ASCL2 is a key regulator of the proliferation-differentiation equilibrium in the esophageal epithelium

Project description:The esophagus is protected from the hostile environment by a stratified epithelium, which renews rapidly. Homeostasis of this epithelium is ensured by a rare population of stem cells in the basal layer: Keratin 15+ (Krt15+) cells. However, little is known about the molecular mechanisms regulating their distinct features, namely self-renewal, potency, and epithelial regeneration. Achaete-Scute Family BHLH Transcription Factor 2 (ASCL2) is strongly upregulated in Krt15+ stem cells and is known to contribute to stem cell maintenance in other tissues. Herein, we investigated the role of ASCL2 in maintaining homeostasis under normal and stress conditions in the esophageal epithelium. ASCL2 overexpression severely dysregulated cell differentiation and cell fate. Proliferation was also reduced due potentially to a blockage in the G1 phase of the cell cycle or an induction of quiescence. Mass spectrometry analysis confirmed alterations in several proteins associated with differentiation and cell cycle. In addition, overexpression of ASCL2 enhanced resistance to radiation and chemotherapeutic drugs. Overall, these results denoted the role of ASCL2 as a key regulator of the proliferation-differentiation equilibrium in the esophageal epithelium.

Cytokeratin 15 marks basal epithelia in developing ureters and is upregulated in a subset of urothelial transitional cell carcinomas

Project description:The mammalian ureter contains a water-tight epithelium surrounded by smooth muscle. Key molecules have been defined which regulate ureteric bud initiation and drive the differentiation of ureteric mesenchyme into peristaltic smooth muscle. Less is known about mechanisms underlying the developmental patterning of the multilayered epithelium characterising the mature ureter. In skin, which also contains a multilayered epithelium, cytokeratin 15 (CK15), an intermediate filament protein, is a marker for cells whose progeny contribute to epidermal regeneration following wounding. Moreover, CK15+ precursor cells in skin can give rise to basal cell carcinomas. In the current study, using transcriptome microarrays of embryonic wild type mouse ureters, we detected Krt15, coding for CK15.

Keratin15 Regulates Cell Proliferation in Outer Enamel Epithelium

Project description:The keratins are the typical intermediate filament proteins of epithelia, presenting with highly specific expression patterns related to the epithelial type and stage of cellular differentiation. They are important for the cytoplasmic stability and integrity of epithelia, and also involved in various intracellular signaling pathways. Several keratins have been proved to be associated with enamel formation. However, there is still lack of information on the expression patterns of keratins during tooth morphogenesis. In the present study, we analyzed the spatiotemporal expression of the keratin family members using cap analysis gene expression and single-cell RNA-sequence analysis, revealing that Krt15 and Krt17 were not only highly expressed during tooth development, but also specifically expressed in non-ameloblasts. Immunostaining of both embryotic and postnatal mouse tooth germs showed that keratin 15 was mostly expressed in outer enamel epithelium, whereas keratin 17 was mainly found in stratum intermedium. Microarray analysis discovered that a core set of proliferation related genes were upregulated in Krt15-knockdown dental epithelial cells. The results of both CCK8 assay and EdU staining indicated that the loss of keratin 15 resulted in a higher cell proliferation activity. Our findings suggested that Krt15 could be a marker gene of outer enamel epithelium, and might take a part in regulating cell proliferation of dental epithelial cells.

Expression data from airway basal cells and mucociliary-differentiated epithelium of COPD patients and normal subjects

Project description:Continuous stress caused by smoking induces changes in the cell population of small airway epithelium, with basal cell hyperplasia and goblet cell metaplasia at the expense of ciliated cells, and there is now compiling evidence that basal cells play a key role in the early pathogenesis of Chronic Obtructive Pulmonary Disease (COPD). We hypothesized that COPD airway basal cells undergo transcriptomic changes during differentiation that are different from those observed in normal cells and can explain the formation of an abnormal epithelium. We performed microarray analysis of basal cells obtained from healthy non-smoker and COPD subjects and also mucociliary-differentiated cell cultures from the same basal cells. We compared the transcriptome of normal and COPD basal cells, mucociliary-differentiated normal and COPD cell cultures and also of basal cells with corresponding mucociliary-differentiated cultures for both normal and COPD.

Project description:In this dataset, we report the analysis by RNA-sequencing (RNA-seq) of the transcriptional profile of three sub-populations of epithelial cells contained in the mouse colonic epithelium: 1) Epcam+/Cd44+/Kit-neg cells, which correspond to a population of epithelial cells located at the bottom of colonic crypts and enriched in Lgr5+ columnar basal cells (CBCs); 2) Epcam+/Cd44+/Kit+ cells, which correspond to a population of epithelial cells located at the bottom of colonic crypts and enriched in goblet cells; 3) Epcam+/Cd44-neg/Cd66-high cells, which correspond to a population of epithelial cells located at the top of colonic crypts and enriched in mature enterocytes (Rothenberg et al., Gastroenterology, 142:1195-1205, 2012).
