Project description:The nervous system requires metabolites and oxygen supplied by the neurovascular network, but this necessitates close apposition of neurons and endothelial cells. We find motor neurons attract vessels with long-range VEGF signaling, but endothelial cells in the axonal pathway are an obstacle for establishing connections with muscles. It is unclear how this paradoxical interference from heterotypic neurovascular contacts is averted. Through a mouse mutagenesis screen, we show that Plexin-D1 receptor is required in endothelial cells for development of neuromuscular connectivity. Motor neurons release Sema3C to elicit short-range repulsion via Plexin-D1, thus displacing endothelial cells that obstruct axon growth. When this signaling pathway is disrupted, epaxial motor neurons are blocked from reaching their muscle targets and concomitantly vascular patterning in the spinal cord is altered. Thus, an integrative system of opposing push-pull cues ensures detrimental axon-endothelial encounters are avoided while enabling vascularization within the nervous system and along peripheral nerves.

Project description:A double cropping system has been commercially adopted in subtropical regions in southern China, where there is abundant sunshine and heat resources. In this viticulture system, the first growing season normally starts as a summer cropping cycle; then, the vine is pruned and forced by hydrogen cyanamide, resulting in a second crop in January of the next year. Due to climate differences between the two growing seasons, flavonoid content and composition varies greatly. In this study, changes in the transcriptome of flavonoid-associated pathways were compared in berries grown under the double cropping system; in addition, the accumulation of flavonoid compounds was compared. Specific alterations in MYB transcription factors occurred in winter cropping berries around veraison. Then, the winter cropping cycle distinctly induced the flavonoid metabolic pathways while triggering the ripening-associated pathways. Notably, the climate conditions in winter cropping positively affected flavonoid biosynthesis, while the summer season took a major toll on anthocyanin accumulation. In addition, the three classes of flavonoid compounds responded differently to the changing climate; the anthocyanins and flavonols were promoted several fold, whereas no consistent increase was found for flavan-3-ols. Conclusively, flavonoid biosynthesis in grapes grown under a double cropping system showed seasonal or climatic-specific accumulation patterns.

Abstract:
According to receptor theory, the effect of a ligand depends
on the amount of agonist-receptor complex. Therefore, changes
in receptor abundance should have quantitative effects.
However, the response to pheromone in Saccharomyces cerevisiae
is robust (unaltered) to increases or reductions in the
abundance of the G-protein-coupled receptor (GPCR), Ste2,
responding instead to the fraction of occupied receptor. We
found experimentally that this robustness originates during
G-protein activation. We developed a complete mathematical
model of this step, which suggested the ability to compute
fractional occupancy depends on the physical interaction
between the inhibitory regulator of G-protein signaling (RGS),
Sst2, and the receptor. Accordingly, replacing Sst2 by the
heterologous hsRGS4, incapable of interacting with the
receptor, abolished robustness. Conversely, forcing hsRGS4:Ste2
interaction restored robustness. Taken together with other
results of our work, we conclude that this GPCR pathway
computes fractional occupancy because ligand-bound GPCR-RGS
complexes stimulate signaling while unoccupied complexes
actively inhibit it. In eukaryotes, many RGSs bind to specific
GPCRs, suggesting these complexes with opposing activities also
detect fraction occupancy by a ratiometric measurement. Such
complexes operate as push-pull devices, which we have recently
described.
