Project description:Soil microorganisms play an irreplaceable role in agricultural production, however, an understanding of response of soil microorganisms to slow-release and common fertilizer applications is limited. In this study, different amounts of slow- release fertilizer were used to overwintering Chinese chives growing area in a plastic greenhouse to investigate the effects of on rhizosphere soil physicochemical properties and soil microbial communities (bacteria and fungi) of Chinese chives. The result displayed that application of slow-release fertilizer significantly improved soil nutrients, soil enzyme activity, and soil microbial community structure and diversity compared to conventional fertilizer application. Compared with T1 treatment, the content of total nitrogen (TN) and available phosphorus (AP), and the SU-E activity in the soil of T2 (NPK: 62.8 kg · 667 m-2) increased by 42.58%, 16.67%, and 9.70%, respectively, showing the best effects. In addition, soil bacterial diversity index and soil microbial community structure were improved as indicated by increased relative abundance of each species, such as Byssovorax, Sandaracinus, and Cellvibrio. Oppositely, the both soil fungal diversity and the number of species decreased after fertilizationthe relative abundance of Ascomycota increased in each fertilization treatment detected by ITS sequencing. Further, the relative abundance of pathogenic fungi such as Pezizomycetes, Cantharellales, and Pleosporales decreased in the T2 treatment. Principal Coordinates Analysis (PCoA) showed that both the amount of fertilizer applied and the type of fertilizer applied affected the soil microbial community structure. RDA evidenced that soil bacteria, Proteobacteria and Gemmatimonadetes, were closely correlated with soil AN, SOM, and AK. Acidobacteria were closely correlated with soil pH, TN, and AP. Ascomycota was closely correlated with soil pH and TN. In conclusion, the application of slow-release fertilizers and reduced fertilizer applicationcould improve soil physical and chemical properties as well as soil microbial community structure and diversity, contributing to sustainable soil development. The recommended fertilization rate for overwintering Chinese chives is NPK: 62.8 kg · 667 m-2.
Project description:Despite the lower reactivity of natural phosphates compared to soluble fertilizers, their P bioavailability can increase over the cultivation years, due to the physicochemical processes and the activity of soil microbiota. Therefore, this work aimed to evaluate the α and β diversity of the rhizosphere microbiota of maize and sorghum genotypes grown under different sources and doses of phosphate fertilizers. Four commercial maize and four sorghum genotypes were grown under field conditions with three levels of triple superphosphate (TSP) and two types of rock phosphate sources: phosphorite (RockP) and bayóvar (RP) during two seasons. Maize and sorghum presented a significant difference on the genetic β diversity of both rhizosferic bacterial and arbuscular mycorrhizal fungi. Moreover, P doses within each phosphate source formed two distinct groups for maize and sorghum, and six bacterial phyla were identified in both crops with significant difference in the relative abundance of Firmicutes and Proteobacteria. It was observed that RockP fertilization increased Firmicutes population while Proteobacteria was the most abundant phylum after TSP fertilization in maize. In sorghum, a significant impact of fertilization was observed on the Acidobacteria and Proteobacteria phyla. TSP fertilization increased the Acidobacteria population compared to no fertilized (P0) and RockP while Proteobacteria abundance in RockP was reduced compared to P0 and TSP, indicating a shift toward a more copiotrophic community. Our results suggested that the reactivity of P source is the predominant factor in bacterial community' structures in the maize and sorghum rhizosphere from the evaluated genotypes, followed by P source.

Project description:We present metaproteome data from maize rhizosphere from sodic soil. Isolation of proteome from maize rhizosphere collected from Experimental Farm, ICAR-IISS, Mau, India was done with the standardized protocol at our laboratory and metaproteome analysis was done with the standardized pipepline. In total 696 proteins with different functions representing 245 genus and 395 species were identified. The proteome data provides direct evidence on the biological processes in soil ecosystem and is the first reported reference data from maize rhizosphere.
Project description:IntroductionThis article endeavors to investigate the influence of various fertilization methods on the characteristics of rhizosphere soil and organic carbon mineralization in the mixed soil of Mu Us Sandy land under maize cultivation, with the objective of laying the groundwork for low-carbon agriculture and the development of high-quality farmland.MethodsThe research focuses on soft rock and sand composite soil with a 1:2 ratio, and it comprises four treatments: no fertilization (CK), only chemical fertilization (CF), only cattle manure application (MF), and only oil residue application (DF).ResultsThe findings revealed that the use of organic fertilizer substantially elevated nutrient content and enzyme activity in the maize rhizosphere soil. Furthermore, it had a notable influence on both soil aggregate diameter and stability. Specifically, the DF treatment led to a significant increase in both soil aggregate diameter and stability. The mineralization rate of organic carbon in the maize rhizosphere soil could be categorized into two distinct phases: a rapid initial decline followed by a slower release. By the end of the incubation period, the cumulative mineralization of organic carbon in the MF, DF, and CF treatments showed a significant increase of 119.87%, 57.57%, and 24.15%, respectively, in comparison to the CK treatment. Additionally, the mineralization rate constants of the DF and MF treatments experienced a substantial rise, with increments of 23.52% and 45.97%, respectively, when contrasted with the CK treatment. The bacterial phyla Actinobacteriota, Proteobacteria, Chloroflexi, Acidobacteriota, and Firmicutes were dominant in the rhizosphere soil bacterial community. Specific genera such as Nocardioides and Sphingomonas showed significant correlations with organic carbon mineralization. The application of different organic fertilizer can improve soil physical, chemical and biological properties, and promote the mineralization process of organic carbon in maize rhizosphere soil.DiscussionNotably, the DF treatment exhibited the most favorable outcome, improving the overall quality of maize rhizosphere soil while incurring a minimal loss of unit organic carbon. These findings hold significant implications for optimizing field management practices and augmenting soil quality.
Project description:The rhizosphere is a critical interface supporting the exchange of resources between plants and their associated soil environment. Rhizosphere microbial diversity is influenced by the physical and chemical properties of the rhizosphere, some of which are determined by the genetics of the host plant. However, within a plant species, the impact of genetic variation on the composition of the microbiota is poorly understood. Here, we characterized the rhizosphere bacterial diversity of 27 modern maize inbreds possessing exceptional genetic diversity grown under field conditions. Randomized and replicated plots of the inbreds were planted in five field environments in three states, each with unique soils and management conditions. Using pyrosequencing of bacterial 16S rRNA genes, we observed substantial variation in bacterial richness, diversity, and relative abundances of taxa between bulk soil and the maize rhizosphere, as well as between fields. The rhizospheres from maize inbreds exhibited both a small but significant proportion of heritable variation in total bacterial diversity across fields, and substantially more heritable variation between replicates of the inbreds within each field. The results of this study should facilitate expanded studies to identify robust heritable plant-microbe interactions at the level of individual polymorphisms by genome wide association, so that plant-microbiome interactions can ultimately be incorporated into plant breeding.
Project description:IntroductionThe selection and application of nitrogen-efficient maize hybrids have significantly bolstered contemporary food security. Nevertheless, the effects of heightened nitrogen fertilizer demand of these crops on the composition and assembly of soil microbial communities in agricultural production require further elucidation.MethodsIn this study, the effects of four nitrogen fertilizer managements on rhizosphere bacterial and fungal community assembly, co-occurrence network and function of two maize hybrids (LD981 and DH605) were compared.Results and discussionFindings revealed that the bacterial community was primarily shaped by deterministic processes, while stochastic processes played a pivotal role in fungal community assembly. N-efficient hybrid DH605 had a more stable microbial network than N-inefficient hybrid LD981. At N3 (130 g N/m2) rate, the bacterial and fungal community networks were the most complex but unstable, followed by N2 (87 g N/m2), N0 (0 g N/m2), and N1 (43 g N/m2) rates. Excessive nitrogen rate (N3) increased the relative abundance of denitrification genes nirK and norB by enriching nitrogen-related genus such as Nitrolancea and Nitrosospira. It led to an increase in the relative abundance of pathways such as cysteine and methionine metabolism and pyruvate metabolism. The effects of management practices (i.e. maize hybrids and N rates) on microbial communities were ultimately directly or indirectly reflected in microbial functions. Our findings illustrate the relationship between the appropriate selection of crop hybrids and management measures in optimizing rhizosphere microbial community assembly and promoting nitrogen use, which is necessary for sustainable food security.
