Project description:Cyanidin-3-glucoside (C3G) is one of the most prevalent anthocyanins among dietary polyphenols. Experimental studies suggest that C3G exhibits antioxidant, anti-inflammatory, and cytoprotective effects against various disorders induced by oxidative stress. We examined its in vivo anti-neoplastic properties using MMTV-PyVT transgenic mice, a well-validated model for the development and progression of multistage mammary carcinoma.

Characterization of Mammary Tumors Arising from MMTV-PyVT Transgenic Mice.

Project description:Among genetically engineered mouse models of breast cancer, MMTV-PyVT is a mouse strain in which the oncogenic polyoma virus middle T antigen is driven by the mouse mammary tumor virus promoter. The aim of the present study was to perform morphologic and genetic analyses of mammary tumors arising from MMTV-PyVT mice. To this end, mammary tumors were obtained at 6, 9, 12, and 16 weeks of age for histology and whole-mount analyses. We conducted whole-exome sequencing to identify constitutional and tumor-specific mutations, and genetic variants were identified using the GRCm38/mm10 mouse reference genome. Using hematoxylin and eosin analysis and whole-mount carmine alum staining, we demonstrated the progressive proliferation and invasion of mammary tumors. Frameshift insertions/deletions (indels) were noted in the Muc4. Mammary tumors showed small indels and nonsynonymous single-nucleotide variants but no somatic structural alterations or copy number variations. In summary, we validated MMTV-PyVT transgenic mice as a multistage model for mammary carcinoma development and progression. Our characterization may be used as a reference for guidance in future research.

Comparative analysis of the effect of different diets on histone H3K4me3 distribution in mouse liver chromatin reveals the epigenetic efficacy of cyanidin-3-O-glucoside dietary intake

Project description:Background: Different diets result in significantly different phenotypes through metabolic and genomic reprogramming. Epigenetic marks, identified in humans and mouse models upon caloric restriction, high fat diet or the intake of specific bioactives, suggest that genomic reprogramming drives this reprogramming and mediates the effect of nutrition on health. Histone modifications encode the epigenetic signal that adapts genome functions to environmental conditions, including diets, by tuning the structure and properties of chromatin. So far, the effect of different diets on the genome-wide distribution of critical histone marks has not been determined Methods: We investigated by chromatin immunoprecipitation sequencing the distribution of the trimethylation of lysine 4 of histone H3 in the liver of mice fed for one year with five different diets including: chow containing corn powder as extra source of plant bioactives or specifically enriched with the cyanidin-3-O-glucoside, high fat enriched obesogenic and caloric restricted pro-longevity diets Conclusions: The comparison of the resulting histone mark profiles revealed functional food containing cyanidin determines broad effect.

miRNA expression during early, mid and late breast cancer disease progression in mammary glands from MMTV-PyMT transgenic mice

Project description:MicroRNAs (miRNAs) are small noncoding RNAs that typically inhibit the translation and stability of messenger RNAs (mRNAs), controlling genes involved in a variety of cellular processes. miRNA dysregulation is recognized to play an essential role in the development and progression of cancer. MMTV-PyMT mice (Jax Strain: FVB/N-Tg(MMTV-PyVT)634Mul/J) are a well-characterized transgenic mouse model of breast cancer. Upon activation of the MMTV-PyVT transgene (mouse mammary tumor virus (MMTV) long terminal repeat upstream of a cDNA sequence encoding the Polyoma Virus middle T antigen (PyVT)) female carriers develop palpable mammary tumors as early as 5 weeks of age. We performed miRNA microarrays on samples from the MMTV-PyMT transgenic mouse model to investigate the differential expression of miRNAs during development of malignant disease in this model.

Understanding the functional role of cyanidin-3-O-glucoside in mouse osteoblastic cell line MC3T3-E1 by RNA-seq analysis

Project description:Cyanidin-3-O-glucoside (C3G) is one of the most widely distributed anthocyanins and has the effect of reducing the risks of osteoporosis. However, the molecular mechanism by which C3G promotes bone formation remains incompletely understood. Our aim was to investigate the effect of C3G on gene expression profiles, during the proliferation and differentiation in MC3T3-E1, a kind of osteoblast cell line from mice, by RNA sequencing. The results identified 34 up-regulated and 17 down-regulated genes. We further performed Gene Ontology (GO), Kyoto Encyclopedia of Genes and Genomes analyses (KEGG), and other bioinformatics methods to do further analysis of differentially expressed genes (DEGs). Analysis of sequencing data showed that these genes were highly enriched in functions related to Lysosomal and Glycolipid biosynthetic. Lastly, we used RT-qPCR to validate the differentially expressed mRNAs. The results showed that C3G increased the expression levels of Ly6a (Lymphocyte antigen 6 family member A), Atp6v0c (ATPase H+ transporting v0 subunit c), and decreased the expression levels of Cx3cl1 (C-X3-C motif chemokine ligand 1). This study suggested that C3G activated osteoblast via Glycosylphosphatidylinositol(GPI)-anchor biosynthesis and the Cx3cl1/Cx3cr1 signaling pathway.

The genome-wide enhancer profiling of breast cancer in the MMTV-PyVT mice [ChIP-seq]

Project description:Activation of transcription enhancers, especially super enhancers, is one of the important epigenetic features of tumorigenesis. However, only very few studies reported how the enhancer landscape evolves during tumorigensis. Here we utilized a proteomics approach and found that H3K27ac and H4K8ac are elevated in the mammary tumor of MMTV-PyVT mouse model, which was then confirmed by IHC results in human BRCA chips. ChIP-seq studies revealed that H4K8ac is highly co-localized with H3K27ac on chromatin, especially on the distal enhancers . We further identified a subgroup of super enhancers which are marked by H3K4me3 peaks in the intergenic regions. These H3K4me3-enriched super enhancers are covered with higher level of H3K27ac and H4K8ac in tumor than normal tissue , and accompanies higher transcription level of the adjacent genes. Combined with global gene expression data, we identified 148 tumor H3K4me3-enriched super enhancer adjacent genes with higher expression in the tumor tissues , which may be critical for breast cancer. We further utilized one inhibitor of p300, C646, to inhibit H3K27ac level, which successfully repressed tumor formation process probably through inhibiting Vegfa and other oncogenes. Taken together, our work identifies novel regulators and provides important resource to the genome-wide enhancer studies in breast cancer, and raises the possibility of cancer treatment through modulating enhancer activity.
