbar DRG (L1-6) between Zfhx2 p.R1907K BAC transgenic mice (n=5, genomic BAC copy number of 4) and wild-type controls (n=7). Probucol ameliorates renal and metabolic sequelae of primary CoQ deficiency in Pdss2 mutant mice

Project description:To discern whether the same transcriptional signature of mitochondrial dysfunction previously reported in B6.Alb/cre,Pdss2loxP/loxP liver-conditional knockout mice (Peng, Falk et al. PLoS Genetics, 2008 [PMID 18437205]) was present regardless of Pdss2 mutation type, as well as to assess whether pharmacologic therapies modulate expression of particular pathways, genome-wide transcriptional profiling was performed in liver from B6.Pdss2(kd/kd) missense mutant mice on standard chow or supplemented long-term with either probucol or CoQ10. For analysis of lifelong probucol and CoQ10 supplementation effects in B6.Pdss2(kd/kd) missense mutant mice, total RNA was isolated from the livers of B6 mice and from Pdss2(kd/kd) missense mutant mice fed a normal diet, or a diet supplemented with Probucol or CoQ10. Total RNA was individually hybridized to 20 total Illumina Mouse WG-6 v2.0 arrays, which included 5 biological replicates from each of the 4 groupings: untreated B6.Pdss2(kd/kd) missense mutant mice, probucol-treated B6.Pdss2(kd/kd) mice, CoQ10-treated B6.Pdss2(kd/kd) mice, and untreated B6 wild-type mice. TBK1 p.E696K mutation causes autophagolysosomal dysfunction and ALS/FTD-like symptoms in mice

Project description:While deleterious mutations are responsible for the vast majority of TBK1-linked ALS/FTD cases, the ALS/FTD causing missense mutation p.E696K leads to a selective loss of TBK1/optineurin binding. Knock-in of this specific missense mutation causes progressive autophagolysosomal dysfunction and an ALS/FTD-like phenotype in mice, while, as opposed to TBK1 deletion, RIPK/TNF-α-dependent necroptosis or overt inflammation are absent. Our results highlight the role of autophagolysosomal dysfunction as a therapeutic target in TBK1-ALS/FTD.

A novel model of binge ethanol exposure reveals enhanced neurodegeneration with advanced age

Project description:Binge drinking is rising among aged adults (>65 years of age). The distinct effects of binge ethanol exposure on neurodegeneration in the aged brain are not well described. Using our model of intermittent binge ethanol exposure in young and aged mice, we investigated how binge ethanol expoure may promote neurodegenerative diease development. Spatial transcriptomics revealed that binge ethanol exposure enriched neurodegenerative disease pathways in the aged hippocampus, including tau hyperphosphorylation and neuronal death. These data suggest binge drinking during advanced aging may promote neurodegenerative disease development.
