Project description:In order to investigate the diurnal oscillations of ruminal protozoa, and their responses to the changes in different feeding patterns, we conducted an animal experiment by feeding the sheep ad libitum with a hay-based diet (50% of alfalfa hay and 46% of oats hay) and a grain-based diet (45% of corn meal and 11% of soybean meal) for 30 days, and ruminal fluid samples were collected at six different timepoints from T2 to T22 in one day, and the composition and diversity of the protozoal communities in rumen microbiomes of the sheep in the Grain-diet and Hay-diet groups at different timepoints were analyzed through 18S rRNA sequencing.

Project description:In the present study, we studied the effect of dietary selenium (Se) supplementation on the transcriptomic profile of sheep. The main objective was to evaluate the effect of Se-supplementation on the overall transcriptome of sheep, the altered pathways, and the biological processes related to it . A custom oligo microarray platform (AMADID: 070119) was designed, then used to profile gene expression from 20 samples from 10 sheep at two time points (T0; before Se-supplementation, and T40; at the end of a 40-d Se-supplementation period). Isolated and purified total RNAs were individually hybridized to the custom (4x44k) DNA microarray. The comparison of control and treated animal transcriptomes revealed a large set of differentially expressed genes. After functional analysis and qPCR validation, the result showed several pathways and biological processes that have been altered following Se-supplementation to the diet.

Project description:The possible benefits of selenium (Se) supplementation are currently under investigation for prevention of certain cancers and treatment of neurological disorders. Little is known concerning the response of the brain to increased dietary Se under conditions of Se sufficiency, despite the majority of Se supplementation trials occurring in healthy subjects considered Se sufficient. We evaluated the transcriptional response of the zebrafish (Danio rerio) brain to supplementation with nutritionally relevant levels of dietary Se (sodium selenite) during conditions of assumed Se sufficiency. We used a microarray approach to analyze the global gene expression response of the brain to dietary Se supplementation for 14 days. The experiment used Affymetrix microarrays to compare whole brain RNA from 8 adult zebrafish (Danio rerio) fed a diet with control selenium levels (1.4ppmSe) and 8 fed a diet supplemented with sodium selenite (5.6ppmSe) for 14 days, and with an equal sex ratio within each diet.

Project description:PURPOSE: Previous mouse studies using corn oil (ω-6) as the dietary fat source suggest that decreasing dietary fat content can slow prostate cancer (PCa) growth. However, other studies, in which the diet was composed around saturated fat, showed no difference in outcomes between high-fat and low-fat diets. The relative effects of other fats, such as fish oil and olive oil, also remain unexplored. To our knowledge, no trial has yet compared the effect of various fats on prostate cancer progression. Therefore, we sought to systematically study the effect of fish oil, olive oil, corn oil, and saturated fat on prostate cancer progression. METHODS: A total of 96 male SCID mice were injected with LAPC-4 human PCa cells. Two weeks following injection, mice were singly-housed and randomized to either a fish oil, olive oil, corn oil, or saturated fat based diet. Animals were euthanized when tumors reached 1,000 mm3. Serum was collected at sacrifice and assayed for PSA, insulin, IGF-1, IGFBP-3, and PGE-2 levels. Tumors were also assayed for PGE-2, and COX-2 levels, and gene array analysis was performed. RESULTS: Mice weights and tumor volumes were equivalent across groups at randomization. Overall, fish-oil consumption was associated with improved survival, relative to all other dietary groups (Log-rank, all p<0.05). We did not detect any significant difference in serum PSA, insulin, IGF-1, IGFBP-3, and PGE-2 levels. Glucose at the time of sacrifice was statistically different between groups, with the fish-oil fed mice having the highest levels of serum glucose (Kruskal-Wallis, p=0.03). CONCLUSIONS: In this prostate cancer xenograft model, we found that consuming a diet in which fish-oil was the only fat source slowed tumor growth in improved survival, compared to mice consuming diets composed of olive oil, corn oil, or saturated fat sources. These results suggest that type of dietary fat consumed may be as important as amount of dietary fat consumed in the setting of prostate cancer.

Project description:<p>Background: With the pursuit of rapid intramuscular fat deposition in beef cattle, basal diets have increasingly shifted toward grain-based, energy-dense formulations. However, high-starch dietary regimens may induce host metabolic dysregulation, which is closely linked to both ruminal microbial dysbiosis and systemic inflammatory responses. Guanidinoacetic acid (GAA) and taurine (TAU), two endogenous bioactive compounds, have garnered significant interest for their anti-inflammatory and antioxidant effects. This study applied a multi-omics approach to analyze the effects of GAA and TAU supplementation on ruminal microorganisms, ruminal metabolites, blood metabolites, and antioxidant and inflammatory indicators in both rumen and blood. It also aimed to explore the underlying biological functions and mechanisms for GAA and TAU in fattening cattle fed a high-starch diet.</p><p>Results TAU supplementation significantly decreased the concentrations of lipopolysaccharide (LPS), intestinal permeability markers, proinflammatory cytokines, and oxidative stress markers in the plasma of fattening cattle fed high-starch diets. Moreover, TAU supplementation modified the rumen microbiota and enhanced the levels of ruminal propionate and butyrate. Notably, the potentially beneficial bacteria taxa Lachnospiraceae_UCG_008, Roseburia, Anaerovorax, and Christensenellaceae_R_7_group were found to be upregulated following TAU supplementation and showed positive correlations with rumen metabolites and host metabolites. Untargeted metabolomics of rumen and plasma demonstrated that TAU supplementation upregulated the abundance of metabolites related to intestinal mucosal repair and anti-inflammatory effects, including phosphocholine, proline, rumenic acid, L-pyrrolidone, L-pipecolate, and myristoleic acid. Differential metabolites (including butyrate) showed significant correlations with plasma diamine oxidase (DAO), D-lactic acid (D-LA), TNF-α, IL-6, malondialdehyde (MDA), superoxide dismutase (SOD), and rumen probiotics. Moreover, TAU participated in the regulation of amino acid, carbohydrate, lipid, and bile acid-related metabolic pathways in both rumen and plasma, showing potential benefits for improving nitrogen and energy utilization efficiency. Another noteworthy finding is that the Christensenellaceae_R_7 group and the associated plasma biosynthesis pathways of unsaturated fatty acids suggest a potential enhancement in the unsaturated fatty acid content of meat. GAA supplementation decreased the concentration of malondialdehyde (MDA) while increasing the activity levels of superoxide dismutase (SOD), glutathione peroxidase (GSH-Px), and total antioxidant capacity (T-AOC) in plasma, thereby effectively mitigating oxidative stress in the host. However, it exerted minimal influence on the composition of ruminal microorganisms and the profiles of plasma metabolites, and no significant correlation was observed between differential microorganisms or metabolites and the improvement of oxidative stress markers.</p><p>Conclusion: Our study indicates that TAU supplementation effectively mitigates systemic inflammation and oxidative stress in fattening cattle fed high-starch diets, primarily through the modulation of rumen microbial communities and their associated metabolites. In contrast, while GAA also shows a beneficial effect on alleviating host oxidative stress, this effect appears to be independent of alterations in microbial metabolic pathways.</p>

Project description:A healthy rumen is crucial for normal growth and improved production performance of ruminant animals. Rumen microbes participate in and regulate rumen epithelial function, and the diverse metabolites produced by rumen microbes are important participants in rumen microbe-host interactions. SCFAs, as metabolites of rumen microbes, have been widely studied, and propionate and butyrate have been proven to promote rumen epithelial cell proliferation. Succinate, as an intermediate metabolite in the citric acid cycle, is a final product in the metabolism of certain rumen microbes, and is also an intermediate product in the microbial synthesis pathway of propionate. However, its effect on rumen microbes and rumen epithelial function has not been studied. It is unclear whether succinate can stimulate rumen epithelial development. Therefore, in this experiment, Chinese Tan sheep were used as experimental animals to conduct a comprehensive analysis of the rumen microbiota community structure and rumen epithelial transcriptome, to explore the role of adding succinate to the diet in the interaction between the rumen microbiota and host.
