Impact of recurrent hypoglycemia on brain metabolite profile

ABSTRACT: The goal of this study is to determine the effect of mild / moderate hypoglycemia on brain metabolism. To achieve this goal insulin-treated diabetic rats will be subjected to recurrent mild / moderate hypoglycemia.

Chronic hypercapnia-induced gene expression profiles in the brain

Project description:To test the hypothesis that increasing severity of chronic hypercapnia elicits distint gene expression profiles in important brainstem regions of cardiorespiratory control, three groups of goats (female; n=6/group) were exposed either 14 days of room air (RA), 14 days of 6% inspired CO2 (mild CH), or 7d of 6% InCO2 followed by 7d of 8% InCO2 (moderate CH). Following 14 days of exposure 4 target sites were collected (3 brainstem/1 cortical) for gene expression profiling analysis using bulk-tissue RNA-sequencing. Comparisons were made across all target sites comparing room air animals to either mild or moderate CH.

The impact of low molecular weight hyaluronan on the lipidomic profile and global gene expression of peripheral blood mononuclear cells in asthma

Project description:Hyaluronan (HA) is the major glycosaminoglycan in the extracellular matrix involved in the pathogenesis of asthma and other chronic inflammatory diseases. During inflammation there is an increased breakdown of HA, resulting in the local and systemic accumulation of low molecular weight (LMW) HA. Eicosanoids, derived from cytosolic phospholipase A2 group IVA activation, are potent lipid mediators also attributed to acute and chronic inflammation. We investigated the effect of LMW HA on lipidomic profile and global gene expression in PBMCs of patients with mild-to-moderate and severe asthma. We found that LMW HA increased production of 68 unique lipid species, among which PGE2, PGA2, PGD2, PGF2a, 15-HETE, TxB2, 11,12-EET, 14,15 EET, 13-HOTrE(y) and 16 (17) EpDPE were significantly upregulated in severe asthmatics. We also performed a systematic genome-wide expression analysis of LMW HA signaling, confirming its highly immunostimulatory potential. However, in severe asthmatics the LMW HA-induced global gene expression profile showed a comprehensive impairment in interferon signaling, cell apoptosis and cell movement, leading to diminished antiviral responses. Likewise, LMW HA-induced production of IL12 p40, CXCL10, CXCL11 and CCL8was markedly reduced in severe asthmatics. Our findings suggest a previously unforeseen link between extracellular matrix, global lipid production and antiviral responses in the pathogenesis of severe asthma. Profile of Rabbit Vocal Fold Tissue Systemically Dehydrated with Furosemide

Project description:The objective of this study was to obtain a comprehensive proteome of euhydrated and systemically dehydrated rabbit vocal folds. We hypothesized that a mild-moderate systemic dehydration level would lead to changes in protein levels. New Zealand White rabbits were randomly assigned to euhydrated (control) or furosemide-dehydrated groups. Systemic dehydration was assessed by body weight loss and changes in blood markers. Vocal fold specimens were processed for proteomic analysis using liquid chromatography-tandem mass spectrometry. Systemic dehydration induced 5.5 percent body weight loss (range: 4.5 to 6.5 percent) and significant changes in blood analytes compared to controls. More than 1500 proteins were identified across all vocal fold samples, associated with a variety of cellular components and ubiquitous cell functions such as protein synthesis and degradation, and mitochondrial translation and respiration. The comparison analysis identified 27 proteins differentially expressed in the systemically dehydrated vocal folds. These proteins are mainly involved with mitochondrial translation and metabolism, which are downregulated in response to systemic dehydration. We identified a large group of proteins representing the rabbit vocal fold tissue. The data provide novel insights on the response of vocal folds to a mild-moderate level of systemic dehydration that involves the downregulation of mitochondrial metabolism, suggesting a biologic mechanism to prevent oxidative damage associated with furosemide-induced dehydration.

Repeated hypoglycemia in diabetic mice induce hypothalamic mitochondrial and synaptic activity deregulations characteristic of neurodegenerative diseases

Project description:Repeated exposures to insulin-induced hypoglycemia in diabetic patients progressively impair the counter-regulatory response (CRR) that restores normoglycemia. This defect is characterized by reduced secretion of glucagon and other counter-regulatory hormones. Evidence indicates that glucose responsive neurons located in the hypothalamus, orchestrate the CRR. Here, we aimed at identifying the changes in hypothalamic gene and protein expression that underlie impaired CRR. High fat diet fed and low dose streptozocin-treated C57BL6N mice were exposed to one (acute hypoglycemia, AH) or multiple (recurrent hypoglycemia, RH) insulin-induced hypoglycemic episodes. Single nuclei RNA sequencing (snRNAseq) data were obtained from the hypothalamus and cortex of AH and RH mice. Proteomic data were also obtained from hypothalamic synaptosomal fractions. The present study shows that repeated moderate hypoglycemia in diabetic mice lead, in the hypothalamus, to multiple cellular physiology changes, affecting all cell types and their interactions, which show striking features of neurodegenerative diseases. It also shows that repeated hypoglycemic episodes affect very differently the hypothalamus and the cortex.
