Exercise-associated metabolomic changes in mouse prostate tumors

ABSTRACT: This study investigated exercise-associated metabolic alterations in prostate tumor tissues from Pb-Cre; Pten (fl/fl) mice. LC-MS-based metabolomic profiling was performed to compare prostate tumor tissues from control and voluntary exercise groups.

RNA sequencing of prostate tumors from control and exercise groups in a prostate-specific Pten knockout mouse model

Project description:Epidemiological studies suggest that physical activity is associated with favorable prostate cancer outcomes, but the underlying molecular mechanisms remain unclear. To investigate transcriptomic changes associated with exercise, we performed bulk RNA sequencing of prostate tumor tissues collected from prostate-specific Pten knockout mice assigned to control or voluntary wheel running conditions. These data may help clarify the molecular basis of exercise-mediated tumor suppression in prostate cancer.

Expression data from Pten, Apc, and Smad4 knockout prostate cancer mouse models

Project description:Prostate-specific deletions of tumor suppressor genes such as Pten, Apc, and Smad4 contribute to diverse molecular programs in prostate cancer. To investigate the transcriptional effects of these alterations, we generated genetically engineered mouse models with prostate-specific knockout of Pten alone or in combination with Apc or Smad4, using PB-Cre. We used microarrays to profile gene expression in tumors derived from different genotypes, comparing bulk tumor tissues and isolated cancer cells. This dataset enables investigation of gene expression changes driven by distinct oncogenic drivers and tumor cell populations in prostate cancer.

PB-Cre/Pten/Smad4 mouse prostate cancer micorarray for CRPC and lung metastasis studies

Project description:PB-Cre/Pten/Smad4 is a transgenic mouse model of metastatic prostate adenocarcinoma (PMID: 21289624). To study the transcriptomic alterations associated with castration-resistant prostate cancer (CRPC), the PB-Cre/Pten/Smad4 males with established prostate cancer were treated with surgical castration followed by enzalutamide-admixed diet. After about 4 weeks, dorsolateral prostate (DLP) lobes of treatment-naïve prostate tumors (N=2) and CRPC tumors (N=3) were harvested and extracted for RNA purification and microarray profiling. To further study the transcriptomic changes associated with lung metastases of the PB-Cre/Pten/Smad4/mTmG CRPC model, the PB-Cre/Pten/Smad4 males with established prostate cancer were treated with surgical castration followed by enzalutamide-admixed diet. About 3 months later, from one mouse anterior prostate (AP), dorsolateral prostate (DLP), ventral prostate (VP) and GFP+ lung metastasis nodules were each harvested for RNA purification and microarray profiling.

Project description:We treated the prostate tumor cell line LNCaP with serum from prostate cancer patients that participated in a 12-week physical exercise and nutrional study. Exercise was conducted in form of whole-body electromyostimulation (WB-EMS). All patients from the control and the intervention group received nutritional counseling to achieve high-protein dietary intake (please refer to Schink, Herrmann et al, 2018; PMID: 30208857). Serum from all patients was taken at study entry and at the end of the intervention in week 12. Additionally, human primary skeletal muscle cells were treated with electric pulse stimulation to simulate in vitro exercise. EPS-conditioned medium was then also used to incubate prostate tumor cells. Treatment with either study patient serum or conditioned myotube medium was done for 24 h and for 72 h. Total RNA was isolated and cancer-related gene expression was analyzed using Nanostring technology.

Voluntary exercise suppresses tumor growth through exercise-directed recruitment and intratumoral infiltration of NK cells

Project description:Voluntary exercise reduces the risk of cancer and lowers the risk of disease recurrence. Yet the mechanisms for this protection remain to be elucidated. Here we demonstrate that exercise halves tumor growth through an exercise-dependent mobilization and intratumoral infiltration of NK cells in malignant melanoma. Using voluntary wheel running, we show that exercise prior to and during B16 tumor challenge reduced tumor growth by 67%, and this reduction was associated with increased inflammation and immune cell infiltrates, especially NK cells, in the tumors from exercising mice. Depletion of NK cells blunted the exercise-dependent reduction in tumor growth. Moreover, during exercise, NK cells were engaged through an epinephrine-dependent mobilization to the circulation and redistributed to peripheral tissues through an IL-6 dependent mechanism. Project description:Oxidative stress, induced by chronic prostatic inflammation, impairs normal protective mechanisms in the prostate and increases the risk for cancer progression. The Phellodendron amurense extract (Nexrutine®) had similar effects as exercise in reducing highly aggressive tumors in the transgenic adenocarcinoma of the mouse prostate (TRAMP) model. In this study, we examined changes in the transcriptome to understand how these two interventions affect prostate cancer.

Gene expression profiling of prostate tissues and seminal vesicles from PB-Pten-NICD mice

Project description:To investigate the identity of the tissue origin for metastases, we compared gene expression profiles of the prostate tissues and seminal vesicles from PB-Pten-NICD mice and those of 7 lung metastases from different mice. Three respective cell lines established from primary seminal vesicle tumors, primary prostate tumors, and lung metastases in PB-Pten-NICD mice were also included in the microarray analysis. We hypothesized that the gene expression profiles of primary and metastatic tumors should resemble those of their tissue origins. We identified genes differentially expressed between the seminal vesicle and prostate tumors. The expression profiles of the two cell lines established from primary tumors resembled those of their respective tissue origins, which supports our hypothesis. The expression profile of the metastatic cell line resembled those of the prostate tumor tissues, indicating the prostate as its tissue of origin. The expression profiles of 2 metastatic specimens each resembled those of the seminal vesicle tumors and the prostate tumors, respectively, while the expression profiles of the remaining 3 samples displayed a mixed pattern. This study suggests that metastatic tumors in this model may have originated from either the prostate, or the seminal vesicles, or both.

Spermine is a natural suppressor of AR signaling in castration-resistant prostate cancer

Project description:In castration-resistant prostate cancer (CRPC), clinical response to androgen receptor (AR) antagonists is limited mainly due to AR-variants expression and restored AR signaling. The metabolite spermine is most abundant in prostate and it decreases as prostate cancer progresses, but its functions remain poorly understood. Here, we show spermine inhibits full-length androgen receptor (AR-FL) and androgen receptor splice variant 7 (AR-V7) signaling and suppresses CRPC cell proliferation by directly binding and inhibiting protein arginine methyltransferase PRMT1. Spermine reduces H4R3me2a modification at the AR locus and suppresses AR binding as well as H3K27ac modification levels at AR target genes. Spermine supplementation restrains CRPC growth in vivo. PRMT1 inhibition also suppresses AR-FL and AR-V7 signaling and reduces CRPC growth. Collectively, we demonstrate spermine as an anticancer metabolite by inhibiting PRMT1 to transcriptionally inhibit AR-FL and AR-V7 signaling in CRPC, and we indicate spermine and PRMT1 inhibition as powerful strategies overcoming limitations of current AR-based therapies in CRPC.
