Project description:Epigenetic changes such as DNA methylations regulate gene expression patterns in response to environmental cues including infections. Microbial infections induced DNA methylation may play a potential role in modulating host-immune response. In the present study, we sought to determine DNA methylation changes induced by the mastitis causing Escherichia coli (E coli) in porcine mammary epithelial cells (PMEC). Two time points (3hr and 24 hr) were selected based on the specific transcriptomic changes as early and late phase immune responses. Genome-wide methylation information was obtained to identify significant differential methylation patterns in E coli infected PMEC.

Project description:Sow milk yield and quality is crucial for the survival and growth of piglets. Mammary epithelial cell proliferation, differentiation, milk component production and galactopoietic hormone and cytokine synthesis result from the quantities of differentially expressed genes. This study used the Affymetrix Porcine GeneChip to acquire a comprehensive picture of transcriptional adaptations in the mammary glands of lactating sows. Mammary tissue samples were collected from six sows at -17, 1 and 17 d relative to parturition. Mammary tissues from two sows were used to extract RNA and were subsequently pooled in equal volumes. Nine pooled samples were hybridized to porcine Affymetrix GeneChips.

Pregnancy-associated alterations in DNA methylation patterns of mammary epithelial stem cells [inhibitor study]

Project description:Mammary gland development and luminal differentiation occur largely postnatally during puberty and pregnancy. To explore the role of DNA methylation in luminal cell differentiation and pregnancy-induced changes, we determined the genome-wide DNA methylation and gene expression profiles of mammary epithelial stem, luminal progenitor, and mature luminal cells at different reproductive stages. We found that pregnancy had the most significant effects on stem cells, inducing a distinct epigenetic state that remained stable through life. Integrated analysis of gene expression, DNA methylation, and histone modification profiles revealed cell type and reproductive stage-specific changes in molecular signatures. We also identified p27 and TGFβ signaling as key regulators of luminal progenitor cell proliferation based on their expression patterns and by the use of explant cultures. Our results suggest relatively minor changes in DNA methylation during luminal cell differentiation as compared to the significant effects of pregnancy on mammary epithelial stem cells. Mammary glands were collected from mice treated with DNA methylation inhibitor 5-azacytidine (AzaC) and histone deacetylase inhibitor valproic acid (VPA) at non-pregnant and pregnant stages for RNA extraction and hybridization on Affymetrix microarrays.
