Project description:Long-term high-fat diet feeding increases the circulating concentration of stearic acid (SA), which has a potent toxic effect on β-cells, but the underlying molecular mechanisms have not been fully elucidated. Here, we evaluated the role of lncRNA TCONS_00077866 (lncRNA-866) in SA-induced β-cell inflammation. lncRNA-866 was selected for study because it demonstrated the largest fold-difference in expression of five lncRNAs that were affected by SA treatment using lncRNA High-throughput Sequencing analysis. Knockdown of lncRNA-866 by virus-mediated shRNA expression in mice or Smart Silencer in β-TC6 cells significantly inhibited the SA-induced reduction in insulin secretion and β-cell inflammation. lncRNA-866 directly bound to miR-297b-5p, thereby preventing it from reducing the expression of its target serum amyloid A3 (SAA3), according to lncRNA-miRNA-mRNA co-expression network analysis and luciferase reporter assay. Furthermore, overexpression of miR-297b-5p or inhibition of SAA3 also had marked protective effects against the deleterious effects of SA in β-TC6 cells and mouse islets. In conclusion, lncRNA-866 silencing ameliorates SA-induced β-cell inflammation by targeting the miR-297b-5p/SAA3 axis. lncRNA-866 inhibition may represent a new strategy to protect β-cells against the effects of SA during the development of type 2 diabetes.

Manipulation of Mir140 3p and 5p expression

Project description:The goal of this study was to change the stoichiometry of Mir140-3p and Mir140-5p expression, thereby increasing the expression levels of Mir140-5p, and observe the biological effects on skeletal development. This was done by introducing two types of nucleotide changes into the seed regions of Mir140. The first was by changing the initial bases of Mir140-3p from 'TACCACAGGGTAGAACCACGGACAGGGTACTGGAGC' to 'CGCCACAGGGTAGAACCACGGACAGGGTACTGGAGC' lowering the expression levels of Mir140 3p and disrupting its function. The second change to alter Mir140 expression was by changing the Mir140-5p seed region from 'CAGTGGTTTTACCCTATGGAGG' to 'TAGTGGTTTTACCCTATGGAGG' increasing mi140-5p's ability to be bound by Argonaute.

Project description:From a previous microarray study we developed a small chondrogenesis model. We performed qPCR and measured how knockdown of miR-199a-5p or miR-199b-5p could modulate chondrogenesis. Several experiments were used to determine the parameters of this model. We utilised parameter scan and manual sliding to refine the model. Within are two models - an initial model which only comprises of genes which we have data for, and an enhanced model which expands of the initial model to make more predictions - e.g. how miR-140-5p is indirectly regulated by miR-199a-5p and miR-199b-5p.
