Candida utilis as a protein source for Atlantic salmon

Project description:Single cell proteins, such as Candida utilis, are known to have immunomodulating effects in the distal intestine (DI) of Atlantic salmon, whereas soybean meal (SBM) can cause soybean meal induce enteritis (SBMIE). Inflammatory or immunomodulatory stimuli at the local level in the intestine may alter the plasma protein profile of Atlantic salmon. These changes can be helpful tools in diagnosis for fish diseases and indicators for fish health. The present work aimed to identify local intestinal tissue responses and changes in plasma protein profiles of Atlantic salmon fed C. utilis yeast, SBM, or combined diets. Fish meal (FM) based diet was used as a control diet and the six experimental diets were: FM diet with 200 g/kg C. utilis (FM200CU) and five diets containing 200 g/kg SBM together with 0 (SBM group), 25, 50, 100 or 200 g/kg C. utilis (SBM25CU, SBM50CU, SBM100CU and SBM200CU groups, respectively). Intestine morphology of fish fed FM200CU where not affected whereas SBM group presented changes characteristic of SBMIE. Low inclusion of C. utilis in SBM diet showed a modulation of immune cell populations, but did not alleviate inflammatory symptom.

Dietary soyasaponin supplementation to pea protein concentrate reveals nutrigenomic interactions underlying enteropathy in Atlantic salmon

Project description:Atlantic salmon was fed with diets based on five plant protein sources combined with soyabean saponins. Diets with corn gluten, sunflower, rapeseed and horsebean produced minor effects while combination of saponins with pea protein concentrate caused enteritis and major transcriptome changes in the distal intestine. Microarray analyses were perfromed in distant intestine. Individual samples from fish that received saponing were hybridized to pools from salmon fed with the same feeds without saponins.

The farmed Atlantic salmon (Salmo salar) skin-mucus proteome

Project description:Norway is the largest producer and exporter of farmed Atlantic salmon (Salmo salar) worldwide. Skin disorders correlated with bacterial infections represent an important challenge for fish farmers due to the economic losses caused. Little is known about this topic, thus studying the skin-mucus of Salmo salar and its bacterial community depict a step forward in understanding fish welfare in aquaculture. In this study, we used label free quantitative mass spectrometry to investigate the skin-mucus proteins associated with both Atlantic salmon and bacteria. In addition, the microbial temporal proteome dynamics during 9 days of mucus incubation with sterilized seawater was investigated, in order to evaluate their capacity to utilize mucus components for growth in this environment.

Changes in the liver transcriptome of farmed Atlantic salmon (Salmo salar) fed diets based on terrestrial alternatives to fish meal and fish oil

Project description:After a 14-week feeding trial, Atlantic salmon fed a diet based on animal by-product meal and vegetable oil (ABP) showed lower growth rates compared to others fed an FM/FO-based diet (MAR). Interestingly, feeding a vegetable meal/oil-based diet (VEG) resulted in growth rates similar to the FM/FO-based diet. A 44K microarray experiment conducted on liver total RNA samples from these three dietary groups identified 46 genes related to metabolic and growth-relevant pathways, 25 immune-related, and 12 related to oxidation-reduction processes.

Dietary cholesterol supplementation to a plant-based diet for Atlantic salmon

Project description:The study evaluated effects of dietary cholesterol (1.5%) in Atlantic salmon fed a plant based diet for 77 days. Cholesterol supplementation did not affect growth or organ weights of Atlantic salmon, but promoted induction of cholesterol and plant sterol efflux in the intestine, whereas sterol uptake was suppressed. Microarray analyses in the liver indicated decreased cholesterol biosynthesis and enhanced conversion to bile acids. The marked effect of cholesterol on bile acid synthesis suggests that dietary cholesterol can be used to stimulate bile acid synthesis in fish. The study clearly demonstrated how Atlantic salmon adjusted metabolic functions in response to the dietary load of cholesterol, and has expanded our understanding of sterol metabolism and turnover that adds to the knowledge of these processes in fish. Atlantic salmon received feeds based on plant ingredients with (CH) and without (K) supplementation of cholesterol. Liver samples were collected after 77 days. Five individuals from each group were analyzed with microarrays, pooled liver sample of salmon fed with commerical fish meal based feed was used as a reference.

Functional genomic analysis of the impact of camelina (Camelina sativa) meal on Atlantic salmon (Salmo salar) distal intestine gene expression and physiology

Project description:Fish were fed a standard fish meal (FM) diet or a diet with partial replacement of FM with solvent extracted camelina meal (CM) (8%, 16% or 24% CM inclusion) during a 16-week feeding trial. A significant decrease in growth performance was seen in fish fed the CM inclusion diets. A 44k oligonucleotide array experiment was used to identify any differentially expressed transcripts in the distal intestine of the fish fed the 24% CM diet compared to the control. The expression level of these genes was validated using quantitative polymerase chain reaction, which was also used to measure transcript expression in the fish fed the 8% CM and 16% CM diets. Histopathological analysis was used to quantify any physical signs in inflammation in the distal intestine of the Atlantic salmon fed the CM-containing diets.

Feed supplementation with choline improved lipid absorption and suppressed abnormal accumulation of fat in the gut.

Clues from the intestinal mucus proteome of Atlantic salmon to counter inflammation

Project description:Intestinal inflammation in Atlantic salmon was studied by profiling the intestine mucus proteome, employing isobaric tags for relative and absolute quantification and 2D LC-MS/MS approach. Two fish groups were fed soy saponin-containing (inflammation inducer, SO) or devoid (CO) diets for 36 days. Two more fish groups were fed each of the aforementioned diets with a health additive (CP, SP). Inflammation characteristics in the intestine were milder in the SP-fed fish compared to the SO-fed fish, based on histology results. The fish that had intestinal inflammation (SO) was characterised by alterations of many proteins. Based on the enriched KEGG pathways and GO terms of the SO vs CO comparison pathways such as phagosome and lipid binding were affected due to upregulation of protein like Integrin beta 2 precursor, Coronin 1A, Cathepsin S precursor, Vesicle-trafficking protein, and Neutrophil cytosol factors. On the other hand, the SP group had fewer inflammation characteristics and altered proteins; this fish group had higher abundance of protein linked to the pathways aminoacyl-tRNA biosynthesis and ribosome due to the higher abundance of many large and small subunit of ribosomes. The alteration of Glycogen phosphorylase and Glutamin synthetase could also have alleviated intestinal inflammation in the SP group. Elevating the abundance of ribosomal proteins, aminoacyl-tRNA ligases, and lowering the abundance of Glycogen phosphorylase and Glutamine synthetase could be effective to alleviate intestinal inflammation. The proteins described here can be exploited to alleviate intestinal inflammation in fish and higher vertebrates.
