Project description:Amniotic fluid (AF) reflects fetal and placental physiology and provides a valuable window into intrauterine development. However, the extent to which fetal sex influences the mid-gestation AF proteome remains poorly characterized. The present study applied a data-independent acquisition mass spectrometry (DIA-MS) approach to AF samples collected at 16–18 weeks of gestation from pregnancies carrying male and female fetuses. Approximately 1,700 proteins were identified in AF_male and AF_female samples.

Global Gene Expression Analysis of Term Amniotic Fluid Cell-Free Fetal RNA

Project description:The objective of this study was to identify the tissue expression patterns and biological pathways enriched in term amniotic fluid cell-free fetal RNA by comparing functional genomic analyses of term and second-trimester amniotic fluid supernatants. There were 2,871 significantly differentially regulated genes. In term amniotic fluid, tissue expression analysis showed enrichment of salivary gland, tracheal, and renal transcripts as compared with brain and embryonic neural cells in the second trimester. Functional analysis of genes upregulated at term revealed pathways that were highly specific for postnatal adaptation such as immune function, digestion, respiration, carbohydrate metabolism, and adipogenesis. Inflammation and prostaglandin synthesis, two key processes involved in normal labor, were also activated in term amniotic fluid. This was a prospective whole genome microarray study comparing eight amniotic fluid samples collected from eight women at term who underwent prelabor cesarean delivery and eight second-trimester amniotic fluid samples from routine amniocenteses. A functional annotation tool was used to compare tissue expression patterns in term and second-trimester samples. Pathways analysis software identified physiologic systems, molecular and cellular functions, and upstream regulators that were significantly overrepresented in term amniotic fluid.

DNA hypermethylation in placental tissues: placental tissues vs. maternal peripheral bloods

Project description:Noninvasive prenatal diagnosis currently used does not achieve desirable levels of sensitivity and specificity. Recently, fetal methylated DNA biomarkers in maternal whole blood have been explored for noninvasive prenatal detection. However, such efforts cover only chromosomal aneuploidy; fetal methylated DNA biomarkers for detecting single-gene disease remain to be discovered. To address this issue, we systematically screened significantly hypermethylated genes in fetal tissues compared with maternal blood for noninvasive prenatal diagnosis of various inherited diseases. First, Methylated-CpG island recovery assay combined with CpG island array was performed in four maternal peripheral bloods and their corresponding placental tissues. Subsequently, direct bisulfite sequencing and combined bisulfite restriction analysis (COBRA) were carried out to validate the reliability of methylation microarray analysis. As results, 310 significantly hypermethylated genes in fetal tissues were detected by microarray. Two of five randomly selected hypermethylated genes detected by microarray were confirmed to be hypermethylated in fetal tissue samples by direct bisulfite sequencing. All four randomly selected hypermethylated genes detected by microarray were confirmed to be hypermethylated in five independent amniotic fluid samples and five independent chorionic villus samples from 10 pregnant women by CORBA. In conclusions, We found a lot of hypermethylated genes and methylation sites in fetal tissues, some of which have great potential to be developed into molecular markers for noninvasive prenatal diagnosis of monogenic disorders. Amniotic-fluid metabolomics identifies phospholipid remodeling as a metabolic signature of intrauterine exposure in pregnancies with polycystic ovary syndrome

Project description:Background: Polycystic ovary syndrome is associated with metabolic and hormonal disturbances during pregnancy, but whether these alterations are reflected in the fetal intrauterine exposure environment remains incompletely understood. This study aimed to identify polycystic ovary syndrome-related intrauterine metabolic signatures using late-gestation amniotic fluid. Methods: Untargeted metabolomic profiling was performed on late-gestation amniotic fluid samples from women with polycystic ovary syndrome and controls. Differential metabolite analysis, pathway-level analysis, and multilevel sensitivity analyses were conducted to identify robust metabolic alterations associated with maternal polycystic ovary syndrome. Exploratory placental transcriptomic analysis was further used to provide tissue-level evidence for lipid-related metabolic alterations.

Placental TRPV2 supports trophoblast function and normal fetal development [MERSCOPE]

Project description:While placental functioning arbitrates fetal outcome, the molecular players that underpin normal placental development remain poorly understood. Here, we provide evidence that the Transient Receptor Potential Vanilloid 2 (TRPV2) channel plays an important role in normal placental development, determining fetal size and survival. Its absence severely affects placental morphology, causing fetal growth restriction and embryonic lethality, which was found to be allocated to a non-redundant role in the trophoblast, but not in the embryo. CRISPR/Cas9-mediated Trpv2 deletion in mouse trophoblast stem cells demonstrated that Trpv2 contributes to the differentiation towards Syncytiotrophoblast layer II (SynTII) cells, which was confirmed by the growth restriction phenotype in SynTII-specific Trpv2 knockout animals. Finally, TRPV2 was functionally expressed in human trophoblast cells and enriched in trophoblast populations undergoing syncytial differentiation. Together, our findings identify an important role for TRPV2 during placental development and consequently fetal outcomes.
