Project description:We have observed that the expression of IFIT1, IFIT2, IFIT3 and the other genes related interferon are all elevated by GLP1R knockdown whereas reduced by GLP1RA and 2GRA in multiple lineages differentiation systems of PDLSCs. It was reported that IFIT proteins could block the formation of 43S pre-initiation complex and 48S initial complex via interacting with eIF3E and eIF3C. RIP-seq was conducted to study the transcriptome-wide occupancies of IFIT1 and eIF3C in osteogenic PDLSCs treated with GLP1R knockdown compared to regular osteogenesis.

Transcriptome sequencing of PDLSCs induced by GIPR and GLP1R alteration

Project description:Our previous study (Environ Pollut. 2023;323:121307) has indicated that Glucose-dependent insulinotropic polypeptide receptor (GIPR) and Glucagon-like peptide-1 receptor (GLP1R) are down-regulated in periodontal ligament stem cells (PDLSCs) by cigarette and E-cig. In the current study, we focus on investigating the functional roles of these two G protein-coupled receptors in PDLSCs, as well as their molecular mechanisms behind it. In brief, human PDLSCs treated with GIPR-siRNA, GIPR agonist (GIPRA) as well as GIPR, GLP1R dual agonist (2GRA) were used to study the effects of GIPR on cell growth of PDLSCs. Moreover, PDLSCs were induced to undergo osteogenic, adipogenic, chondrogenic, and neurogenic differentiation in vitro. GLP1R siRNA, GLP1R agonist (GLP1RA), and 2GRA were respectively administered to investigate the effects of GLP1R on the differentiation potential of PDLSCs.

Genome-wide occupancies of multiple transcription activators in PDLSCs induced by GIPRA

In the current study, ChIP-seq was conducted to study the genome-wide occupancies of phosphorylated CREB (p-CREB), p-STAT, p-ERK and TCF4 in PDLSCs treated with GIPR agonist (GIPRA) (Pro3) GIP (0.5 nM, 24 h, Cat. No. HY-P3584, MCE, China) and ERK1/2 inhibitor SCH772984 (0.5 μM, 24 h, HY-50846, MCE) compared to normal PDLSCs.

RNA sequencing of human periodontal ligament stem cells (PDLSCs) with Illumina HiSeq2000

Project description:To study the molecular mechanisms underlying PDLSC osteogenic differentiation, we developed RNA sequencing with Illumina HiSeq2000 to comprehensively identify RNA expression profiles in normal PDLSCs, osteogenic inductive PDLSCs and mechanical force-induced PDLSCs. Plenty of RNAs were found to be differentially expressed by RNA sequencing. Furthermore, the potential functions of these RNAs were investigated using bioinformatic analysis.

Controlling tissue patterning by translational regulation of signaling transcripts through the core translation factor eIF3c [II]

Project description:Although gene expression is tightly regulated during embryonic development, the impact of translational control has received less experimental attention. Here, we find that eukaryotic translation initiation factor-3 (eIF3) is required for Shh-mediated tissue patterning. Analysis of loss-of-function eIF3 subunit c (Eif3c) mice reveal a unique sensitivity to the Shh receptor Patched 1 (Ptch1) dosage. Genome-wide in vivo enhanced cross-linking immunoprecipitation sequence (eCLIP-seq) shows unexpected specificity for eIF3 binding to a pyrimidine rich motif present in subsets of 5’-UTRs and a corresponding change in the translation of these transcripts by ribosome profiling in Eif3c loss-of-function embryos. We further find that while Eif3c loss-of-function embryos do not show a global decrease in protein synthesis, translation of Ptch1 through this pyrimidine rich motif is specifically sensitive to eIF3 amount. Altogether, this work uncovers hidden specificity of housekeeping translation initiation machinery for the translation of key developmental signaling transcripts.
