Project description:Foxf1 is enriched and differentially expressed in lung cells derived from mesoderm. We used single cell RNA sequencing (snRNAseq + snATAC) to analyze the dynamic transcriptional regulation in various lung cell types. The single cell analysis show the cell specific regulation of Foxf1 by distal enhancers that contribute to the molecular pathology of ACDMPV.

Foxf1-specific differential gene expression in collagen I-positive murine mesenchymal cells.

Project description:Type I Collagen and Foxf1 expressing double transgenic mice that were labeled for green flourescent protein (GFP) and tdTomato reporters, respectively, were used to sort out three populations. We found that mesenchymal populations which expressed both type I collagen and Foxf1 also expressed high levels of Gli and this population carried a Col14a1 fibroblast signature while the type I collagen expressing MCs that did not express Foxf1 expressed high levels of integrin alpha 8 and carried a Col13a1 fibroblast signature.

Spatially resolved multiomics of human cardiac niches - Multiome GEX

Project description:Using Multiome and previously published sc/snRNA-seq data, we studied eight anatomical regions of the human heart including left and right ventricular free walls (LV and RV), left and right atria (LA and RA), left ventricular apex (AX), interventricular septum (SP), sino-atrial node (SAN) and atrioventricular node (AVN). For the first time, we profile the cells of the human cardiac conduction system, revealing their distinctive repertoire of ion channels, G-protein coupled receptors and cell-cell interactions. We map the identified cells to spatial transcriptomic data to discover cellular niches within the eight regions of the heart.

Foxf1 enchances pulmonary inflammation and mastocytosis

Project description:The Forkhead Box f1 (Foxf1) transcriptional factor (previously known as HFH-8 or Freac-1) is expressed in endothelial and smooth muscle cells in the embryonic and adult lung. To assess effects of Foxf1 during lung injury, we used CCl4 injury model. Foxf1+/- mice developed severe airway obstruction and bronchial edema, associated with increased numbers of pulmonary mast cells and increased mast cell degranulation following injury. Pulmonary inflammation in Foxf1+/- mice was associated with diminished expression of Foxf1, increased mast cell tryptase and increased expression of CXCL12, the latter being essential for mast cell migration and chemotaxis. Foxf1 haploinsufficiency caused pulmonary mastocytosis and enhanced pulmonary inflammation following chemically-induced lung injury, indicating an important role for Foxf1 in the pathogenesis of pulmonary inflammatory responses. Carbon tetrachloride (CCl4; Sigma, St Louis, MO) was dissolved in mineral oil at a 1:20 ratio v/v and a single intraperitoneal (i.p.) injection of CCl4 (0.5 l of CCl4/ 1g of body weight) was administered to male Foxf1+/- mice or their wild type (WT) littermates as described.
