Project description:Apis mellifera syriaca is the native honeybee subspecies of Jordan and much of the Middle East. It expresses behavioral adaptations to a regional climate with very high temperatures, nectar dearth in summer, attacks of the Oriental wasp Vespa orientalis and in most cases it is resistant to varroa mites. The Thorax control sample of A. m. syriaca in this experiment was originally collected and stored since 2001 from Wadi Ben Hammad a remote valley in the southern region of Jordan. Using morphometric and Mitochondrial DNA markers it was proved that bees from this area had show higher similarity than other samples collected from the Middle East as represented by reference samples collected in 1952 by Brother Adam. The samples L1-L5 are collected from the National Center for Agricultural Research and Extension breading apiary which was originally established for the conservation of Apis mellifera syriaca. Goal was to use the genetic information in the breeding for varroa resistant bees and to determine the successfulness of this conservation program. Project description:After collection, live honeybees were brought to geneOmbio Technologies Central Processing Laboratory under controlled room temperature (25.9C, 72% RH) until dissection was made. The honeybees selected for analysis were collected from Pune region (18°31′13″N 73°51′24″E) from the state of Maharashtra, India. These were identified as Apis cerena based on mitochondrial COI gene sequencing. Project description:We analyzed the changes in the brain tissue of Apis mellifera ligustica at the molecular level by sequencing after using fluvalinate. We found that the differentially expressed miRNAs (DEM) may be involved in hippocampal cell apoptosis and damage to memory functions. This result may be related to behaviors observed after the administration of this medication, such as a lack of homing at night and behavioral disturbances. Overall, our results provide new information about the molecular mechanisms and pathways of fluvalinate action in the brain tissue of Apis mellifera ligustica.

Project description:Our aims in this study were: 1) to identify the miRNAs of the bumble bees Bombus terrestris and B. impatiens; 2) to compare the total numbers of miRNAs between both bumble bee species and between them and the honey bee, Apis mellifera; and 3) to test whether the sequences and expression patterns of miRNAs were conserved between species. To investigate each of these aims we used miRNA-seq (deep sequencing of miRNA-enriched libraries) in B. terrestris, and bioinformatics prediction programs to identify miRNAs in both Bombus species. We identified 131 miRNAs in B. terrestris, and 114 in B. impatiens; of these, 17 were new miRNAs that had not previously been sequenced in any species. We found a striking level of difference in the miRNAs present between Bombus and A. mellifera, with 103 miRNAs in A. mellifera not being present in the genomes of the two bumble bees.

Project description:We aim to evaluate the effects of four Nosema spores’ isolates, (i) and (ii) N. ceranae isolated from A. mellifera hosts from two different geographical origins, (iii) N. ceranae from A. cerana host and (iv) N. apis from A. mellifera, on the A. mellifera on gut proteomics at the early stage of infection. To dissect the molecular mechanism responsible of the susceptibility of A. mellifera to Nosema, we investigated by high-resolution proteomics (LC-ESI-MS/MS) and differential label-free quantification of proteins (LFQ) the molecular cross-talk existing between different species and isolates of N. apis and N. ceranae, and the targetted gut tissue of A. mellifera. To reach the objectives of this study, we performed a bottom-up proteomic analysis on the different anatomical sections of the gut tissue (esophagus, crop, midgut, ileum and rectum) at an early stage of the exposition to Nosema spores (4 days). Then, we focused on the midgut, the region targeted by Nosema sposres for germination and, as we found out, the second region with the highest load of Nosema proteins, after the rectum, to perform differential quantitative proteomic analyses and acquire series of up- and down-regulated proteins. We discussed the different pathways observed to be impacted by different Nosema species and isolates with a main focus on the deregulated metabolic and response to stimuli processes.

Project description:Here, we demonstrate that Nematostella vectensis, Ciona intestinalis, Apis mellifera, and B. mori, show two distinct populations of genes differentiated by gene-body CpG density. Genome-scale DNA methylation profiles for A. mellifera spermatozoa reveal CpG-poor genes are methylated in the germ line, as predicted by the depletion of CpGs. We find an evolutionarily conserved distinction between CpG-poor and -rich genes: the former are associated with basic biological processes, the latter with more specialized functions. This distinction is strikingly similar to that recently observed between euchromatin-associated genes in Drosophila that contain intragenic histone 3 lysine 36 trimethylation (H3K36me3) and those that do not, even though Drosophila doesnM-CM-"M-BM-^@M-BM-^Yt display CpG density bimodality or methylation. We confirm that a significant number of CpG-poor genes in N. vectensis, C. intestinalis, A. mellifera and B. mori are orthologs of H3K36me3- rich genes in Drosophila. We propose that over evolutionary time, gene-body H3K36me3 has influenced gene-body DNA methylation levels, and consequently the gene-body CpG density bimodality characteristic of invertebrates that harbor CpG methylation.
