Project description:The fungal cell wall provides protection and structure to cells, and serves as an important target for antifungal compounds. The cell wall integrity (CWI) signaling pathway regulates the transcriptional response to various cell wall stresses, but emerging evidence suggests posttranscriptional pathways play an important complementary role. Here, we show that the RNA-binding proteins (RBPs) Mrn1 and Nab6 specifically target the 3′ UTRs of a largely overlapping set of cell wall-associated mRNAs. These mRNAs are downregulated in the absence of Nab6, indicating that Nab6 directly or indirectly stabilizes its target mRNAs. We further show that Nab6 acts in parallel to conventional CWI signaling to maintain adequate expression of cell wall mRNAs. Cells lacking both pathways are hypersensitive to antifungal compounds targeting the cell wall, while deletion of MRN1 partially alleviates the growth and molecular phenotypes associated with the Dnab6 strain. Taken together, our results uncover a novel posttranscriptional pathway which mediates resistance to cell wall stress and antifungal compounds.

Project description:Candida albicans, an opportunist fungal pathogen ranked top among those causing hospital-acquired infections, exhibits diverse virulence factors aiding its survival within the human body. The rise of multidrug resistance (MDR) in C. albicans requires urgent development of new alternative antifungal therapies. Through genetic engineering gene fusion technologies, our group developed Lectin- Fc(IgG2a) proteins (WGA-Fc(IgG) and Dectin-1-Fc(IgG)) exhibiting antifungal effects against various endemic mycosis and consisting of promising options to combat fungal infections. This study explores the direct impact of Lectin-Fc treatments on C. albicans, shedding light on potential mechanisms disrupting fungal survival and growth. Our results revealed Lectin-Fc(IgG2a) proteins' ability to reduce key polysaccharides on the fungal cell wall. Co-incubations also resulted in oxidative stress in the fungus and alterations in the yeast micronutrient content and lipid reserves. Furthermore, Lectin-Fc(IgG2a) impacted fungal secretion and impeded the release of extracellular vesicles (EVs). The proteomic analysis demonstrated differential protein expression in both the whole C. albicans yeast and EVs post-Lectin-Fc(IgG2a). Notably, these antibody-like structures are slowly degraded by secreted aspartyl proteases (Saps) in vitro, as sap mutants are more prone to growth inhibition. Finally, Lectin-Fc(IgG2a) opsonization enhanced the activation of dendritic cells, contributing to fungal killing. These findings affirm Lectin-Fc(IgG2a) proteins as viable alternatives for therapeutic interventions against fungal infections. Beyond their antibodies-like functions, they induce fungal morphological alterations, and oxidative and metabolic stress, increasing the susceptibility of their cell wall to host immune recognition and elimination.

Project description:Innate lymphoid cells (ILCs) are crucial regulators of tissue immunity, yet their role in antifungal defense remains incompletely understood. Through our findings, we demonstrate that pulmonary ILCs can sense fungal pathogens by recognizing fungal cell wall components via pattern recognition receptors, leading to the activation and type-1 polarization in vitro. Mechanistically, we identify Syk/Akt-dependent signaling as one of the key drivers of ILC activation upon fungal encounters. Aspergillus fumigatus infection reshapes this response in vivo, where elevated TGF-β and IL-1β levels facilitate a shift from type-1 ILC activity to IL-17A release. Ultimately, IL-17-producing ILCs are crucial in resolving the infection and restoring normal tissue homeostasis. Moreover, adoptive transfer models in lymphocyte-deficient Rag2-/-γc-/- mice reveal that ILCs restrain excessive inflammation while hindering fungal clearance, highlighting their dual role in regulating antifungal immunity. Remarkably, ILCs lacking the non-receptor tyrosine kinase Tec disrupts caspase-8 activation, leading to enhanced proliferation of type-1 ILCs. Consequently, improved fungal clearance enhances host survival by strengthening antifungal immunity. Our findings uncover hitherto unrecognized roles of ILCs as fungal sensors and early modulators of antifungal immunity. Therefore, targeting Tec kinase signaling in ILCs holds great promise as a therapeutic option to enhance antifungal immunity, especially in patients at risk for invasive A. fumigatus infections.

Project description:Fungal infections pose a great threat to public health and the existing four classes of antifungals have limitations due to high toxicity, drug-drug interactions, and emerging drug-resistance. Streptomyces spp. represent an important source of antimicrobial substances, notably including the antifungal agent amphotericin B. The rapamycin-producer Streptomyces iranensis displayed strong antifungal activities against Aspergillus. Revisiting its genome revealed several intriguing biosynthetic gene clusters, including one unparalleled Type I polyketide synthase, which codes for uncharacterized metabolites. The identification of a novel macrolide spirolactone (1) was facilitated through CRISPR-based gene editing, HR-ESI-MS analysis, followed by fermentation and purification processes. Their structures and absolute configurations were confirmed by NMR, MS and X-ray crystallography. Spirolactone harbors an undescribed carbon skeleton with 13 chiral centers, featuring a rare beta-lactone moiety, a [6,6]-spiroketal ring, and an unprecedented 7-oxo-octylmalonyl-CoA extender unit. Spirolactone displayed profound antifungal effects against numerous fungal pathogens, e.g. the genus Talaromyces and several sections of Aspergillus including clinically relevant species such as Aspergillus niger and A. tubingensis (section Nigri), A. terreus (section Terrei) and the azol-resistant A. calidoustus (section Usti). Proteomics analysis revealed spirolactone potentially disrupted the integrity of fungal cell walls and induced the expression of stress-response proteins in A. niger. Spirolactone represents a new class of potential agents leading to combat fungal infections.

Project description:Heat-stable antifungal factor (HSAF) isolated from Lysobacter enzymogenes has shown a broad-spectrum of antifungal activities. However, little is known about its mode of action. In this study, we used the model filamentous fungus Neurospora crassa to investigate the antifungal mechanism of HSAF. We first used HSAF to treat N. crassa strain for different time points. Spore germination, growth phenotype and differential gene expression analysis were conducted by utilizing global transcriptional profiling combined with genetic and physiological analyses. Our data showed that HSAF could significantly inhibit the germination and aerial hyphae growth of N.crassa. RNA-seq analysis showed a group of genes associated with cell wall formation and remodeling were highly activated. Screening of N. crassa gene deletion mutants combined with scanning electron microscopic observation revealed 3 fungal cell wall integrity related genes played important role in the interaction between N. crassa and L. enzymogens. In addition, WGCNA analysis, accompany with confocal microscopy observation revealed that HSAF could trigger autophagy mediated degradation and eventually result in cell death in N. crassa. The findings of this work provided new insights into the interactions between the predatory Lysobacter and its fungal prey.

Project description:Invasive fungal infections threaten the lives and health of humans, especially immunodeficient patients or hospitalized patients with serious underlying diseases, and impose a heavy economic burden on society. The emergence of drug-resistant fungi, the formation of biofilms, and the limits and side effects of existing antifungal drugs increase the difficulty of clinical treatment, and there is an urgent need for the development of novel antifungal drugs. Therefore, based on previous kinase chemical library antifungal activity screening studies, this paper further investigates the activity of the ALK inhibitor HG-14-10-04 (HG) against various fungi and elucidates its mechanism of action. The in vitro antifungal activity of HG was evaluated by micro liquid-dilution method, time-killing curve, mycelium and biofilm formation. The results showed that HG exhibited inhibitory and even fungicidal effects against sensitive and resistant Candida albicans, Candida krusei, Cryptococcus neoformans, Candida tropicalis, Candida glabrata and Candida parapsilosis (MICs=8-16 μg/mL); HG significantly inhibited the mycelium and biofilm formation, and destroyed the mature biofilm; and it exhibited synergistic antifungal effects with amphotericin B. The antifungal mechanism of HG was investigated by flow cytometry and transmission electron microscopy, etc. Sequencing analysis showed a total of 1041 differentially expressed genes, of which 666 were up-regulated and 375 were down-regulated. According to the GO functional classification results, the up-regulated genes were mainly involved in ribosome production, oxidation-reduction and other functions, while the down-regulated genes were mainly involved in the synthesis of carbohydrate, glycoproteins, glycolipids and their metabolism, GPI anchor synthesis, and cytoskeleton and other functions. In addition, HG could significantly increase the level of ROS, induce the fungal necrosis, block the cell cycle at the G0/G1 phase, and change the ultrastructure of the fungi, especially the structure of the fungal cell wall. Therefore, the enhanced inhibitory and fungicidal activity of HG may be attributed to the elevation of ROS, alteration of cellular ultrastructure (especially cell wall structure), cell cycle arrest at the G0/G1 phase, and induction of fungal necrosis, providing a basis for the discovery of novel antifungal drugs or lead compounds.

Project description:Fungal infections have increased dramatically in the last two decades and fighting infectious diseases require innovative approaches such as the combination of two drugs acting on different targets, or even target a salvage pathway of one of the drugs. The fungal cell wall biosynthesis is inhibited by the clinically used antifungal drug caspofungin. This antifungal activity has been found to be potentiated by humidimycin, a new natural product identified from the screening of a collection of 20,000 microbial extracts and with no major effect when used alone. The analysis of transcriptomes and selected A. fumigatus mutants indicated that humidimycin affects the high osmolarity glycerol response pathway. By combining humidimycin and caspofungin a strong increase of caspofungin efficacy was achieved, demonstrating that targeting different signaling pathways provides an excellent basis to develop novel anti-infective strategies.
