Project description:Background: Despite endothelial dysfunction being considered as the initial step of the development of hypertension and associated cardio-renal syndromes, effective therapeutic strategies to prevent endothelial injuries are still lacking. GPR183 is a recently identified GPCR for oxysterols and hydroxylated metabolites of cholesterol. Here, we explored a previously unappreciated role of GPR183 in hypertension and age-related cardio-renal injuries. Methods and results: Mice were treated with DOCA/Salt or AngII to induce hypertension. we found that endothelial GPR183 was significantly induced in aged or hypertensive mice, which was further confirmed in renal biopsies from elderly people or subjects with hypertensive nephropathy. In addition, we found that endothelial-specific GPR183 deletion markedly ameliorated endothelial senescence and cardio-renal injuries in hypertensive mice. Mechanistically, we found that GPR183 disrupted circadian signaling by inhibiting PER1 expression, thereby facilitating endothelial senescence and dysfunction through the cAMP/PKA/CREB signaling pathway. Importantly, pharmacological inhibition of the oxysterol-GPR183 axis by NIBR189 or Clotrimazole ameliorated endothelial senescence and cardio-renal injuries in hypertensive mice. Conclusions: GPR183 is an attractive therapeutic target for hypertension and age-related diseases. Targeting GPR183 could provide novel strategies for managing hypertension and its associated cardio-renal injuries.

Project description:Hypertension is a long-term condition that can increase organ susceptibility to insults and lead to severe complications such as chronic kidney disease (CKD). Extracellular vesicles (EVs) are cell-derived membrane structures that participate in cell-cell communication by exporting encapsulated molecules to target cells, regulating physiological and pathological processes. We here demonstrate that multiple administration of EVs from adipose-derived mesenchymal stromal cells (ASC-EVs) in deoxycorticosterone acetate (DOCA)-salt hypertensive model can protect renal tissue by maintaining its filtration capacity. Indeed, ASC-EVs downregulated the pro-inflammatory molecules monocyte chemoattracting protein-1 (MCP-1) and plasminogen activating inhibitor-1 (PAI1) and reduced recruitment of macrophages in the kidney. Moreover, ASC-EVs prevented cardiac tissue fibrosis and maintained blood pressure within normal levels, thus demonstrating their multiple favorable effects in different organs. By applying microRNA (miRNA) microarray profile of the kidney of DOCA-salt rats, we identified a selective miRNA signature associated with epithelial-mesenchymal transition (EMT). One of the key pathways found was the axis miR-200-TGF-β, that was significantly altered by EV administration, thereby affecting the EMT signaling and preventing renal inflammatory response and fibrosis development. Our results indicate that EVs can be a potent therapeutic tool for the treatment of hypertension-induced CKD in cardio-renal syndrome.

Project description:We assigned carboxypeptidase X 2 (Cpxm2) to a genetic locus for left ventricular mass. The functional role of Cpxm2 was investigated in Cpxm2-deficient (KO) and wild-type (WT) mice exposed to deoxycorticosterone acetate (DOCA)-salt hypertension and control conditions (SHAM). Both WT and KO animals developed severe and similar systolic hypertension in response to DOCA. WT mice developed severe LV damage. These changes were significantly ameliorated or even normalized (i.e. ejection fraction) in KO-DOCA animals. LV transcriptome analysis in WT, but not in KO mice, showed a molecular cardiac hypertrophy/remodeling signature with significant upregulation of 1234 transcripts including Cpxm2 in response to DOCA.

Project description:Recent studies show that acute injury to non-aged podocytes induces many similarities to healthy aged podocytes, such as decreased lifespan and health-span. Like healthy aged podocytes, injury to young mouse and human podocytes can induce a senescent phenotype. This begs the question if injury to young podocytes phenocopies a healthy middle-aged podocyte, and if the pathways underlying senescence and other injury responses overlap between injured young podocytes and healthy middle-aged podocytes. To address this knowledge gap we induced hypertension, a major cause of chronic kidney disease, in young mice (4m of age~20-year-old human) and in middle-aged mice (18m of age, ~55+ year old human) with deoxycorticosterone and high salt (DOCA) and compared outcomes to non-hypertensive healthy middle-aged mice. In both healthy middle-aged mice and in young mice with hypertension, the increase in age-related senescent genes p16 and p19, along with the stress-related senescent genes p21 and p53 were similar. Bulk RNA-sequencing of podocytes showed that the senescent associated secretory phenotype and individual genes from several aging gene sets were also similar between middle-aged mice and young mice with hypertension. Of the highest enriched Hallmark pathways in middle-aged podocytes, 95% were also enriched in young mice treated with DOCA. Gene set enrichment analysis of podocytes showed that 36 genes overlapped between middle-aged mice, and young and middle-aged mice given DOCA, while 119 genes identified were “DOCA-specific”. These results suggest that hypertension in young mice induces podocyte injury and an aging/senescent phenotype that is similar to the one of podocytes from healthy middle-aged mice.

Project description:This study aimed to profile exosomal miRNAs in both plasma and cerebrospinal fluid (CSF) from male and female mice subjected to DOCA-salt treatment—a widely used model for inducing hypertension—and from their respective baseline controls. Exosomes were isolated using size exclusion chromatography (SEC), followed by small RNA extraction and quality control. Exosomal miRNAs were sequenced using the QIAseq miRNA Library Kit (Qiagen) on the Illumina NovaSeq 6000 platform. Libraries incorporated unique molecular identifiers (UMIs) to mitigate PCR bias. High-throughput sequencing data were analyzed for quality, aligned to multiple databases (miRBase, piRNABank, and Rfam), and normalized using CPM values. Differentially expressed miRNAs were identified and subjected to target prediction and pathway analysis using miRTarBase, GO, and KEGG. These findings may provide insight into the molecular signatures associated with hypertension and sex-specific responses in extracellular vesicle-derived miRNAs.
