Zinc Transporter 8 Haploinsufficiency Protects Against Beta Cell Dysfunction in Type 1 Diabetes by Increasing Mitochondrial Respiration

Project description:Zinc transporter 8 (ZnT8) is a major humoral target in human type 1 diabetes (T1D). Polymorphic variants of Slc30A8, which encodes ZnT8, are also associated with protection from T2D, suggesting that ZnT8 might play a role beyond simply being a target of autoimmunity in the pathophysiology of T1D. To begin to investigate this possibility, we transferred a global Slc30a8 null allele to the well-established NOD mouse model of T1D. Unexpectedly, complete loss of ZnT8 accelerated spontaneous T1D, while heterozygosity was partially protective. In vivo and in vitro studies using ZnT8 mutants on a NOD.scid background suggested that the accelerated disease was due to more rampant autoimmunity. Conversely, beta cells in heterozygous animals uniquely displayed increased mitochondrial fitness under mild proinflammatory conditions. In pancreatic beta cells and immune cell populations, Zn2+ plays a key role as a regulator of redox signaling and as an independent secondary messenger. Importantly, Zn2+ also plays a major role in maintaining mitochondrial homeostasis. Our results suggest that regulating mitochondrial fitness by altering intra-islet zinc homeostasis may provide a novel mechanism to modulate T1D pathophysiology.

Adaptation of Cupriavidus metallidurans CH34 to toxic zinc concentrations involves an uncharacterized ABC-type transporter

Project description:Cupriavidus metallidurans CH34 is a well-studied metal-resistant β-proteobacterium and contains a battery of genes involved in the resistance and processing of numerous metals. Here, we performed a directed evolution experiment to obtain derivatives adapted to grow in the presence of zinc at concentrations above the minimal inhibitory concentration for the parental strain. Genetic characterization of one derivative, CH34ZnR, which was also more resistant to cadmium, revealed seven mutations, including inactivation of glpR coding for a DeoR-type transcriptional repressor by insertion of IS1088. The latter resulted in the constitutive expression of the neighboring ABC-type transporter. GlpR and the adjacent ABC transporter are highly similar to the glycerol operon regulator and ATP-driven glycerol importer of Rhizobium leguminosarum bv. viciae VF39, respectively. Deletion of glpR or the ABC transporter and complementation of CH34ZnR with the parental glpR gene further demonstrated that loss of GlpR function, with concomitant induction of the adjacent ABC transporter, is pivotal in the observed phenotype. Moreover, addition of glycerol, presumably by glycerol-mediated reduced GlpR activity, also promoted increased zinc and cadmium resistance. Thus, the ABC-type transporter is proposed to be a new adaptation route during exposure to toxic zinc concentrations.

Zinc limitation in Klebsiella pnuemoniae

Project description:Nutrient adaptation is key in limiting environments for the promotion of microbial growth and survival. In microbial systems, zinc is an important component for many cellular processes and bioavailability varies greatly among different conditions. In the bacterium, Klebsiella pneumoniae, the impact of zinc limitation has not been explored at the protein level. Here, we apply a mass spectrometry-based quantitative proteomics strategy to profile the global impact of zinc limitation on the cellular proteome and extracellular environment (secretome) of K. pneumoniae. Our data defines the impact of zinc on proteins involved in transcriptional regulation and emphasizes the modulation of a vast array of proteins associated with zinc acquisition, transport, and binding. We also identify proteins in the extracellular environment associated with conventional and nonconventional modes of secretion. In particular, we focus on the impact of zinc on an uncharacterized cation transporter and the role of the ChaB transporter in influencing bacterial growth, capsule production, nad virulence. Moreover, we validate our results through transcriptional profiling of genes reprsessed under high abudnance of a histidine repression operon, hutC. Overall, we provide evidence of novel connections between zinc availability and capsule production in a bacterial system and define new roles for a transporter.
