Mapping the cardiac vascular niche in heart failure

Project description:Various transgenic CreER;tdTomato mouse-lines (Pdgfrb-CreER, Col1a1-CreER, Gli1-CreER, Cdh5-CreER, Myh11-CreER, Ng2-CreER) were pulsed with tamoxifen and subjected to either Sham or TAC surgery at 21 days after tamoxifen induction. Two and four weeks later, tdTomato-positive cells were isolated from hearts by FACS sorting and 10x Genomics scRNA 3'RNA sequencing pipeline was applied. We identified fibroblast, endothelial cells, mural cells and Schwann cells as major cardiac cell types in our samples. Gene expression changes between subtypes of the major cell types point towards a general upregulation of extra cellular matrix related genes, with fibroblasts being the highest producer.

Project description:Purpose: Comparision transcriptomes of isolated tdTomato+ and tdTomato- OLs from the brains and spinal cords of infected mice at 30 dpi to those of OLs isolated from mock-infected central nervous system (CNS) samples by nextgen sequencing Methods: Brains and spinal cords were harvested at day 30 p.i. and processed as described above. After Live/Dead staining and treatment with Fc block, OLs were identified using APC-anti-O4 (1:50; Miltenyi). RNA was extracted from sorted OLs using an RNeasy Micro Kit (Qiagen). RNA sequence analysis was performed at the University of Minnesota Genomics Center. Results: We identified 158 and 115 genes, that showed a significant difference (p<0.05) when tdTomato+ and tdTomato- cells were compared to cells from mock-infected mice, respectively, with 61 mRNAs common to both tdTomato- and tdTomato+ cells

High fluid shear stress induces Hippo/YAP pathway in articular cartilage superficial layer cells: a potential mechanistic link to 0steoarthritis

Project description:Abnormal mechanical loading, which can lead to articular cartilage damage, is a significant contributor to the onset of osteoarthritis (OA). Articular cartilage superficial layer cells are among the first cells to respond to changes in the mechanical environment and are highly sensitive to mechanical stimuli. This study aimed to investigate the effects of high fluid shear stress on the articular cartilage superficial layer cells and the underlying mechanisms. We found that high fluid shear stress of 20 dyne/cm² induces inflammation and promotes catabolic processes in these cells. Short-term high fluid shear stress has a protective effect, but its efficacy varies with time. YAP plays a crucial role in mediating the effects of high fluid shear stress and may represent a potential therapeutic target for early-stage osteoarthritis. The study also established osteoarthritis models using anterior cruciate ligament transection (ACLT) or injection of sodium iodoacetate (MIA) to further confirm the findings.
