Differential roles of Stella in the modulation of DNA methylation during oocyte and zygotic development

Project description:In order to obtain genome-wide profiles, base-resolution methylomes of oocytes and zygotes were generated using the bisulfite sequencing (BS-seq) method for small samples. We find that global loss of DNA methylation during zygotic development in HFD mice. A total of 412 DMRs were identified, of which 294 were hypomethylated (hypo-DMRs; 71.4%) and 118 were hypermethylated (hyper-DMRs; 28.6%) (Fig. 6A and 6B), showing a predominance of hypo-DMRs. Furthermore, we show that hyper-DMRs are significantly depleted from CGI, but enriched in short interspersed elements (SINEs) and non-CGI regions. Strikingly, hypo-DMRs are specifically depleted from SINEs, with a concurrent enrichment in DNA transposons.

STELLA and MuERV-L activation are essential for early mouse development

Project description:The maternal-to-zygotic transition (MZT) marks the period when the embryonic genome is activated and acquires control of development. Maternally inherited factors play a key role in this critical developmental process, which occurs at the 2-cell stage in mice. Here we investigated the function of the maternally inherited factor STELLA (DPPA3) using single-cell/embryo approaches. This submission concerns itself with transcriptional profiling of wild type and Stella knockout (Stella-/-) oocytes, wild type and Stella maternal/zygotic knockout (StellaM/Z-/-) 1-cell and 2-cell embryos. We show that loss of maternal STELLA results in widespread transcriptional mis-regulation and a partial failure of MZT. Strikingly, activation of the LTR class of transposable elements (TE), and particularly 2-cell specific MuERV-L elements, is significantly impaired in StellaM/Z-/- embryos, which leads to a failure to upregulate selected chimeric transcripts. We propose that STELLA is involved in ensuring activation of TEs that themselves play a key role during early development, in part through regulating embryonic gene expression.

Stella prevents excessive de novo DNA methylation during mouse oogenesis

This stage is accompanied with large-scale alteration in transcription, and series of genome-wide epigenetic reprogramming, including de novo establishment of DNA methylation. However, our understanding of mechanisms regulating this process is limited. To investigate the role of Stella (Dppa3) in de novo methylation during mouse oogenesis, here we measured DNA methylation by RRBS and expression profiles by RNA-seq in PGCs and oocytes at serveral development stages, including genotypes of both Stella (Dppa3) +/- and Stella -/-.

Zygotic Arrest 1 Pathogenic Variants Disrupt Maternal mRNA Stability and Cause Oocyte Maturation Defects in Humans

Project description:Zygotic arrest 1 (ZAR1) is one of the earliest identified maternal-effect genes that function during the maternal-to-zygotic transition (MZT) in mice. However, the role of human ZAR1 was unclear. This study investigated the association of ZAR1 gene variants and infertility in women, focusing on their impact on oocyte and early embryonic development. In five individuals with unexplained female infertility, defects in oocyte maturation and early embryonic arrest were observed during in vitro fertilization treatment. Whole-exon sequencing identified either homozygous or compound heterozygous variants of ZAR1 in the patients. Functional analysis revealed that the V118A mutation disrupted the localization of ZAR1 to the mitochondria-associated ribonucleoprotein domain (MARDO) structure, a key mRNA storage site in oocytes. In contrast, R397Q and S121* mutations impaired ZAR1’s RNA-binding capability. These variants disrupt MARDO formation and function, thereby negatively affecting oocyte maturation and early embryonic development. Additionally, transcriptome analysis of oocytes and embryos from ZAR1 variant carriers showed downregulation of maternal mRNAs and altered translation profiles, indicating a disrupted MZT. This study highlights the critical roles of ZAR1 and MARDO in human reproduction and provides insights into the molecular mechanisms underlying some forms of female infertility.

A low-input proteomic analysis of mouse maternal-to-zygotic transition

Project description:Maternal-to-zygotic transition (MZT) is a conserved and fundamental process during which the maternal environment of oocyte transits to the zygotic genome driven expression program, and terminally differentiated oocyte and sperm are reprogrammed to totipotency. It is initiated by maternal mRNAs and proteins during the period of zygotic genome quiescence after fertilization, followed by a gradual switch to zygotic genome activation and accompanied by clearance of maternal RNAs and proteins. A key question for embryonic development is how MZT process is regulated. Here we used a low-input proteomic analysis to measure the proteomic dynamics during early development of mouse maternal-to-zygotic transition.

Metaphase II (MII) oocytes and zygotes (one-cell embryo) serve as the mature oocyte and the initiation of pre-implantation embryo development respectively, and characterizing their molecular landscapes at protein levels plays an important role in uncovering MZT and zygotic genome activation (ZGA )in mammals. Here we used an ultrasensitive proteomic approach to depict an in-depth landscape for the very early stage of mouse MZT.

Expression data from cell lines forced expressed PGC7/Stella

Project description:Global DNA hypomethylation and DNA hypermethylation of promoter regionsâincluding tumor suppressor genesâare frequently detected in human cancers. Although many studies have suggested a contribution to carcinogenesis, it is still unclear whether the aberrant DNA hypomethylation observed in tumors is a consequence or a cause of cancer. We found that overexpression of Stella (also known as PGC7, Dppa3), a maternal factor required for the maintenance of DNA methylation in early embryos, induced global DNA hypomethylation and transformation in NIH3T3 cells. This hypomethylation was due to the binding of Stella to Np95 (also known as Uhrf1, ICBP90) and the subsequent impairment of Dnmt1 localization. In addition, enforced expression of Stella enhanced the metastatic ability of B16 melanoma cells through the induction of metastasis-related genes by inducing DNA hypomethylation of their promoter regions. Such DNA hypomethylation itself causes cellular transformation and metastatic ability. These data provide new insight into the function of global DNA hypomethylation in carcinogenesis. We used microarrays to detail the global programme of gene expression by PGC7/Stella overexpression. RNA was extracted from NIH3T3 or B16F10 murine cell lines overexpressed PGC7/Stella and was hybridized on Affymetrix microarrays. We compared gene expression levels between control and PGC7/Stella-overexpressed cells. Microarray analysis was performed in NIH3T3 cells including two independent Stella-expressing NIH3T3 clones and a mixture of Stella-expressing NIH3T3 clones and in B16-F10 cells including three independent Stella-expressing B16-F10 clones.
