Project description:Transcriptional profiling of immortalized human prostate epithelial cells after CTCF knockdown by short hairpin RNA. Knockdown by shRNA targeting CTCF was conducted for 5 days by addition of doxycycline to induce shRNA expression. RNA was purified from triplicate biological replicates of induced and uninduced shRNA samples. Gene expression was quantified by hybridization to Affymetrix Human Transciptome 2.0 arrays.

Methylated DNA was immunoprecipitated with anti-methylcytosine antibodies from genomic DNA and detected by Affymetrix CytoScanHD chips. Methylated DNA was purified by 5-mC antibodies and Affymetrix CytoScanHD arrays were performed according to the manufacturer's protocols.

Project description:A subpopulation of prostate luminal epithelial cells has been previously reported to be sufficient to regenerate prostatic architecture following consecutive rounds of androgen deprivation/repletion. This functional characteristic suggest prostate luminal epithelial cells as the putative cell-of-origin for castration-resistant prostate cancer - which more notizable fenotype is the lack of response to androgen deprivation thereapy. We used microarrays to profile the androgen-induced transcripts in luminal prostate epithelial cells involved in a cycle of prostate regression/regeneration in Hoxb13-rtTA|TetO-H2BGFP transgenic mice.
