Protein Corona of 60nm Silver Nanoparticles

ABSTRACT: We studied the composition of the protein corona formed on 60 nm silver nanoparticles with citrate coating interacted with the human blood plasma under various pH and temperature conditions. The protein corona was analyzed using LC-MS and label-free quantitation.

Plasma Protein Corona on Polystyrene Nanoparticles Facilitates their Transfer across the Human Placenta

Project description:Nanoparticles exposed to biological fluids are rapidly surrounded by proteins. It is known that this formed protein corona influences the interplay of nanoparticles with cells or tissue barriers. In this study, we report the impact of a formed human plasma protein corona on the transfer of 80 nm polystyrene (PS) nanoparticles across the human placenta. We used the human ex-vivo placental perfusion model, as it reflects the intact and physiological tissue barrier between mother and unborn. Our results show an enhanced transfer of polystyrenes, exposed to human plasma, across the placenta compared to bovine serum albumin which served as control setting. We isolated nanoparticles before and after tissue exposure and analyzed their protein corona via shotgun proteomics and LC-MS/MS. The corona profile of particles that crossed the placenta highlighted several proteins as possible drivers for elevated tissue transfer. Subsequently two distinct proteins, human albumin and immunoglobulin G, were selected and incubated with the nanoparticles to form a sole protein corona. Strikingly, the protein corona formed by albumin induced significantly the transfer of polystyrenes across the tissue as compared to corona formed by immunoglobulins. To conclude, our study provides a comparative analysis between different formed protein coronas on nanoparticles and a corona dependent transfer behavior of polystyrenes across placental tissue. Our findings suggest that protein corona analyses of nanoparticles might help to understand better their properties at biological barriers.

Project description:We are human embryologists from center for reproductive medicinel of Anhui Provincial Hospital Affiliated to Anhui Medical University, and we have the expertise to do all that properly in humans. By deep sequencing, the current experiment determined the miRNA profile of two intrafollicular somatic cell types: CRCs and COCs, isolated from women undergoing controlled ovarian stimulation and in vitro fertilization treatment. Ovarian follicles, which are a densely-packed shell of granulosa cells that contains an immature or mature oocyte, are above all responsible for the development, maturation, and release of mature egg for fertilization. They are also responsible for synthesizing and secreting hormones that are essential for follicular development, menstrual and estrous cycle, maintenance of the reproductive tracts and their functions, development of female secondary sex characteristics, and metabolism. During folliculogenesis, ovarian granulosa cells surrounding the oocyte differentiate into mural granulosa cells, involved in gonadal steroidogenesis, and into cumulus cells, which are ovulated with the oocyte at ovulation. These cumulus cells derive from the same population of early follicles, but differentiate into two distinct groups of cells: 1) Those directly lie on the zona pellucida are composed of the so called corona radiata cells.(CRCs) 2) The other group surrounds the CRCs and consists of more numerous cells, forming the so called cumulus oophorus cells (COCs). In the present study, we described the miRNA expression profile to characterize the ensemble of both known and novel miRNAs expressed in CRCs, as well as in COCs, by using high-throughput Solexa technology.

Plasma protein corona on superparamagnetic iron oxide nanoparticles (SPIONs)

Project description:We determined the composition of the plasma protein corona on silica-coated versus dextran-coated iron oxide nanoparticles using mass spectrometry-based proteomics approaches. Gene ontology (GO) enrichment analysis and Ingenuity Pathway Analysis (IPA) revealed distinct protein corona compositions for the two different SPIONs. Relaxivity of silica-coated SPIONs was modulated by the presence of a protein corona. Moreover, the viability of primary human monocyte-derived macrophages was influenced by the protein corona on silica-coated, but not dextran-coated SPIONs, and the protein corona promoted cellular uptake of silica-coated SPIONs, but did not affect internalization of dextran-coated SPIONs.

Nanoparticle Enrichment Mass-Spectrometry Proteomics Identifies Protein Altering Variants for Precise pQTL Mapping

Project description:Proteogenomics studies generate hypotheses on protein function and provide genetic evidence for drug target prioritization. Most previous work has been conducted using affinity-based proteomics approaches. These technological face challenges, such as uncertainty regarding target identity, non-specific binding, and handling of variants that affect epitope affinity binding. Mass spectrometry (MS)-based proteomics can overcome some of these challenges. Here we report a pQTL study using the Proteograph™ Product Suite workflow (Seer, Inc.) where we quantify over 18,000 unique peptides from nearly 3,000 proteins in more than 320 blood samples from a multi-ethnic cohort in a bottom-up, peptide-centric, MS-based proteomics approach. We identify 184 protein-altering variants (PAVs) in 137 genes that are significantly associated with their corresponding variant peptides, confirming target specificity of co-associated affinity binders, identifying putatively causal cis-encoded proteins and providing experimental evidence for their presence in blood, including proteins that may be inaccessible to affinity-based proteomics.

Combining confocal microscopy, dSTORM, and mass spectroscopy to unveil the evolution of the protein corona associated with nanostructured lipid nanocarriers during blood-brain barrier crossing

Project description:Upon entering in contact with the biological environment, nanostructures are immediately covered by biomolecules, and in particular proteins forming the so-called “protein corona” (PC). The phenomenon of PC formation has gained great attention in recent years due to its implication for the use of nanostructures in biomedicine. In fact, it has been shown that the formation of the PC can impact the performance of nanostructures by reducing their stability, causing aggregation, increasing their toxicity, and causing unexpected and undesired nanostructure-cell interactions. In this work, we decided to study for the first time the formation and the evolution of PC on the surface of nanostructured lipid carriers loaded with superparamagnetic iron oxide nanoparticles (SPIONs), before and after the crossing of an in vitro model of the blood-brain barrier. Combining confocal microscopy, direct STochastic Optical Reconstruction Microscopy (dSTORM), and proteomic analysis, we were able to provide a complete analysis of the PC formation and evolution. In particular, we evidence that PC formation is a fast process, being formed around particles even after just 1 min of exposure to fetal bovine serum. Moreover, PC formed around particles is extremely heterogeneous: while some particles have no associated PC at all, others are completely covered by proteins. Lastly, the interaction with an in vitro blood-brain barrier model strongly affects the PC composition: in particular, a large amount of the proteins forming the initial PC is lost after the BBB passage and is partially replaced by new proteins derived from both the brain endothelial cells and from the cell culture medium. Altogether, the presented data could potentially provide new insights into the design and fabrication of lipid nanostructures for the treatment of central nervous system disorders.

Revealing the immune perturbation of black phosphorus nanomaterials to macrophages by understanding the protein corona

Project description:The increasing number of biological applications for black phosphorus (BP) nanomaterials has precipitated considerable concern about their interactions with physiological systems. Here, we demonstrate the adsorption of plasma protein onto BP nanomaterials and the subsequent immune perturbation effect on macrophages. Using liquid chromatography tandem mass spectrometry, we discovered that BP nanosheets (BPNSs) were able to bind 23.3 percent of immune proteins from plasma, while BP quantum dots (BPQDs) bound 41.8 percent of immune proteins. In particular, the protein corona dramatically reshaped BP nanomaterial-corona complexes, influenced cellular uptake, activated the NF-κB pathway and even increased cytokine secretion by 2-5-fold. BP nanomaterials induced immunotoxicity and immune perturbation in macrophages in the presence of a plasma corona. These findings offer important insights into the development of safe and effective BP nanomaterial-based therapies.

Global genetic impact of surface-modified gold nanoparticles on two model human cell lines

Project description:Gold nanoparticles (Au NPs) are uniquely suited for various biomedical applications due to the combination of their optical properties with their easily functionalized surfaces. The Au NP surface can be tailored to improve biocompatibility while also attaching targeting ligands or drugs. However, information on how these tailored surface chemistries may affect cell gene expression is scarce. Using two model human cells line, human dermal fibroblasts and prostate cancer cells, microarray experiments measured gene expression over 27,000 human genes. Each of the cell lines was exposed to four related types of surface-modified Au NPs at two different concentrations, and the microarray data was analyzed by weighted gene correlation network analysis and gene functional annotation. Au NPs were shown to affect genes associated with a variety of cellular functions, and surface charge and chemistry were linked with the types of parthways changed and the degree of which those changes occured. Nanoparticle induced gene expression in PC3 and HDF cells was measured after 24 hour exposure to nanoparticles of four different surface coating types. RNA from three separate culture samples were used for each nanoparticle-cell combinations, along with three control samples not exposed to nanoparticles at all.
