Project description:Although therapies for atherosclerotic cardiovascular disease (ASCVD) have advanced, substantial residual risk of cardiovascular (CV) events remains. Lipoprotein (a) [Lp(a)] is a low-density lipoprotein (LDL) particle that is causally linked to both ASCVD and CV events. Lp(a) is thought to drive ASCVD through multiple mechanisms including its effects on cholesterol accumulation, inflammation, and thrombosis. Although the associations between Lp(a) and ASCVD drivers are clear, the mechanisms that integrate Lp(a)-mediated cholesterol accumulation, inflammation, and thrombosis remain largely unknown. In this study, we employed systems biology approaches consisting of proteomics, transcriptomics, and mass cytometry to define the immune cellular and molecular phenotypes in ASCVD subjects with high and low Lp(a) levels and the mechanisms through which Lp(a) mediates monocyte activation. In a cohort of stable ASCVD subjects (n=64 [41 with high Lp(a) and 23 with low Lp(a]), we found that circulating markers of inflammation (CCL28, IL-17D) and vascular dysfunction (BNP), tissue factor [TF]) was elevated in subjects with high Lp(a) levels compared with those with low Lp(a) levels. Additionally, although total monocytes levels and hs-CRP levels were similar between the groups, CD14+ monocytes from ASCVD subjects with an elevated Lp(a) were primed and expressed more TF at baseline and in response to stress. Mechanistically, we found that Lp(a) itself can activate monocytes through Toll-like receptors (TLR) and nuclear factor kappa B (NFκB) signaling, driving both the induction of TF and TF activity. Overall, these studies are the first to link Lp(a) to monocyte-mediated inflammation and thrombosis, demonstrating a novel mechanism through TLR2, NFκB, and monocyte TF which Lp(a) amplifies immunothrombotic risk.

Project description:The “quality” rather than “quantity” of lipoprotein particles is more relevant to cardiovascular diseases (CVD), since lipoprotein particles are highly heterogeneous. However, the targeted separation, detection, and exploration of the heterogeneous on omic-scale for lipoprotein remain challenging and rarely touched. Here, we established a high-resolution proteomics and lipidomics (HiPL) method. Briefly, the lipoprotein particles were separated into nine fractions (Lipo-HiPL) using anion-exchange chromatography (AEC), in which HDL, LDL, and VLDL were fractionated into three more fractions respectively. Furthermore, the HDL subclasses were delicately purified within nine fractions (HDL-HiPL), covering diverse protein-lipid heterogeneity. Integration of proteomic and lipidomic analysis revealed proteome-lipidome connectivity (PLC) in lipoprotein and HDL particles, which highly correlated with the atherosclerotic phenotype in rabbit model. Importantly, the lipoprotein “quality” revealed by PLC was found to be related more closely to clinical status in acute coronary syndrome patients than direct proteome and lipidome quantification. Thus, proteome-lipidome connectivity (PLC) as novel HDL “quality” features could be highly associated with atherosclerotic cardiovascular diseases.

One of the leading risk factors for development of atherosclerosis is familial hypercholesterolemia. Familial hypercholesterolemia is an autosomal dominant disorder, which is caused by mutations mainly located in the low-density lipoprotein receptor (LDLR) gene. It is characterized by elevated levels of low-density lipoprotein cholesterol, the presence of tendon xanthomas, and premature cardiovascular disease. Aim of this study was to find atherosclerotic markers in white blood cells of patients compared to healthy controls. None of these patients exhibited symptoms of atherosclerosis by standard diagnostic methods; however, transcriptome analysis of blood RNA indicated changes in ubiquitin proteolysis and cell adhesion system as expected in initiation steps of atherosclerosis.

Project description:Atherosclerotic cardiovascular disease remains a major cause of death worldwide. Autoimmunity to low-density lipoprotein (LDL) has been implicated in the disease process and immunization with LDL components proposed as a preventive strategy. However, mechanisms of atheroprotective immunity remain insufficiently characterized. Two different T cell receptor transgenic mouse strains with T cells reactive against the LDL protein, apoB100 (BT1 and BT3) were crossbred with human APOB100-transgenic Ldlr-/- (HuBL) mice, and atherosclerosis was evaluated. In 52-week-old BT1xHuBL mice, a lower atherosclerotic burden was observed in the aorta, mirrored by reduced plasma cholesterol and elevated anti-apoB100 antibodies. BT3xHuBL mice also exhibited a decrease in aortic atherosclerosis; however, no reduction in plasma cholesterol levels was noted. Instead, type 1 regulatory T cell responses with interleukin (IL)-10 production were mounted. This response was associated with reduced collagen content and a lower stability index of the atherosclerotic lesions. In human carotid artery plaques, IL10 mRNA levels were negatively correlated with collagen expression, and IL-10 was found to inhibit collagen production in vascular smooth muscle cells. We conclude that atheroprotective apoB immunity elicits two distinct pathways: lipid-lowering immune reactions and local anti-inflammatory IL-10 production. Since the latter is associated with decreased plaque stability and cardiovascular events, treatments aimed at increasing IL-10 signaling over extended periods to reduce vascular inflammation need reconsideration.

Project description:Tirzepatide (TZP) is a novel dual agonist of glucagon-like peptide -1/glucose-dependent insulinotropic polypeptide receptors. TZP represents a significant advance in the treatment of diabetes and obesity. Clinical trials have shown that TZP significantly reduces cardiovascular risk in patients with diabetes or obese patients who are at high CV risk. We report here that TZP exerts anti-atherosclerotic effects by altering macrophage polarization and inflammatory expression. Under oxLDL, the expression of inflammatory factors in RAW264.7 cells increased, and the phenotype of M1 macrophages increased. After TZP treatment, M2 macrophages increased and anti-inflammatory factor expression increased in RAW264.7 cells. In summary, our data provide a new mechanism for TZP in the treatment of oxLDL-induced foam cell formation.
