Project description:Nano-catalytic therapy mediated by reactive oxygen species (ROS) is a highly promising tumor treatment strategy. However, the unique physiological and biochemical characteristics of tumor microenvironment, such as hypoxia and overexpression of antioxidants and glutathione, limit the production of ROS and their oxidation capacity within the tumor. In this study, an S-scheme heterojunction (Au/MoS2/Bi2S3) with multiple enzyme activities was constructed to achieve significant anti-tumor effects through photothermal/photodynamic combination therapy (PTT/PDT). Under near infra-red irradiation, Au/MoS2/Bi2S3 heterojunction not only provided high fever for PTT but also generated singlet oxygen for PDT. In addition, the Au/MoS2/Bi2S3 heterojunction also could generate H2O2 in-situ (Glucose oxidase-like activity), meanwhile simultaneously complete the decomposition of endogenous H2O2 (Peroxidase-like and Catalase-like activity) and depletion of glutathione (Glutathione-peroxidase-like activity) through the conversion between Mo4+ and Mo6+, further enhancing the generation of ROS. In vitro and in vivo studies demonstrated that the photoactivated Au/MoS2/Bi2S3 heterojunction with multiple enzyme activities could completely eliminate tumors without recurrence, meanwhile this heterojunction also exhibited the high biological safety. This study provided a new perspective for tumor therapy based on nano-catalytic method.

Integrated tissue and serum proteomics on magnet-guided photothermal therapy using Au/Fe/Au trilayer nanodiscs

Project description:Nanodiscs are emerging as promising nanomaterials for localized therapies as they enable magnetic targeting and near-infrared (NIR) laser–driven photothermal stimulation. In this study, we performed comprehensive proteomic analyses of both tissue and serum from mouse models to elucidate the effects of photothermal therapy using Au/Fe/Au trilayer nanodiscs (AuFeAuNDs) on both local and systemic protein regulation. Protein expression in tissue and serum was analyzed under six experimental exposure conditions: control, control plus laser irradiation, Au nanodiscs, AuFeAuNDs without magnetic induction, AuFeAuNDs with magnetic induction, and AuFeAuNDs with magnetic induction plus laser irradiation. Tissue proteomes showed enrichment of proteins involved in histone modification and transcriptional regulation, while serum proteomes revealed alterations in immune responses and coagulation pathways, with the combined AuFeAuNDs, magnet, and laser treatment promoting fibrinolysis. Fifty-two shared proteins across tissues and serum were implicated in ferroptosis, coagulation, and defense responses. Overall, these findings provide mechanistic insights into the biological impact and biosafety of nanodisc-mediated photothermal therapy, as well as highlight potential biomarkers for monitoring therapeutic responses.

Photothermal Modulation of Human Stem Cells Using Light-responsive 2D Nanomaterials

Project description:Two-dimensional (2D) molybdenum disulfide (MoS2) nanomaterials are an emerging class of biomaterials that are photo-responsive at near-infrared wavelengths (NIR). Here, we demonstrate for the first time the ability of 2D MoS2 to modulate cellular functions of human stem cells through photothermal mechanisms. The interaction of MoS2 with human stem cells is investigated using whole transcriptome sequencing (RNA-seq). Global gene expression profile of stem cells reveals significant influence by NIR stimulation of MoS2 on integrins, cellular migration and wound healing. Overall, the combination of MoS2 and NIR light may provide new approach to control and regulate cellular migration and functions.

A study of the mechanism whereby Au-Ag-PDA@MSCM-nanoparticle-mediated photothermal therapy induces sebaceous gland ferroptosis during the treatment of acne

Project description:Branched gold and silver nanoparticles with a polydopamine coating (Au-Ag-PDA) are promising nanomaterials with high photothermal conversion efficiency. Previous studies have shown that a low concentration of Au-Ag-PDA nanoparticles coated with mesenchymal stem cell membranes (Au-Ag-PDA@MSCM), in conjunction with 808 nm LASER irradiation, can be used to treat acne by shrinking the sebaceous gland tissue to inhibit sebum secretion. However, the exact mechanism underlying this treatment remains unclear. We systematically studied the mechanism by which Au-Ag-PDA@MSCM-mediated photothermal therapy (PTT) inhibits sebum secretion in vitro and vivo, using proteomic analyses, biochemical indicators of ferroptosis, bio-transmission electron microscopy, and western blotting assays. To the best of our knowledge, we revealed for the first time that Au-Ag-PDA@MSCM-mediated PTT induced early ferroptosis by upregulation acyl-coenzyme A synthetase long-chain family member 4 (ACSL4) in sebaceous gland cells, including features such as mitochondrial contraction, increased membrane density. sebaceous gland cells iron deposition, lipid peroxidation, and glutathione (GSH) depletion. Using proteomics, we also found that the sebaceous gland cells had mitochondrial respiratory chains and redox imbalances after Au-Ag-PDA@MSCM-mediated PTT. We also found that the Hippo signaling pathway was enriched after this procedure. The results of this study will aid our understanding of how Au-Ag-PDA@MSCM-mediated PTT inhibits sebum secretion and treats acne. Our study also provides an important theoretical basis for studying the mechanisms involved in the use of nanomaterials for photothermal treatment. Turning foes to friends: H2S-Consumed Multifunctional Nanoplatform for Photothermal–Nanocatalytic Synergistic Therapy Inducing Ferroptosis to Enhance Colorectal Cancer Immunotherapy

Project description:Colorectal cancer treatment remains difficult, with traditional approaches restricted by indications, toxicity, and resistance. In this study, we present a tumor microenvironment-responsive nanoplatform, Cu2O@AuMn@HA, that induces ferroptosis through photothermally enhanced dual enzyme-like activities, thereby improving the immunotherapy outcome of CRC. Leveraging hyaluronic acid, the platform selectively targets CD44-overexpressing cells and undergoes conversion to CuxS in response to the elevated H2S levels within tumors, achieving in situ photothermal therapy (PTT) while consuming endogenous hydrogen sulfide (H2S). The nanoplatform exhibits both peroxidase-like (POD-like) and glutathione oxidase-like (GSHOx-like) catalytic activities, promoting reactive oxygen species (ROS) generation while depleting glutathione, thereby disrupting redox homeostasis and driving ferroptosis. Simultaneously, the photothermal effect accelerates these catalytic reactions, further enhancing therapeutic efficacy. Ferroptosis, combined with photothermal stress, provokes immunogenic cell death (ICD), which facilitates immune cells infiltration, and alleviates the immunosuppressive tumor microenvironment. Moreover, the incorporation of Au and Mn endows the platform with triple-modal imaging capabilities -fluorescence imaging, computed tomography (CT), and magnetic resonance imaging (MRI)- enabling precise localization and real-time monitoring at both the cellular and animal levels. In vitro and in vivo experiments demonstrate near-complete tumor eradication in CRC models via a photothermal-nanocatalytic-immune triple-synergistic mechanism, with favorable safety profiles. This study introduces a multifunctional, scalable, precision-based diagnostic and therapeutic strategy for CRC, providing novel insights for future clinical translation.
