DIA-PASEF Proteomic Profiling of Metabolic Regulation of Extracellular Vesicle Uptake

ABSTRACT: This project uses untargeted proteomics to investigate how cellular metabolic state and metabolite adjuvants regulate extracellular vesicle (EV) uptake in neuroblastoma cells. Proteomic profiling was performed on cells cultured in in vivo–like medium (HPLM) or standard medium, as well as on HPLM-conditioned cells treated with or without metabolite adjuvants. Comparative pathway analyses identified metabolic rewiring under physiologic conditions and revealed upregulation of endocytosis-associated pathways following metabolite supplementation. These proteomics data support a model in which modulation of the cellular metabolome enhances EV uptake primarily through increased endocytic activity.
