Proteome changes in retinal Mueller glial cells from a spontaneous equine model for autoimmune uveitis compared to healthy controls

Project description:Retinal Müller glial cells (RMG) play a central role in retinal neuroinflammation, actively participating in the immune response and adopting a dynamic inflammatory phenotype. However, the precise functions of RMG in retinal inflammation remain incompletely understood. In this study, we investigated the proteome of RMG isolated from healthy horses and horses affected by equine recurrent uveitis (ERU). ERU serves as a unique spontaneous model of autoimmune uveitis, exhibiting key clinical and pathophysiological features observed in humans, such as a relapsing-remitting course of disease and a CD4+ T cell-driven autoimmune etiology [1, 2]. To gain insights into the molecular landscape of RMG in a spontaneous animal model of autoimmune uveitis and their potential contribution to retinal autoimmune neuroinflammation, we used discovery proteomics for the analysis of RMG from healthy horses and horses with ERU. Our data are not only relevant to veterinary medicine but also hold significant translational value for human ophthalmology as well. The equine granulocyte proteome: expression changes in autoimmune disease and after stimulation

Project description:We investigated the granulocyte proteome from healthy horses, horses with a spontaneous autoimmune disease (equine recurrent uveitis) and the reaction of granulocytes to different stimuli (IL8, PMA, LPS) on protein level. A total of 2032 proteins describing the whole granulocyte proteome were identified, including 245 proteins (12% of proteome) newly associated to in vivo expression in primary granulocytes (hypothetical proteins). Our studies provide novel information on the granulocyte proteome and contribute to a better understanding of molecular mechanisms involved in activation and recruitment of neutrophils and the role of these cells in modulating and regulating autoimmune diseases. The data presented here is highly relevant for veterinary medicine and has translational quality for spontaneous autoimmune uveitis in man.

CMTM4 is a subunit of the IL-17 receptor and mediates autoimmune pathology

Project description:Interleukin-17A (IL-17A) is a key mediator of protective immunity to yeast and bacterial infections but also drives the pathogenesis of several autoimmune diseases, such as psoriasis or psoriatic arthritis. Here, we show that the tetra-transmembrane protein CMTM4 is a subunit of the IL-17 receptor (IL-17R). CMTM4 constitutively associated with IL-17R subunit C (IL-17RC) to mediate its stability, posttranslational modification, and plasma membrane localization. Both mouse and human cell lines deficient in CMTM4 were largely unresponsive to IL-17A, due to their inability to assemble the IL-17 receptor signaling complex. Accordingly, CMTM4-deficient mice were largely resistant to experimental psoriasis. Collectively, our data identified CMTM4 as an essential component of the IL-17 receptor and a potential therapeutic target for treating IL-17-mediated autoimmune diseases.

Regulatory T cells targeting a pathogenic MHC class II: Insulin peptide epitope postpone spontaneous autoimmune diabetes

Project description:In spontaneous type 1 diabetes (T1D) non-obese diabetic (NOD) mice, the insulin B chain peptide 9-23 (B:9-23) can bind to the MHC class II molecule (IAg7) in register 3 (R3), creating a bimolecular IAg7/InsulinB:9-23 register 3 conformational epitope (InsB:R3). Previously, we showed that the InsB:R3-specific chimeric antigen receptor (CAR), constructed using an InsB:R3-monoclonal antibody, could guide CAR-expressing CD8 T cells to migrate to the islets and pancreatic lymph nodes. Regulatory T cells (Tregs) specific for an islet antigen can broadly suppress various pathogenic immune cells in the islets and effectively halt the progression of islet destruction. Therefore, we hypothesized that InsB:R3 specific Tregs would suppress autoimmune reactivity in islets and efficiently protect against T1D. To test our hypothesis, we produced InsB:R3-Tregs and tested their disease-protective effects in spontaneous T1D NOD CD28-/- mice. InsB:R3-CAR expressing Tregs secrete IL-10 dominated cytokines upon engagement with InsB:R3 antigens. A single infusion of InsB:R3 Tregs delayed the onset of T1D in 95% of treated mice, with 35% maintaining euglycemia for two healthy lifespans, while whereas control Tregs did not. Our data demonstrate that Tregs specific for MHC class II: Insulin peptide epitope (MHCII/Insulin) protect mice against T1D more efficiently than polyclonal Tregs lacking islet antigen specificity, suggesting that the MHC II/insulin-specific Treg approach is a promising immune therapy for safely preventing T1D.

Several proteins showed changed abundance in horses affected by a spontaneous autoimmune disease (equine recurrent uveitis, ERU). Findings provide novel knowledge about changes in the CD4+ immune cell membrane proteome in a spontaneously and naturally occurring autoimmune disease in horses. The data is highly relevant for veterinary medicine and has proven translational quality for autoimmune uveitis in man.

In this study, pooled sera from six 8-10 month old BXD2 mice was profiled for anti-mouse IgG (H+L) analysis of autoantibody reactivity to peptide auto-epitopes printed in duplicate on a PepperPrint Chip.
