In vitro activity of quisinostat against Toxoplasma gondii

ABSTRACT: We performed quisinostat drug bath on Toxoplasma gondii RH strain. Next, we carried out research using data-independent acquisition (DIA) quantitative proteomics technology. A total of 4,243 proteins were identified in the four samples, all of which were quantifiable. Proteins with significant differential expression were screened based on the criteria of fold change ≥1.5 (up-regulation >1.5-fold or down-regulation <0.67-fold) and P value <0.05. Among them, 282 proteins showed significant changes in expression levels, with 77 proteins significantly up-regulated and 205 proteins significantly down-regulated. Based on the above data, systematic bioinformatics analysis was conducted for all identified proteins.

Bovine blood gamma/delta T cell stimulation study

Project description:Blood gamma/delta T cells were purfiied from holstein calves less than six months old. Blood from 4 individual calves were pooled and gamma/delta T cells were purified by immunomagnetic separation, rested in RPMI medium for 12h, and underwent ConA & hIL-2 stimulation. Total RNAs were extracted by using Trizol reagents. 15 ug of total RNA was used for cDNA synthesis and microarray hybridization. Table 1: regulated genes These genes were identified, in GeneSpring software, via filtering of genes with relative expression ratio of 1.5 and over or 0.667 or below, equivalent to up- and down-regulation by fold change 1.5 or more, in both experiments. Table 2: down-regulated genes These genes were identified, in GeneSpring software, via filtering of genes with relative expression ratio of 0.667 or below, equivalent to down-regulation by fold change 1.5 or more, in both experiments. Expression profiles and bioinformatic analysis of circRNAs in cardiac fibrosis of db/db mice

Project description:To identify the differential expression of circRNAs in cardiac fibrosis of db/db mice, db/db mice cardiac apex tissues were detected through microarray analysis at the age of 21 weeks. Compared with normal control mice, 77 circRNAs were up-regulated and 135 circRNAs were down-regulated in the chromosomes of db/db mice (fold changes ≥1.5, p-values ≤ 0.05). Moreover, Real-time qPCR was performed to validate the expression of circRNAs. In db/db mice, the top four up-regulated circRNAs, mmu_circRNA_28667, mmu_circRNA_32303, mmu_circRNA_20674, mmu_circRNA_25425 were verified. In db/db mice, the top four down-regulated circRNAs which were indexed by circBase database were verified. They were mmu_circRNA_014302, mmu_circRNA_001535, mmu_circRNA_005522 and mmu_circRNA_013422.

Project description:This project uses TMT labeling quantitative proteomics technology to carry out research, and a total of 898 proteins have been identified. Differentially expressed proteins were screened according to the criteria of expression fold change of more than 1.5-fold (up-regulation more than 1.5-fold or down-regulation less than 0.67) and P value<0.05. Among them, taking the comparison group Control VS H2O2 as an example, there were 31 up-regulated differentially expressed proteins and 81 down-regulated differentially expressed proteins. Through GO enrichment and KEGG pathway analysis, it was found that these differentially expressed proteins are mainly involved in important biological processes such as single-organism metabolic process, small molecule metabolic process, organophosphate metabolic process, organophosphate biosynthetic process and carbohydrate derivative biosynthetic process, and are mainly involved in the regulation of Metabolic pathways, Fructose and mannose metabolism, Oxidative phosphorylation, Tyrosine and Degradation of aromatic compounds and other important KEGG metabolic pathways.

Lack of hepatic response of microRNA in mice following DB[a,h]A exposure (Agilent expression)

Project description:Dibenzo[a,h]anthracene DB[a,h]A is a polycyclic aromatic hydrocarbon potent carcinogen. Few studies have investigated the role of DB[a,h]A on mRNA and miRNA expression. In this study a 10-week old male MutaTM Mouse were exposed to 6.25, 12.5, and 25 mg/kg/day DB[a,h]A by oral gavage for 28 consecutive days. DNA adducts were detected in the livers at each DB[a,h]A dose tested, and a dose-dependent increase in lacZ mutants was observed in the same samples. MAANOVA analysis revealed minor changes in the mRNA expression for the two lowest doses. Differential expression of 19 up-regulated and 22 down-regulated transcripts with fold-change > 1.5 (FDR-adjusted P < 0.05) were identified in the 6.25 mg/kg/day DB[a,h]A treatment group. For the 12.5 mg/kg/day treatment group 13 transcripts were up-regulated and 32 down-regulated (FDR-adjusted P < 0.05 and fold-change > 1.5). Major effect on mRNA expression resulted from exposure to the highest dose (25 mg/kg/day) of DB[a,h]A with 135 up-regulated and 104 down-regulated genes with fold-change > 1.5 (FDR-adjusted P < 0.05). The significantly regulated genes are involved in circadian rhythm, drug metabolism, glucose metabolism, cholestrol and lipid metabolism, immune response, cell cycle, and apoptosis. We also investigated miRNA response to the three doses of DB[a,h]A. MiRNA expression was relatively unaffected. Only miR-34a showed significant (FDR-adjusted P < 0.05) up-regulation with a fold change above 1.3-fold.

Expression profiling of lncRNAs, miRNAs and mRNAs and their differential expression in leiomyoma using next generation RNA sequencing

Project description:Total RNA was isolated from leiomyoma and paired myometrium (N=8) and samples from three pairs were subjected to RNA sequencing. After analysis, 5941 lncRNAs (2813 up- and 3128 down-regulated at ≥1.5 fold), 148 miRNAs (56 up- and 96 down-regulated at ≥1.5 fold) and 3855 mRNAs (2030 up- and 1855 down-regulated at ≥1.5 fold) were differentially expressed in leiomyomas. Using QRT-PCR we further confirmed the expression of HULC, lnc-MEG3, LINC00890, TSIX, LINC00473, lnc-KLF9-1 and lnc-POTEM-3 (lncRNA-ATB) in leiomyoma and matched myometrium (N=8). Our results presented here provide a comprehensive expression profile of lncRNAs in leiomyomas with concurrent integrated expression of miRNAs and mRNAs in leiomyomas.
