Project description:This study is to investigate the potential role of Prdm1 in murine intestinal epithelial cells (IEC4.1 cells) in response to IFN gamma stimulation. Prdm1 was knocked down in IEC4.1 cells by using a CRISPR/Cas9 approach. Wild type IEC4.1 cells were used as control (Ctrl). Cells were treated with or without mouse IFN gamma (1ng/ml for 4hrs). Then total RNA was collected for sequencing via DNBSEQ platform to obtain a comprehensive view of the transcriptome.

Transcriptomic profile of IEC4.1 cells treated with an ASO to lncRNA XR_001779380 in response to IFN-gamma

Project description:LncRNAs may play an important role in intestinal epithelial cell autonomous defense against microbial infection. Our previous study showed that lncRNA XR_001779380 can interact with Prdm1 to modulate gene transcriuption in intestinal epithelial cells and may regualte IFN-gamma-stimulated gene transcription. Here we designed an antisense oligos-morpholine (ASO4) targeting the sequence of lncRNA XR_001779380 to interfere with its interaction with PRDM1. Murine intestinal epithelial cells (IEC4.1 cells) were treated with ASO4 or a standard control ASO-morpholine (ST) for 24h, followed by exposure to IFN-gamma (1 ng/ml) for 4 hours. Then total RNA was collected for sequencing via DNBSEQ platform to obtain a comprehensive view of the transcriptome.

Project description:This study is to investigate the potential impact of an lncRNA NR_126553 (Nostril) in murine intestinal epithelial cells (IEC4.1 cells) in response to IFN-gamma protein (IFNγ) stimulation. NR_126553 (Nostril) was knocked down by using a pool of gene specific siRNA (SiNostril). Cells treated with scramble non-specific siRNA were used as control (siNegative control). After siRNA transfection 24h, cells were treated with or without mouse IFNγ (1ng/ml for 4hrs). Then total RNA was collected for sequencing via DNBSEQ platform to obtain a comprehensive view of the transcriptome.

The underlying mechansims of host immune regulation remains obscure and increasing evidence has shown long non coding RNAs (lncRNAs) can regulating gene expression at both the transcriptional and posttranscitpional levels. In the current study, we analyzed the transcriptome of murine intestinal epithelial cells (IEC4.1 cells) or stable CRISPR/cas9 U90926KO IEC4.1 cells following treatment with IFNγ to identify potential immune gene tragets of this lncRNA. Total RNA was collected for the genome-wide analysis, for sequencing using DNBSEQ platform.

Interferon (IFN)-γ promotes monocyte-mediated lung injury during influenza pathogenesis

Project description:Influenza A Virus (IAV) triggers an exuberant host response that promotes acute lung injury. However, the determinants of the pathological host response to IAV remain incompletely understood. In the current study, we identified interferon (IFN)-γ-regulated subset of monocytes, CCR2+ monocytes, as a driver of lung damage during IAV pathogenesis. IFN-γ regulated the recruitment and inflammatory phenotype of CCR2+ monocytes, and CCR2 (CCR2-/-) and IFN-γ (IFN-γ-/-) deficient mice exhibited reduced lung inflammation, pathology, and increased resistance against bacterial co-infection by Streptococcus pneumoniae (Spn). Adoptive transfer of WT (IFN-γR1+), but not IFN-γR1 deficient (IFN-γR1-) CCR2+ monocytes, restored the wild-type (WT)-like pathological phenotype of lung damage in IAV-infected CCR2-/- mice. The CD8+ T cells were the most significant source of IFN-γ in IAV-infected lungs. Collectively, our data highlight that IFN-γ regulates CCR2+ monocyte-mediated lung pathology during IAV pathogenesis.

T-bet-depended but IFN-gamma-independent genes in GVHD

Project description:Given T-bet is a master regulator for Th1 differentiation that produce IFN-gamma, underlining mechanisms accounted for the distinct GVHD outcomes caused by T-bet- versus IFN-gamma-deficient donor T cells are unclear. We hypothesize that GVHD is regulated by T-bet-dependent but IFN-gamma-independent molecules. To identify these molecules, we isolated donor T cells from spleen cells of the recipients that were transplanted with WT, T-bet-/- or IFN-gamma-/- CD4 T cells for 7 days. The gene profile of isolated T cells was measured by microarray analysis using GeneChip Mouse Genome 430 2.0 arrays (Affymetrix) according to the manufacturerM-^Rs instructions. Array images were analyzed and processed by robust multi-array average (RMA) procedure.
