Project description:We employed the Affymetrix GeneChip technology to evaluate the patterns of expression in two different in vivo models of cardiac remodeling and in two different regions (left ventricle free wall and septum) of the heart. Mice underwent transverse aortic constriction (TAC); myocardial infarction (MI) or Sham operation and RNA from the left ventricle free wall and the septum was isolated 1 week later.

Gene expression profiling in Wistar male rat left ventricle with chronic and severe aortic valve regurgitation

Project description:Aortic valve regurgitation (AR) imposes a severe volume overload to the left ventricle (LV) which results in dilation, eccentric hypertrophy and eventually loss of function. Little is known about the impact of AR on LV gene expression. We therefore conducted a gene expression profiling study in the LV of male Wistar rats with chronic (9 months) and severe AR. Five male Wistar rats had one or two aortic valve leaflets punctured with a catheter under echocardiographic guidance in order to induce severe regurgitation (>65% of blood regurgitating during diastole from the aorta to the left ventricle). Five additional rats were sham-operated. The animals were sacrificed 9 months after the procedure and their left ventricle collected for RNA extraction and microarray analysis.

Expression data from uninephrectomized dogs compared to sham operated dogs

Project description:Renal insufficiency is a risk factor for development of cardiovascular disease. In this study our aim was to investigate functional and structural changes in the heart of dogs with mild renal insufficiency induced by uninephrectomy (UNX). In addition to comprehensively assessing functional parameters 12 week after UNX or sham surgery, we harvested tissue for assessment of structural changes. Microarray analyses were performed on snap frosen tissue from the left atrium (LA) and left ventricle (LV) of UNX and sham operated animals. Gene expression in LA was compared between the groups, and gene expression in the LV was compared between the groups. We aimed to assess whether a potentially altered gene expression in the heart of the UNX group also included genes associated with cardiac remodeling. Six dogs were assigned to unineprectomy and 6 dogs were assigned to sham operation. Microarray analyses were performed on 5 samples from the left atrium of uninephrectomized animals and on 5 samples from the left atrium of sham operated animals. Further, microarray analyses were performed on 5 samples from the left ventricle of uninephcretomized animals and on 5 samples from the left ventricle of sham operated animals. No replicates were perfomed.
