Project description:Background: Immune mechanisms underlying elicitation in allergic contact dermatitis (ACD) are not disclosed. Previous studies have shown a double-faceted nature of ACD with both common biomarkers between different allergens, as well as allergen-specific imprinting, with discordance in terms of relevant involved immune pathways. Objectives: To characterize molecular signatures and their immune mechanisms between different relevant allergens (nickel, 2-hydroxyethylmethacrylate (2-HEMA), methylisothiazolinone (MI), formaldehyde) in strong and extreme patch test reactions of patients without atopic dermatitis using RNA sequencing.

Integrative analysis of miRNA/mRNA expression patterns in allergic contact dermatitis

Project description:Allergic and irritant contact dermatitis induces different immunological cascades, involving a plethora of immune cells as well as keratinocytes. This highly interwoven network is regulated by miRNAs. However, which miRNAs are involved during allergic- and irritant-induced contact dermatitis is not well investigated. Therefore, we analyzed miRNA and mRNA expression data of the same sample from positive patch test reactions from 27 patients topically exposed to allergens (nickel (Ni), epoxy resin (EP) and methylchloroisothiazolinone (CM); each n=5), irritants (sodium lauryl sulfate (SL, n=9) and nonanoic acid (NO, n=5)), as well as healthy skin (n=5).

Nickel-induced contact dermatitis induce changes in human skin transcriptome over 96 hours

Project description:We aimed to characterize transcriptomic changes over time (baseline, 2 hours, 48 hours and 96 hours) after nickel-induced contact dermatitis in 6 human probands. As a contrast, 7 patients with SLS-induced irritant contact dermatitis were included. Therefore, the patch tests were applied onto the back of the individuals and punch biopsies were taken at the respective time points. Only skin with clear reactions was further used for transcriptomic analysis. Using gene set enrichment analysis and leukocyte deconvolution algorithm, we were able to identify important immune cell subsets and related functions within the human skin.

Quantitative multiplexed ChIP-Seq (MINUTE-ChIP) from formaldehyde-fixed material

Project description:In crosslinked MINUTE-ChIP, formaldehyde-fixed chromatin is fragmented using sonication, blunted and ligated to double-stranded DNA adaptors that include a T7 RNA polymerase promoter and a sample barcode sequence. Finally, samples are combined and subsequent ChIP reactions are performed with the pooled samples. ChIP material is prepared into an Illumina-compatible library using linear amplification by virtue of T7 RNA polymerase, reverse transcription and a low-cycle library PCR amplification. Here, we demonstrate a MINUTE-ChIP for Nanog from formaldehyde-fixed cells using fragmentation by sonication.
