Transcriptomic analyses reveal genes with sexual dimorphic expression in the Arapaima gigas gonads and brain

Project description:Pirarucu (Arapaima gigas, or A. gigas) is a native fish species to Amazon basin and, economically important in the Brazilian Amazonian for its great potential to aquaculture. In the natural environment the A. gigas is captured only in the sustainable development reserves of the State of Amazonas by applying a system of rotation in fishing in order to avoid overfishing of this important fishing resource. The reproductive biology of A. gigas has long been intriguing; however, very little is known about the molecular pathways underlying their sexual differentiation and determination. Using the SOLiD sequencing platform, a total of 432,058,560 short sequencing reads were produced. An average of ~30% of sequencing reads could be mapped to Asian arowana reference cDNAs. 305 genes showed higher expression in female brain against 8 gene with higher expressing in male brain. In gonad, there are 120 genes higher expressed in female against 10 gene higher expressed in male.

Whole-transcriptome RNA sequencing of skin reveals innate immunity differences between wild-type and yellow mutant rainbow trout

Project description:Rainbow trout (Oncorhynchus mykiss) is an important aquaculture fish species that is farmed worldwide, and it is also the most widely cultivated cold water fish in China. This species, a member of the salmonidae family, is an ideal model organism for studying the immune system in fish. Two phenotypes of rainbow trout are widely cultured; wild-type rainbow trout with black skin (WR_S) and yellow mutant rainbow trout with yellow skin (YR_S). Fish skin is an important immune organ, however, little is known about the differences in skin immunity between WR_S and YR_S in a natural flowing water pond aquaculture environment, and very few studies were conducted to investigate the ceRNA mechanism for fish skin.

Targeted proteomic validation of dysregulated proteins in liver tissue of Labeo rohita after Aeromonas hydrophila infection

Project description:Aeromonas hydrophila (Ah) is an opportunistic Gram-negative bacterium and a serious global pathogen causing Motile Aeromonas Septicaemia (MAS) in fish and many other vertebrates. This study aimed at identifying the cellular proteins and processes altered by Ah infection in liver tissue of Labeo rohita, an important aquaculture species. Discovery based proteomics data could identify a panel of differentially expressed proteins (DEPs) between Control and AH condition (Ah infected group). Protein abundance changes in a few selected DEPs was then validated using Selected reaction monitoring (SRM) approach.

Project description:Streptococcus agalactiae (Lancefield’s group B Streptococcus, GBS) is a major bacterial species of genus Streptococcus and has medical and veterinary importance by affecting mainly humans (Maione et al., 2005; Johri et al., 2006), cattle (Keefe, 1997) and fish (Mian et al., 2009). The GBS is the most important pathogen for the Nile tilapia, a global commodity of the aquaculture sector, causing outbreaks of septicemia and meningoencephalitis (Hernández et al., 2009; Mian et al., 2009). This study aimed to evaluate the global abundancy of proteins among the main genotypes of GBS isolated from fish identified in Brazil using a label free shotgun liquid chromatography-ultra definition mass spectrometry (LC-UDMSE) approach and to compare the differential expression of proteins identified between isolates from fish and human.

A coordinated attack by a bacterial secretion system and a small molecule drives prey specificity

Project description:Vibrio species are recognized for their role in food- and water-borne diseases in humans, fish, and aquatic invertebrates. We screened bacterial strains isolated from raw food shrimp for those that are bactericidal to Vibrio strains. Here we identify and characterize Aeromonas dhakensis strain A603 which shows robust bactericidal activity specifically towards Vibrio and related taxa but less potency toward other Gram-negative species. Using the A603 genome and genetic analysis, we show that two antibacterial mechanisms account for its vibriocidal activity -- a highly potent Type Six Secretion System (T6SS) and biosynthesis of a vibriocidal phenazine-like small molecule, named here as Ad-Phen. Further analysis indicates coregulation between Ad-Phen and a pore-forming T6SS effector TseC, which potentiates V. cholerae to killing by Ad-Phen.

Shotgun proteomics of Sparus aurata hepatic stress response

Project description:This study characterizes the liver stress proteome of fish submitted to overcrowding (OC), repeated net-handling (NET) and hypoxia (HYP). Fish trials were conducted in triplicate tanks and two fish per tank were randomly sampled for MS analysis. This work aims to disclose most significant changes in signaling and metabolic pathways involved in the stress response. This data-driven knowledge may ultimately contribute for the improvement of species-specific welfare management protocols, towards a sustainable aquaculture.
