Project description:These data show differences in up- and down-regulation for protein abundances in the hippocampus of radiated vs. sham pigs after total body exposure to low dose radiation (LDR, 1.79 Gy). Tandem mass tags (TMT)-MS results showed 190 up-regulated and 120 down-regulated proteins between the two groups. These TMT-MS findings revealed changes never described before in LDR studies, with several proteins associated with neurodegenerative processes showing altered abundances. These proteomic data may further support the investigation of LDR as a potential beneficial tool to interfere with neurodegeneration and other brain-related disorders.

Project description:Diabetes is at an increased risk for development of dementia and effective treatments for diabetes-induced dementia are required with urgency. Yokukansan, a Kampo medicine, is used clinically to treat several neurological conditions including dementia. In this study, we aimed to show the effects of yokukansan on learning and memory functions in diabetes-induced dementia and to clarify the molecular mechanisms inducing the effects in the hippocampus of yokukansan- and non-treated diabetic model mice. Morris water maze test showed that yokukansan admin-istration significantly improved learning and memory functions of diabetic model mice. In the proteomic analysis, significant changes in expression of 10 (4 up- and 6 down-regulated) proteins and 8 (3 up- and 5 down-regulated) phosphoproteins were observed in the hippocampus of yokukansan-treated diabetic model mice. These identified proteins and phosphoproteins could be functionally classified as neuronal development, cellular cytoskeleton, energy metabolism, oxi-doreductase, protein deubiquitination, proton transport, and chaperone. Additionally, western blotting validated the changes in CAPZB, tubulin beta-5 chain, and LDHB. Taken together, these results showed that yokukansan improved learning and memory functions in mice with diabe-tes-induced dementia, underlying the changed expression of proteins and phosphoproteins in the hippocampus. We propose that yokukansan could be effective for care of diabetes-induced de-mentia.

Project description:The acute stress response mobilizes energy to meet situational demands and re-establish homeostasis. However, little is known about the nature and dynamics of the underlying molecular cascades. We used a brief forced swim exposure to trigger a strong stress response in mice, which transiently increases anxiety, but does not lead to lasting maladaptive behavioral changes. Using multiomic profiling we characterize the stress-induced molecular events in the hippocampus over time, across molecular scales, and down to the level of cell types and single cells. Our results unveil the complexity and specificity of the healthy stress response, with widespread changes in protein phosphorylation and gene transcription, but tightly regulated protein translation. All observed molecular events resolve efficiently within 4 hours after initiation of stress.

Project description:In this study, we used RNA-Seq to analyze the transcriptome of BK KO/WT mice hippocampus and cortex, and performed transcriptome profiling to characterize the differentially expressed genes. A total of 652 genes were screened with the threshold of significance at P < 0.05 and |log2foldchange| > 0.58, among which 382 genes were down-regulated and 270 genes were up-regulated in hippocampus tissues. In cortex tissues, we detected a total of 561 differentially expressed genes with the threshold of significance at P < 0.05 and |log2foldchange| > 0.58, including 162 up-regulated genes and 399 down-regulated genes. In order to better understand the potential functions of differentially expressed genes, Gene Ontology (GO) enrichment analysis was carried out to assess the involved pathways. Biological process pathway in GO analysis results showed that development process and biological adhesion were enriched. The results of kyoto encyclopedia of genes and genomes (KEGG) showed that insulin secretion, axon guidance, p53 signaling pathway, HIF-1 signaling pathway and calcium signaling pathway were enriched in hippocampus. On the other hand, cortical KEGG results showed that cell adhesion molecules (CAMs), ECM-receptor interaction, phagosome and calcium signaling pathway were enriched. Dysregulation of gene expression, especially astrocyte activation, neuroinflammation and autophagy, might be the molecular mechanism of BK-LOF meditated epilepsy. This study explores the possible mechanism of epilepsy by transcriptome, reveals the relationship between KCNMA1-LOF and epilepsy, and provides a possible molecular template for individualized treatment of epilepsy.

Project description:Among all voltage-gated calcium channels, the T-type Ca2+ channels encoded by the Cav3 genes are highly expressed in the hippocampus, which is associated with contextual, temporal and spatial learning and memory. However, the specific involvement of the Cav3.2 T-type Ca2+ channel in these hippocampus-dependent types of learning and memory remains unclear. To investigate the functional role of the 1H channel in learning and memory, we subjected Cav3.2 homozygous, heterozygous knockout and their wild-type littermates to hippocampus-dependent behavioral tasks, including trace fear conditioning (TFC), the Morris water-maze and passive avoidance. The Cav3.2-/- mice performed normally in the Morris water-maze and auditory trace fear conditioning tasks but were impaired in the context-cued trace fear conditioning, step-down and step-through passive avoidance tasks. Furthermore, long-term potentiation (LTP) could be induced for 180 minutes in hippocampal slices of WTs and Cav3.2+/- mice, whereas LTP persisted for only 120 minutes in Cav3.2-/- mice. To determine whether the hippocampal formation is responsible for the impaired behavioral phenotypes , we next performed experiments locally knock down function of the Cav3.2 T-type Ca2+ channel in the hippocampus. Wild-type mice infused with mibefradil exhibited similar behaviors as homozygous knockouts. Finally, microarray analyses indicated that Cav3.2-/- and WT mice presented distinct hippocampal transcriptome profiles. Taken together, our results demonstrate that retrieval of context-associated memory is dependent on the Cav3.2 T-type Ca2+ channel. After WT and Cav3.2 KO mice retrieval of context-associated memory, three right hippocampi of each group were dissected, pooled together and homogenized. The products of experimental and naive groups were used to acquire expression profiles of a total of 29,922 unique genes. Two replicates per group.
