Single-cell RNA sequencing reveals diverse cell populations and transcriptional differences during skeletal muscle development in Megalobrama amblycephala

Project description:Fish skeletal muscle plays a crucial role in various physiological functions, and providing an important source of meat for human consumption. Therefore, understanding the molecular genetic regulation of muscle development and growth can benefit in enhancing the efficacy of aquaculture. Morphological and skeletal muscle histological analysis of Megalobrama amblycephala at 14 development stages ranging from 5-240 days post-hatching (d) revealed that 30 d and 45 d are the crucial stages for postembryonic muscle fibers hyperplasia and hypertrophy in M. amblycephala, respectively. Then we utilized single cell RNA sequencing (scRNA-seq) to investigate the regulation mechanism of postembryonic muscle development in M. amblycephala skeletal muscle at 30 d and 45 d. Here, scRNA-seq obtained 45,572 cells and identified a total of 13 distinct cell types in the skeletal muscle tissue. We reconstructed the cell differentiation events using all the cell clusters by single cell pseudo time trajectories, and the differentiation trajectory indicates that skeletal muscle cells derive from mesenchymal stem cells (MSCs). Furthermore, we analyzed the cell-specificity of skeletal muscle-related cells and their differences at different developmental stages, and identified a total of 51 crucial genes for muscle development, including 9 genes from muscle progenitors, 35 genes from myoblasts, and 7 genes from skeletal myocytes. Cell-cell communication analysis indicated that IGF, VEGF, and SEMA3 signaling pathways serve as important incoming signaling pathways for MSCs at 30 d. Skeletal myocytes (SMs) and tendon cells etc., might influence the differentiation of MSCs into myogenic lineages via Igf2b - Igf1ra and Igf1 - Igf1ra ligand - receptor interaction. This study revealed a list of new genes involved in M. amblycephala postembryonic myogenesis, and will provide the information for the molecular breeding of muscle mass trait in M. amblycephala.

Comparison of gene expression signatures in human melanocytes after indirect cold physical plasma treatment and direct UV-irradiation

Project description:Cold atmospheric plasma is assembled out of various components such as UV-radiation, electrons, ions and an electromagnetic field at low temperature. In recent years cold atmospheric plasma gains interest as a medical tool used for improved healing of chronic wounds. In order to examine the impact of cold atmospheric plasma on the melanogenesis in human skin, the gene expression signature of differently treated lightly pigmented primary human melanocytes was analysed. Melanocytes were either treated in vitro indirectly by cold atmospheric plasma or irradiated by UV-radiation. After 2 h incubation the microarray analysis was performed.

Atmospheric and Room Temperature Plasma (ARTP) Mutagenesis Improved the Anti-MRSA Activity of Brevibacillus sp. SPR20

Project description:Brevibacillus sp. SPR20 produced potentially antibacterial substances against MRSA. The synthe-sis of these substances is controlled by their biosynthetic gene clusters. Several mutagenesis methods are used to overcome the restriction of gene regulations when genetic information is ab-sent. Atmospheric and room temperature plasma (ARTP) is a powerful technique to make random mutagenesis for microbial strain improvement. This study utilized an argon-based ARTP to conduct the mutations on SPR20. The positive mutants of 40% occurred. The M27 mutant exhib-ited an increase in anti-MRSA activity when compared to the wild-type, by which the MIC values were 250500 and 500 g/mL, respectively. M27 had genetic stability because it exhibited con-stant activity throughout 15 generations. This mutant had similar morphology and antibiotic susceptibility to the wild-type. Comparative proteomic analysis identified some specific proteins that were upregulated in M27. These proteins were involved in the metabolism of amino acids, cell structure and movement, and catalytic enzymes. These might result in the enhancement of the anti-MRSA activity of the ARTP-treated SPR20 mutant. This study supports the ARTP technology to increase the production of valuable antibacterial agents.
