Project description:Enteric nervous system (ENS) development requires migration, proliferation, and neuronal diversification of progenitors to achieve normal gastrointestinal motility. Sox10 deficits disrupt the ratios of mature enteric neuron subtypes. How Sox10 deficiency alters the proportions of neuronal subtypes is still being determined. Sox10's prominent expression in neural crest-derived enteric progenitors and lack of this gene in enteric neurons led us to investigate how Sox10 effects in early enteric progenitors could alter neuronal allocation. To explore this further, enteric progenitors, developing neurons, and enteric glia were isolated from Sox10+/+ and Sox10Dom/+ littermates at 15.5 days post-coitus based on dual Sox10-H2BVenus and Phox2b-H2BCerulean transgene expression for single-cell RNA sequencing (scRNA-seq). The data was processed in R to identify cell-specific markers, differentially expressed genes, biological processes associated with gene ontology, cell fate trajectories, and gene regulatory network activity between genotypes. Hybridization chain reaction (HCR) validated expression changes detected in scRNA-seq.

Single-cell RNA sequencing of small intestine neurons in Hirschsprung disease mice

Project description:This study was undertaken to identify gene expression changes in enteric neurons of Hirschsprung disease mice. Methods: We performed single-cell RNA sequencing of enteric nervous system cells isolated from the small intestine of Ednrb-knockout mice at age P14. Results: Through comparisons with published datasets of WT small intestine enteric neurons, we identify a missing neuronal population in the ganglionated small intestine of Hirschsprung disease mice. Conclusion: Single-cell RNA sequencing permits identification of cellular perturbations in Hirschsprung disease models.

Transcriptomic profiling of aganglionic and ganglionic colon segments from Hirschsprung disease patients

Project description:This dataset contains bulk RNA sequencing data from paired aganglionic and ganglionic colonic tissue specimens obtained from three pediatric patients diagnosed with Hirschsprung disease (HSCR, OMIM 142623). RNA was extracted and sequenced to investigate transcriptomic alterations and signaling pathway dysregulation associated with HSCR pathogenesis. Raw paired-end FASTQ files generated by Illumina NovaSeq 6000 sequencing are provided for each sample, enabling downstream analyses of differential gene expression between diseased and unaffected intestinal segments.

Chromatin plasticity contributes to neuronal selection during memory allocation

Project description:Memory allocation refers to the process by which neurons are recruited into the encoding ensemble to store learned information. This recruitment is regulated by neuronal selection based on differences in intrinsic excitability (IE) and the expression of the transcription factor CREB. However, whether other forms of plasticity influence memory allocation remains unknown. Here, we found that chromatin compaction and histone acetylation in the mouse lateral amygdala display a high degree of heterogeneity, a prerequisite for neuronal selection. Consequently, when we increased histone acetylation by overexpressing histone acetyl transferases (HATs), neurons with elevated histone acetylation were preferentially recruited into the encoding ensemble and memory retention was enhanced, while optogenetic silencing of the epigenetically altered neurons prevented memory expression. Mechanistically, using patch-clamp recordings and single-nucleus multi-ome sequencing, we observed that HAT overexpression increased IE and epitranscriptomic changes favoring synaptic plasticity. Lastly, by merging FRET-based epigenetic beacons with calcium indicators to simultaneously record histone acetylation and neuronal dynamics in real time, we found that epigenetic heterogeneity underlies IE in cell-autonomous manner. These results identify chromatin plasticity as a key factor catalyzing memory allocation.

Astrocyte allocation during brain development is controlled by Tcf4-mediated fate restriction

Project description:Astrocytes within specific brain regions contribute uniquely to regional circuits for higher-order brain function through interactions with local neurons. The regional diversification of astrocytes is dictated by their embryonic origin, yet the mechanisms governing their regional allocation remain unknown. Here we show that allocation of astrocytes to specific brain regions requires the transcription factor 4 (Tcf4) mediated fate restriction during brain development. Loss of Tcf4 in ventral telencephalic neural progenitors alters the fate of oligodendrocyte precursors to transient intermediate astrocyte precursor cells, resulting in mislocated astrocytes in the dorsal neocortex. These ectopic astrocytes originated from the ventral telencephalon engage with neurons and acquire features reminiscent of local neocortical astrocytes. Furthermore, Tcf4 functions as a suppressor of astrocyte fate during differentiation of oligodendrocyte precursors, thereby restricting the fate to oligodendrocyte lineage. Our study reveals that fate restriction governs regional astrocyte allocation, contributing to astrocyte diversification across brain regions.

Pancreas lineage allocation and specifciation are regulated by sphingosine-1-phosphate signalling

Project description:Identifying the signals that regulate the survival, lineage allocation and specification of pancreas progenitors will help elucidate the embryonic origins of pancreas dysfunction and provide important cues for the efficient conversion of pluripotent stem cells into fully functional β cells. Several transcription factors regulating the conversion of the early pancreatic progenitors into terminally differentiated cells have been identified but extracellular signals regulating pancreas development are less well understood. Using a combination of genetic approaches, organotypic cultures of embryonic pancreata and genomics we have found that sphingosine-1-phosphate signalling through plays a key role in this process. S1p signalling stabilizes the Hippo pathway effector YAP to promote progenitor survival, acinar and endocrine specification. Endocrine cell specification relies on Gai subunits revealing an unexpected dependence of lineage specification on selected intracellular signalling components. Independently of YAP stabilization, S1p signalling attenuates Notch levels, thus regulating lineage allocation. These findings identify S1p signalling as a key pathway coordinating cell survival, lineage allocation and specification during pancreas development.
