Project description:Prenatal exposure to maternal stress and depression has been identified as a risk factor for adverse behavioral and neurodevelopmental outcomes in early childhood. However, the molecular mechanisms through which maternal psychopathology shapes offspring development remain poorly understood. We analyzed transcriptome-wide gene expression profiles of 149 UCB samples from neonates born to mothers with prenatal PTSD (n=20), depression (n=31) and PTSD with comorbid depression (PTSD/Dep; n=13), compared to neonates born to carefully matched trauma exposed controls without meeting PTSD criteria (TE; n=23) and healthy mothers (n=62). We also evaluated physiological and developmental measures in these infants at birth, six months and twenty-four months. A multistep analytic approach was used that specifically sought to: 1) identify dysregulated genes, molecular pathways and discrete groups of co-regulated gene modules in UCB associated with prenatal maternal psychopathologies; and 2) to determine the impact of perinatal PTSD and depression on early childhood development outcomes.

by the clinical nurse specialist. Stepped care: The steps include: (i) Watchful waiting. (ii) If psychological distress persists, the guided self-help program. (iii) If psychological distress persists, a problem analysis is performed and an agreement is made with the patient on the next step; treatment consists of problem solving therapy (face-to-face). (iv) If psychological distress persists, psychotherapy, medication or a referral for other services (e.g. social work) is offered.
The TES intervention is compared to usual care.

Project description:Background: This multicenter longitudinal study seeks to investigate the dynamic changes of psychological distress (PD) in nasopharyngeal carcinoma (NPC) patients during radiotherapy, and reveal the expression profiles and regulatory networks of circRNAs in these NPC patients. Methods: 282 newly diagnosed NPC patients from three hospitals in China were included. Participants completed questionnaires and provided blood samples. PD trajectories were identified via a latent class growth model (LCGM). Moreover, the factors that influence the PD trajectories were explored. Whole transcriptome sequencing was performed to investigate genetic factors. The real-time quantitative PCR was applied to validate circRNAs. We predicted the target mirnas, target Mrnas and target RNA-binding proteins (RBPs) of the top 10 malregulated circrnas. Subsequently, circRNA-miRNA-mRNA (ceRNA) and circRNA-RBP networks were constructed. In addition, the role of circRNA and target mRNA parent genes was predicted by KEGG and GO analysis. Results: LCGM identified two of the most important PD trajectories during radiotherapy in NPC patients: Class 1 “decline distress group” (11.0%) and Class 4 “rise distress group” (20.3%). Household monthly per capita income, coping strategies, and perceived social support emerged as important predictors of PD trajectories. Regarding genetic factors, 600 circRNAs and 123 miRNAs were identified as being significantly differentially expressed. Notably, hsa_circ_0004277 demonstrated significant differences between patients in the rise and decline distress groups (P < 0.01). ceRNA and RBP networks may influence the pathophysiology of PD in NPC patients undergoing radiotherapy. Conclusion: This study unraveled that PD trajectories in NPC patients during radiotherapy were heterogeneous, indicating the need for screening and timely interventions within this population. Furthermore, the expression patterns of ceRNA and circRNA–RBP networks and pathways related to these networks suggested a potential role of circRNAs in developing PD among NPC patients receiving radiotherapy.

However, the molecular mechanisms through which maternal psychopathology shapes offspring development remain poorly understood. We applied transcriptome-wide screens to 149 umbilical cord blood samples from neonates born to mothers with posttraumatic stress disorder (PTSD; n = 20), depression (n = 31) and PTSD with comorbid depression (n = 13), compared to carefully matched trauma exposed controls (n = 23) and healthy mothers (n = 62). Analyses by maternal diagnoses revealed a clear pattern of gene expression signatures distinguishing neonates born to mothers with a history of psychopathology from those without. Co-expression network analysis identified distinct gene expression perturbations across maternal diagnoses, including two depression-related modules implicated in axon-guidance and mRNA stability, as well as two PTSD-related modules implicated in TNF signaling and cellular response to stress. Notably, these disease-related modules were enriched with brain-expressed genes and genetic risk loci for autism spectrum disorder and schizophrenia, which may imply a causal role for impaired developmental outcomes. These molecular alterations preceded changes in clinical measures at twenty-four months, including reductions in cognitive and socio-emotional outcomes in affected infants. Collectively, these findings indicate that prenatal exposure to maternal psychological distress induces neuronal, immunological and behavioral abnormalities in affected offspring and support the search for early biomarkers of exposures to adverse in utero environments and the classification of children at risk for impaired development.

Project description:The experience of childhood maltreatment (CM) is highly associated with the occurrence of psychiatric disorders. The impact of CM on neurodevelopmental trajectories is not fully elucidated. Here, we aim to investigate, the impact of different types of CM on the transcriptome expressed by peripheral blood mononuclear cells in adolescence. Adolescents who experienced different types of CM or normal environment were exposed to a comprehensive screening aimed to evaluate psychological and behavioural parameters. Among them, we selected four subpopulations, that experienced respectively Abuse (A), Neglect (N), Abuse and neglect (AN), normal childhood (C). To identify biological markers of their respective experience, mRNA-Seq analysis was performed in PBMC extracted from their blood sampling.

Project description:Background: Prenatal depression, affecting up to a quarter of all pregnancies in the United States, contributes to morbidity and mortality and is associated with increased risk of adverse birth and long-term mental health outcomes. Adverse childhood experiences (ACEs, or experiences of abuse, neglect, or family dysfunction experienced prior to age 18) are a strong predictor of adult depression and adverse health outcomes. The present study investigated whether epigenetic modification in the form of DNA methylation (DNAm) of four stress-related, glucocorticoid pathway genes (CRH, CRHR1, FKBP5, NR3C1) me-diates associations between ACEs and depressive symptoms among Black pregnant women. Methods: Using a cross-sectional design, we examined the mediating role of DNAm on the relationship between depressive symptoms (Center for Epidemiologic Studies Depression Scale (CES-D)) and ACEs (Centers for Disease Control and Prevention 10-item questionnaire), in a subsample (n = 61) of Black pregnant women who were participants of the Biosocial Impacts of Black Births (BIBB) study. Results: A significant association was found between ACEs and depressive symptoms scores (TE α_X = 2.29 with p_TE = 6.60 × 105). DNAm on five CpG sites within two genes significantly mediated the relationship between ACEs and depressive symptoms (cg03238273 on CRHR1, and cg08845721, cg16594263, cg19820298, and cg23430507 on NR3C1). Conclusions: This study provides evidence that DNAm partially mediated the association of ACEs and depressive symptoms during pregnancy among Black pregnant women. Understanding the mo-lecular pathways underlying the mediating effect of ACEs on depressive symptoms among Black pregnant women can illuminate biological markers that help identify and treat pregnant women who are at an increased risk for depression following childhood trauma.

Project description:BackgroundChildhood experiences can exert a huge impact on adult psychological conditions. Previous studies have confirmed the effects of adverse childhood experiences (ACEs) and benevolent childhood experiences (BCEs) on psychological distress (e.g., stress, depression, and suicidal ideation) separately, but few studies explored a combined effect of ACEs and BCEs on psychological distress. The aim of this study was to explore a combined effect of ACEs and BCEs on psychological distress among Chinese undergraduates.MethodsParticipants were undergraduates aged 17-24 years (N = 1,816) and completed a self-reported questionnaire. A series of regression analyses were conducted to examine the association between childhood experiences and psychological distress.ResultsA total of 65.7% of undergraduates had BCEs, 27.1% of undergraduates had ACEs, and 12.9% of undergraduates had ACEs and BCEs simultaneously. Logistic regression analysis indicated that undergraduates who experienced high ACEs were more likely to have a high risk of psychological distress [odds ratio (ORs) = 1.46, 1.84, and 3.15 for uncertainty stress, depressive symptoms, and suicidal ideation, respectively], while undergraduates who experienced High BCEs were less likely to have psychological distress (ORs = 0.33, 0.22, and 0.32 for uncertainty stress, depressive symptoms, and suicidal ideation, respectively) compared with Low-Both group. The combined effect of ACEs and BCEs (High-Both group) could also play as a protective factor in uncertainty stress (OR = 0.56) and depressive symptoms (OR = 0.47).ConclusionOur findings suggested that ACEs and BCEs could not only predict the psychological distress independently, but also BCEs could counteract the negative effect of ACEs in psychological problems. There is an even greater need to identify and support the victims of ACEs and to increase BCEs in early childhood.
