Time series of motor neuronal translatomes over the course of neuroinflammation

Project description:During the course of multiple sclerosis (MS), inflammatory insults drive neuro-axonal loss and disability progression. Here we leverage translating ribosome affinity purification (TRAP) to extract ribosome-bound mRNA from Chat-positive motor neurons of mice undergoing experimental autoimmune encephalomyelitis (EAE), the animal model of MS. This unique dataset allows to follow the temporal dynamics of neuronal responses to inflammation and enables the extractions of molecular targets for therapeutic intervention.

CCDC88B affinity proteomic analysis

Project description:The CCDC88B gene (11q13) is a risk factor for several chronic inflammatory diseases in humans, including inflammatory bowel disease (IBD), multiple sclerosis and psoriasis. A Ccdc88b mutation in mice conveys protection against pathological inflammation in experimental models of microbial (cerebral malaria) and autoimmune (experimental autoimmune encephalomyelitis) of neuroinflammation and colitis. At the cellular level, loss of Ccdc88b function is phenotypically expressed as a migratory defect in vivo, and a cell mobility defect in vitro in mutant dendritic cells and lymphocytes. At the molecular level, CCDC88B belongs to a family of protein scaffold associated with the cytoskeleton and that feature protein:protein interactions domains. We used a co-immunoprecipitation / mass spectrometry-based approach to identify physical interactions of CCDC88B from mouse thymocytes and BI-141 cells.

Peli1 knock-out and wild type mouse brain proteomes during the disease course of experimental autoimmune encephalomyelitis

Project description:The ubiquitin ligase Peli1 has previously been suggested as a potential treatment target in multiple sclerosis since the knock-out induced less activated microglia and less inflammation in the CNS of experimental autoimmune encephalomyelitis in mice. In the present study the brain proteomes of Peli1 knock-out mice and wild-type mice were analyzed and compared before disease induction and after 10 and 20 days of experimental autoimmune encephalomyelitis using quantitative proteomics. The brain samples were analyzed using TMT labeling of small pools of samples and verified using label-free of individual mice.
