Transcription profiling of chicken liver after alpha- and gamma-tocopherol supplementation in case of increased oxidative stress susceptibility

Project description:A 30-day nutritional trial in broiler chickens (Ross 308) was conducted to investigate how specific forms of vitamin E (Î±- and Î³-tocopherol) and their combination impact liver gene expression when oxidative susceptibility of the organism is induced by high n-3 polyunsaturated fatty acids (PUFA) intake. Thirty-six one-day-old male broilers were fed a diet enriched with 5 % linseed oil to induce oxidative susceptibility. Beside negative (N) and positive (P) control group, experimental groups were supplemented with either: 67 mg/kg RRR-Î±-tocopherol (A), 67 mg/kg RRR-Î³-tocopherol (G) or with combination of 33.5 mg/kg of each tocopherol (S). Whole chicken genome microarray analysis was performed on liver RNA and selected differentially expressed genes were confirmed by qRT-PCR.

Transcriptomic analysis of liver in sex-linked Dwarf and wild type chickens

Project description:The sex-linked Dwarf (SLD) recessive mutation is characterized by a significant reduction of body size in hemizygous females (30%) and homozygous males (40%). The SLD chickens of the Leghorn line are affected by a Growth Hormone (GH) resistance condition due to a missense mutation at the Growth Hormone Receptor (GHR) gene causing a loss of function of the GHR. The sex-linked recessive dwarf mutation has large commercial use in maternal lines of the broiler poultry industry since these hens require less food and less housing space. The SLD chickens are also an interesting model for investigating complex traits such as energy expenditure and heat resistance for which the SLD chickens differ compared to normal size ones. In this study we performed a whole genome microarray expression profiling of liver using a 8x60K custom Agilent microarray. In total we analyzed 24 SLD and 24 wild type birds of 12 weeks of age obtained by crossing a heterozygous “dw” Leghorn cock with 12 females of an inbred Leghorn line to obtain wild type and dwarf sib and half-sib hens. The aim of the study was the fine identification of GH-inducible genes, and the characterization of the GH dependent signalling pathways.

Transcriptomic profiling of chicken adipose tissue in response to insulin neutralization and fasting

Project description:Domestic broiler chickens rapidly accumulate adipose tissue due to intensive genetic selection for rapid growth and are naturally hyperglycemic and insulin resistant, making them an attractive addition to the suite of rodent models used for studies of obesity and type 2 diabetes in humans. Furthermore, chicken adipose tissue is considered as poorly sensitive to insulin and lipolysis is under glucagon control. Excessive fat accumulation is also an economic and environmental concern for the broiler industry due to the loss of feed efficiency and excessive nitrogen wasting, as well as a negative trait for consumers who are increasingly conscious of dietary fat intake. Understanding the control of avian adipose tissue metabolism would both enhance the utility of chicken as a model organism for human obesity and insulin resistance and highlight new approaches to reduce fat deposition in commercial chickens. In the present study we simultaneously characterized the effects of a short term (5 hours) fast or neutralization of insulin action (5 hours) on adipose tissue of young (16-17 day-old), fed commercial broiler chickens.

An initial map of chromosomal segmental copy number variations in chickens

Project description:Chromosomal segmental copy number variation (CNV) has been recently recognized as a very important source of genetic variability. Some CNV loci involve genes or conserved regulatory regions. Compelling evidence indicates that CNVs impact genome functions. The chicken is a very important farm animal species which has also served as model animal for biological and biomedical research for hundreds of years. A map of CNVs in chickens could facilitate the identification of chromosome regions that segregate for important agricultural and disease phenotypes. NimbleGen 385k whole genome tiling arrays were used to map CNVs in the chicken. This study has identified 96 CNVs in three lines of chickens (broiler, Leghorn and Rhode Island red). These CNVs encompass 16 Mb (1.3%) of the chicken genome. Twenty six CNVs were found in two or more animals. Smaller sized CNVs mostly affect none coding sequences while larger CNV regions involve genes, for example prolactin receptor, aldose reductase and zinc finger proteins, suggesting chicken CNVs potentially affect agricultural or disease related traits.

Effect of an immune challenge and feed supplements on broiler chicken individual breast muscle protein synthesis

Project description:Optimization of broiler chicken breast muscle protein accretion is key for the efficient production of poultry meat, whose demand is steadily increasing. In a context where antimicrobial growth promoters use is being restricted, it is important to find alternatives as well as to characterize the effect of immunological stress on broiler chicken growth. Despite of its importance, research on broiler chicken muscle protein dynamics has been mostly limited to the study of mixed protein turnover. The present study aims to characterize the effect of a bacterial challenge and the feed supplementation of a citrus and a cucumber extract on broiler chicken individual breast muscle proteins fractional synthesis rates (FSR) using a recently developed dynamic proteomics pipeline. 21 day-old broiler chickens were administered a single 2H2O dose before being culled at different timepoints. A total of 60 breast muscle protein extracts from five experimental groups (Unchallenged, Challenged, Control Diet, Diet 1 and Diet 2) were analyzed using a DDA proteomics approach. Proteomics data was filtered in order to reliably calculate multiple proteins FSR making use of a newly developed bioinformatics pipeline. Broiler breast muscle proteins FSR uniformly decreased following a bacterial challenge, this change was judged significant for 15 individual proteins, the two major functional clusters identified as well as for mixed breast muscle protein. Citrus or cucumber extract feed supplementation did not show any effect on the breast muscle protein FSR of immunologically challenged broilers. The present study has identified potential predictive markers of breast muscle growth and provided new information on broiler chicken breast muscle protein turnover which could be essential for improving the efficiency of broiler chicken meat production.
