Project description:Substantia nigra pars compacta (SNpc) is highly sensitive to normal aging and selectively degenerates in Parkinson's disease. Until now, molecular mechanisms behind SNpc aging have not been fully investigated using high throughput techniques. Here, aging-associated early changes in transcriptome of SNpc were investigated comparing late middle-aged (18 months old) to young (2 months old) mice. Three age groups of C57 wild type mice were used in microarray analysis: young (2 months old), middle aged (10 months old), and late-middle aged (18 months old) mice. Four replicates were included in each age group and each replicate was pooled from 4 mice (4 mice/replicate x 4 replicates x 3 age groups). Total RNA was isolated from SNpc for hybridization on Affymetrix microarrays.

To better understand the underlying mechanisms of this suppression, the goals of this experiment were to compare the transcriptional profiles of the SVZ neural stem cell niche in 3xTg-AD mice versus strain controls. We used early middle-aged mice (7-months-old) rather than old mice, in order to identify genetic changes that are not caused secondarily to other degenerative changes occurring in these mice.

We found several differentially expressed genes, many suggesting an altered immune response to injury with advancing age.

Project description:Purpose: To assess changes in gene expression profiles of mouse ovarian gruanlosa cells from normal control mice, 11-month mice and 11-month mice treated with CEFFE. Methods:The control group comprised 8-week-old female mice, and the 10-month-old female mice were equally divided into two groups, Aged and Aged +CF groups. The mice from Aged +CF group were treated with 200 μL CEFFE (protein concentration: 3 μg/μL) via tail vein every two days for 2 weeks. At the same time, mice from Control and Aged group were injected with the same volume of saline. Then at the forth week ,ovaries were collected. Furthermore, the mRNA-Seq of mouse ovaries were performed to explore underlying mechanism of CEFFE. Results: 1302 genes were significantly increased and 1425 genes were significantly decreased in expression level between Aged vs Ctrl gorup.154 genes were significantly increased and 167 genes were significantly decreased in expression level between Aged+CF vs Aged gorup.
