Project description:Peripheral neuropathies are the most common neurodegenerative diseases affecting more than 20 million people in the USA alone. Chemotherapy, including treatment with the highly effective drug bortezomib (BTZ), is a frequent cause of peripheral neuropathy. We demonstrated recently that while some chemotherapeutics exert direct toxic effects on axons, BTZ leads to axon degeneration via signals from neuronal somata, which reside in the dorsal root ganglia (DRG). However, what these signals are and how they arise is unknown. To address this question, we treated mice intravenously with BTZ twice weekly for 8 weeks and determined the impact on DRG gene expression via single cell RNA-sequencing.

Desciption of the transcriptome profiles of mouse heart innervating DRG and Nodose Ganglia-derived sensory neurons

Project description:To obtain the mRNA expression profiles of vagal and spinal cardiac sensory neurons, mouse Dorsal Root Ganglia (DRG) and Nodose Ganglia (NG) neurons innervating the heart were labelled by retrograde Di-8-ANEPPQ dye which was injected to the heart. Then bilateral NGs and DRGs between C-T segments were dissected and Di-8-ANEPPQ-labelled DRG and NG neurons were FACS sorted. Finally, Di-8-ANEPPQ+ DRG and NG neurons were subjected to total RNA extraction and library preparation protocols for RNA-seq experiment. For control samples, total NG and DRG (C-T) neurons were dissected from the wt mice and same RNA extraction and library preparation protocols were applied. Finally, mRNA profiles of heart-spesific DRG and NG neurons were compared to those of total DRG and NG populations, respectively.

Bortezomib induces anti-multiple myeloma immune response mediated by cGAS/STING pathway activation

Project description:Proteasome inhibitor bortezomib (BTZ) induces apoptosis in myeloma (MM) cells, and has transformed patient outcome. Using in vitro as well as in vivo immunodeficient and immunocompetent murine MM models, we here show that BTZ also triggers immunogenic cell death (ICD) characterized by exposure of calreticulin (CALR) on dying MM cells, phagocytosis of tumor cells by dendritic cells, and induction of MM specific immunity. We identify a BTZ-triggered specific ICD-gene signature which confers improved outcome in two independent MM patient cohorts. Importantly, BTZ stimulates MM cells immunogenicity via activation of cGAS/STING pathway and production of type-I interferons; and STING agonists significantly potentiate BTZ-induced ICD. Our studies therefore delineate mechanisms whereby BTZ exerts immunotherapeutic activity, and provide the framework for clinical trials of STING agonists with BTZ to induce potent tumor-specific immunity and improve patient outcome in MM.

Group 1 was mRNA extracted from wild type dorsal root ganglia (DRG) taken from mice 10-12 weeks of age, Group 2 was mRNA extracted from DRG taken from NT-4 -/- mice of 4-5 weeks of age. Group 3 was mRNA extracted from DRGs taken from NT-4 -/- mice of 12 weeks of age. In all cases mRNA was extracted from 6-10 mice per group. The experiment was carried out a total of three times.
