Project description:Micro and nanoplastics (MNPLs) are contaminants originated mainly from plastic waste degradation that pose potential health risks. Inhalation is a major exposure route, as evidenced by their detection in human lungs, with polyethylene terephthalate (PET) among the most abundant particles in respiratory airways. However, the harmful effects of particle bioaccumulation remain unclear, as chronic effects are understudied. To assess the long-term effects, specifically the carcinogenic effects, BEAS-2B cell line was exposed to PET-NPLs for 30 weeks. Genotoxicity, carcinogenic phenotypic hallmarks, and a panel of genes and pathways associated with cell transformation and lung cancer were examined and compared across three exposure durations. No significant effects were observed after 24 hours or 15 weeks of exposure. However, 30-week exposure led to increased genotoxic damage, anchorage-independent growth, and invasive potential. Transcriptomic analysis showed upregulation of several oncogenes and lung cancer-associated genes at the end of the exposure. Further analysis revealed an increase in differentially expressed genes over time and a temporal gradient of lung cancer-related genes. Altogether, the data suggest PET-NPLs' potential carcinogenicity after extended exposure, highlighting serious long-term health risks of MNPLs. Assessing their carcinogenic risk under chronic, real-life conditions is crucial to address knowledge gaps and eventually develop preventive policies.

Project description:Plastics are emerging pollutants of great concern. Macroplastics degrade into microplastics and nanoplastics, which can accumulate in living organisms with still poorly known consequences. Nanoplastics being particulate pollutants, they are handled in animal organisms by scavenger cells such as macrophages, which are important players in the immune system. Polyethylene terephthalate is one of these plastics of concern, as it is widely used in food packaging where it releases nanoparticles. We have thus undertaken a study on the effects of true-to-life polyethylene terephthalate nanoparticles prepared from water bottles on macrophages. To this purpose, we used a combination of proteomics and targeted validation experiments. Proteomics showed important adaptive changes in the proteome in response to exposure to polyethylene terephthalate nanoparticles. These changes affected for example mitochondrial, cytoskeletal and lysosomal proteins, but also proteins implicated in immune functions. Validation experiments showed that many of these changes were homeostatic, with no induced oxidative stress and no gross perturbation of the mitochondrial function. However, polyethylene terephthalate nanoparticles induced endoplasmic reticulum stress and disturbed the immune functions of macrophages. We indeed observed a slight pro-inflammatory response (1.5-fold increase in TNF secretion). We also observed a decrease in the response to bacterial stimulation (1.6 decrease in IL-6 secretion). We also observed a 20 percent decrease in the expression of important proteins involved in immune responses such as TLR2, TLR7 or collectin 12, and a two-fold decrease in the production of lysozyme. This suggests that macrophages having ingested polyethylene terephthalate nanoparticles are less efficient in their immune functions.

Many of these products are remarkably persistent in the environment, and the accumulation in the environment is recognised as a major threat. Therefore, an increasing interest has been paid to screen for organisms able to degrade and assimilate the plastic. Ideonella sakaiensis was isolated from a plastisphere, a bacterium that solely was thriving on the degradation on PET films. The processes affected by the presence of PET, terephthalic acid, ethylene glycol, ethyl glycolate, and sodium glyoxylate monohydrate was elucidated by differential proteomes. The exposure of PET and its monomers seem to affect two major pathways, the TCA cycle and the β-oxidation pathway, since multiple of the conditions resulted in an increased expression of proteins directly or indirectly involved in these pathways, underlying the importance in the degradation of PET by I. sakaiensis.

Project description:BackgroundAs modern industrial activities have advanced, the prevalence of microplastics and nanoplastics in the environment has increased, thereby impacting plant growth. Potassium is one of the most crucial nutrient cations for plant biology. Understanding how polyethylene terephthalate (PET) treatment affects potassium uptake will deepen our understanding of plant response mechanisms to plastic pollution.ResultsIn this study, we examined the impact of PET micro- and nanoplastics on foxtail millet seedling growth and potassium accumulation. Additionally, we measured reactive oxygen species (ROS) production, antioxidant enzyme activities, and the expression levels of the corresponding enzyme-encoding genes. Our findings indicated that the germination and seedling growth of foxtail millet were not significantly affected by exposure to PET plastics. However, the ROS levels in foxtail millet increased under these conditions. This increase in ROS led to the upregulation of several genes involved in K+ uptake and transport (SiHAK1, SiHAK2, SiAKT2/3, SiHKT2;2, SiHKT1;1, SiGORK, and SiSKOR), thereby increasing K+ accumulation in foxtail millet leaves. Further research revealed that higher K+ concentrations in plant leaves were correlated with increased expression of the antioxidant-related genes SiCAT1, SiPOD1, and SiSOD3, as well as increased activities of the corresponding antioxidant enzymes. This response helps mitigate the excessive accumulation and damage caused by ROS in plant cells after PET nanoplastic treatment, suggesting a potential stress response mechanism in foxtail millet against nanoplastic pollution.ConclusionsOur research indicates that PET nanoplastic treatment induces the expression of genes related to K+ uptake in foxtail millet through ROS signaling, leading to increased K+ accumulation in the leaves. This process mitigates the ROS damage caused by PET nanoplastic treatment by increasing the expression and activity of genes encoding antioxidant enzymes. The present research has unveiled the K+ accumulation-related response mechanism of foxtail millet to PET nanoplastic treatment, contributing significantly to our understanding of both the potassium absorption regulation mechanism in plants and the broader impact of plastic pollution on agricultural crops. This discovery not only highlights the complexity of plant responses to environmental stressors but also underscores the importance of considering such responses when evaluating the ecological and agricultural implications of plastic pollution.

Project description:Polyethylene terephthalate (PET) is one of the most produced plastic materials in the world. The emergence of microplastics and nanoplastics (MPs/NPs) as a significant environmental contaminant has become a matter of increasing concern. While the toxicological effects of PET NPs have been widely researched, there is a lack of methodologies for studying their accumulation. The present study introduces a novel method to monitor the distribution of PET NPs in germinating wheat (Triticum aestivum L.) seeds. This involves the functionalization of superparamagnetic iron oxide nanoparticles (SPIONs) with PET NPs (PET-fSPIONs) coupled with magnetic resonance microimaging (µMRI) to provide insight into their distribution within the seed. The present study has demonstrated that PET-fSPIONs accumulate in specific regions of germinating wheat seeds, including the shoot apical meristem, the radicle, the coleoptile, the plumule, and the scutellum. Furthermore, the accumulation of PET-fSPIONs has been shown to exert a discernible effect on spin-spin relaxation (T2), as observed via MRI and quantitative T2 relaxation time analysis. The accumulation of PET NPs in embryo regions was also confirmed by SEM. Diffusion-weighted magnetic resonance imaging (DW-MRI) and non-invasive chemical shift imaging analyses demonstrated that PET NPs resulted in restricted diffusion within the highlighted areas, as well as an impact on lipid content. Our study reveals that using µMRI with fSPIONs provides a non-invasive method to monitor the biodistribution of PET nanoparticles in wheat seeds. Additionally, it offers valuable insights into the microstructural interactions of PET.
