Project description:We set out to determine if distinct prostate luminal epithelial cell-types exhibited age-related differences in their gene expression profiles. We used fluorescence activated cell sorting to isolate Trop2+ and Trop2- luminal cells and EpCAM- CD49f- stromal cells from dissociated preparations of prostate tissue taken from young adult (3 months) and old (24 months of age) mice. RNA was extracted from sorted cells to evaluate differences in gene expression.

Gene expression profiling of basal and luminal epithelial cells from young adult and old mouse prostates

Project description:We set out to determine if prostate epithelial cell-types exhibited age-related differences in their gene expression profiles. We used fluorescence activated cell sorting to isolate basal and luminal cells from dissociated preparations of prostate tissue taken from young adult (3 months) and old (24 months of age) mice. RNA was extracted from sorted cells to evaluate differences in gene expression. We found significant enrichment for progenitor genes in old luminal cells including an immune/inflammatory profile that overlapped with a previously identified CD38-lo human prostate luminal progenitor signature. This study demonstrates a luminal progenitor signature in old mouse prostate.

Gene expression profiling of TROP2+ cells from human CRC (colorectal carcinoma) specimens

Project description:The Trophoblast cell surface antigen 2 (TROP2) is a transmembrane glycoprotein encoded by the Tumor associated calcium signal transducer 2 (TACSTD2) gene. TROP2 is upregulated in many human malignancies including colorectal cancer, and its overexpression correlates with tumor progression, invasiveness, and poor survival. Functional studies revealed that TROP2 may have a role as both an oncogene and a tumor suppressor. To identify TROP2 function in human cancer cells, we performed gene expression profiling of TROP2+ and TROP2- cells isolated from human colon tumors. Due to the sensitive nature of the data, raw data files have not been uploaded.

Gene expression profiling of Trop2+ cells in mouse Apc-deficient small intestinal early adenomas

Project description:Tumor Associated Calcium Signal Transducer 2 (TACSTD2) is one of the cancer-related genes whose overexpression correlates with tumor progression and invasiveness in human colorectal cancer. TACSTD2 gene encodes for a transmembrane glycoprotein TROP2, which is implicated in altered expression of epithelial-mesenchymal transition (EMT) markers and may play a role in metastasis formation. To determine how TROP2 affects aberrant tumor cell signaling, we isolated early adenoma cells from the mouse small intestine 6 weeks after disruption of the Adenomatous polyposis coli (Apc) gene, one of the first steps in the development of colorectal cancer in human. We performed expression profiling of Trop2+ and Trop2- tumor cells and, in addition, non-tumor cells of the intestinal epithelium, including predominantly differentiated cells.

To monitor the onset of TROP2 in hyperplastic cells and its effect on aberrant signaling potentially leading to tumor growth, we isolated epithelial cells from the mouse small intestine 7 days after disruption of the Adenomatous polyposis coli (Apc) gene. Loss of APC tumor suppressor function is the most common initial step in the development of colon cancer in humans. In the mice, the epithelial hyperproliferation occurs within a few days after Apc disruption, accompanied by the formation of ectopic crypts, followed by the formation of microadenomas over time. We performed expression profiling of Trop2+ and Trop2- hyperproliferating cells and, in addition, non-proliferating cells of the intestinal epithelium, including predominantly differentiated cells.
