We report the application of RNA-sequencing technology (Illumina Hi-Seq) for high-throughput profiling Arabidopsis thaliana transcriptomes upon infection with the model pathogen Pseudomonas syringae (wild-type and mutants).

Characterization of Arabidopsis thaliana MYB47 and MYB95 transcription factors in response to jasmonic acid in leaves.

Project description:We have found that Arabidopsis thaliana paralogous two MYBs, namely MYB47 and MYB95, regulate TSA1 and BGLU18 expression and ER-body formation in response to jasmonic acid (JA). To comprehensively understand the transcriptional regulation of these MYBs in response to JA, we conducted RNA-seq analysis between JA-treated rosette leaves of wild-type and myb47myb95 knockout mutants, together with myc2myc3myc4 mutant that is defective in master transcriptional regulator of JA signaling pathway. The transcriptome data analysis revealed that MYB47 and MYB95 regulate a subset of JA-responsive genes, including ER-body and defense-related genes.

Transcriptomic, proteomic and metabolic changes in Arabidopsis thaliana leaves after the onset of illumination

Project description:Arabidopsis thaliana ecotypes Columbia (Col-0) (wild type: WT) was used in this study. After sterilization, the seeds were placed on Murashige and Skoog medium supplemented with 2% (w/v) sucrose for 10 days and then the seedling were transferred to soil under 16 hours light (22°C) / 8 hours dark (18°C) period in growth chamber at a light intensity of 120?150 µmol m-2 s-1. 20-day-old Arabidopsis leaves without bolting were immediately frozen in liquid nitrogen for RNA and protein and metabolites extraction. Leaves were harvested at three different time points: t = 0 hr (end of night), t = 1 hr (one hour after light turn on) and t = 8 hr (eight hours after light turn on), respectively.

Chromosomal duplication in the grandifolia-D mutants of Arabidopsis thaliana, tiling array data

Project description:The number of cells in an organ is a major factor for the determination of organ size. However, genetic basis of cell number determination is not well understood. Three grandifolia-D (gra-D) mutants of Arabidopsis thaliana developed huge leaves containing two- to three-fold increased number of cells of the wild type. Tiling array and microarray analysis of gra-D mutants suggested that genes found in a lower part of chromosome 4 were upregulated, suggesting the occurrence of segmental chromosomal duplications in the gra-D mutants. These region contain positive regulators of cell proliferation such as AINTEGUMENTA (ANT) and cyclin genes such as CYCD3;1.
