ABSTRACT: this study aims to optimize DNA extraction protocols for ONT sequencing and to explore the practical implications of implementing MinION sequencing as a tool to combat AMR in resource-limited settings. The study will evaluate four commercial kits on both Gram-positive and Gram-negative bacterial isolates, representing different lysis strategies, i.e., enzymatic, chemical, and bead-beating, and purification strategies, i.e., silica spin-column, magnetic bead-based, and glass bead-based.
