RNA from 30 flies from each of 3 genotypes was used to hybridize 4 arrays in a 12-plex array (4 technical replicates, 3 genotypes).
Project description:We report here the transcriptomic analysis of Drosophila melanogaster wing imaginal discs from third instar female larvae expressing Cyclin G deleted of the PEST domain (the 25 COOH-terminal amin-acids) under the control of the daugterless-Gal4 ubiquitous driver. The negative control was transgenic flies wearing only the daugterless-Gal4 driver.

Project description:Targeted DamID (TaDa) measures gene expression using a DNA adenine methyltransferase (Dam) fused to RNA polymerase II (Dam Pol II) to methylate DNA as the genome is transcribed (Southall, Gold et al. 2013, PMID: 23792147). Here we use traditional Illumina RNA-Seq to determine if a cell’s expression profile is affected by Dam Pol II expression. Twenty-four hours prior to collection, we ubiquitously drove UAS Dam or UAS Dam Pol II using a tubulin (tub) GAL4 regulated by a temperature sensitive tub GAL80[ts] (McGuire et al. 2003, PMID: 14657498) in adult Drosophila melanogaster (taxon: 7227) whole males, whole females, testes, and ovaries.
Project description:This project presents a quantitative proteomic analysis of Drosophila melanogaster head tissue expressing human FUS-P525L, using data-independent acquisition (DIA) mass spectrometry. To enable identification of the human transgene, mass spectrometry data were analyzed against a custom database containing the Drosophila melanogaster proteome (UniProt UP000000803) and the human FUS sequence. Head samples were processed through protein extraction, acetone precipitation, and in-solution tryptic digestion. LC-MS/MS data were acquired on a Thermo Scientific instrument and analyzed with the DIA-NN software (v1.8.1), with FDR controlled at 1%. The dataset enables the investigation of protein expression changes, including the transgenic human FUS, under P3 treatment.
