Project description:Bacillus velezensis strain GH1-13 with a native conjugative plasmid (pBV71) is thought to be beneficial to the bacterium, although no information on its effects exists. Here we show that strain GH1-13 frequently lost the plasmid during normal growth conditions in a rich medium and changed the morphology and sensitivity to selenite and tellurite. Compared to the plasmid-cured cells, the wild-type and complemented cells exhibited multicellular behavior with the expression of conjugative type IV pili and regulatory Rap homologous genes that regulate the interconnection between conjugation and biofilm formation. Further omics-based analyses of morphogenesis, biofilm formation, and antibiotic synthesis suggest that the conjugative plasmid activates envelope stress responses in association with increased biosynthesis of extracellular polysaccharide and antibiotics for protective functions of the host during exponential phase.

Project description:Plasmid fitness is directed by two orthogonal processes—vertical transfer through cell division and horizontal transfer through conjugation. When considered individually, improvements in either mode of transfer can promote how well a plasmid spreads and persists. Together, however, the metabolic cost of conjugation could create a tradeoff that constrains plasmid evolution. Here we present evidence for the presence, consequences, and molecular basis of a conjugation-growth tradeoff across 40 plasmids derived from clinical E. coli pathogens. We discover that most plasmids operate below a conjugation efficiency threshold for major growth effects, indicating strong natural selection for vertical transfer. Below this threshold, E. coli demonstrates a remarkable growth tolerance to over four orders of magnitude change in conjugation efficiency. This tolerance fades as nutrients become scarce and horizontal transfer attracts a greater share of host resources. Our results provide insight into evolutionary constraints directing plasmid fitness and strategies to combat the spread of antibiotic resistance.

Project description:P. putida KT2440 was mixed in a conjugation filter with either E. coli BW27783-NxR bearing no plasmid (NP), E. coli BW27783-NxR bearing pSU2007 (ardC+) or E. coli BW27783-NxR bearing pLGM25 (ardC-). RNA-seq results showed that: (a) R388 genes involved in conjugation are highly upregulated in the ardC+ condition with respect to the ardC- condition. (b) a number of donor E. coli genes and pathways involved in flagellar motility, SOS and stress responses and different metabolic pathways are downregulated in the ardC- condition with respect to NP or ardC+ conditions. (c) SOS genes are upregulated in recipient P. putida cells when receiving ardC+ containing plasmid with respect to NP or ardC- conditions.

Project description:Horizontal gene transfer via plasmid conjugation is a major driving force in microbial evolution. Transfer of conjugative plasmids is a complex process that needs to be synchronized with the physiological state of the bacterial host. While several host transcription factors are known to control the plasmid-borne transfer control genes, RNA-based regulatory circuits for host-plasmid communication remain unknown. Here, we describe a post-transcriptional mechanism whereby the Hfq-dependent small RNA, RprA, inhibits transfer of pSLT, the virulence plasmid of Salmonella enterica. RprA employs two different seed pairing domains to recognize and activate the mRNAs of both the sigma-factor S and RicI, a cytoplasmic membrane protein. The latter is a hitherto unknown conjugation inhibitor whose transcription requires S. Together, RprA and S constitute a feed-forward loop with AND-gate logic which tightly controls RicI synthesis for selective suppression of plasmid conjugation under membrane stress. This study reports the first sRNA-controlled feed-forward loop based on double target activation and an unexpected function for a core-genome encoded small RNA in controlling extrachromosomal DNA transfer.

Project description:Conjugative plasmids are a major driving force for the dissemination of antimicrobial resistance. During conjugation, plasmid DNA is transferred from the donor cell as a single-stranded (ss) linear molecule. Recent researches highlighted intriguing DNA reactions at the early stage of conjugation that are important for plasmid establishment in the recipient cell, including prompt expression of anti-defense genes. However, genomics-based approaches to investigate plasmid establishment have been challenging, because identical, fully established plasmids in donor cells mask transconjugant-specific signals. To overcome this limitation, we developed a new EDTA method which exploits a donor mutant hypersensitive to hypoosmotic shock. It allows unprecedently quick and efficient Elimination of Donor population for Transconjugant Accumulation. RNAseq analysis revealed highly selective and robust induction of plasmid genes during early stage of conjugation. We experimentally identified +1 sites of 6 possible operons that are matched with ssDNA promoter predictions. We also showed gene expression program is altered in different recipient cells in which plasmid establishment is perturbed. As the EDTA method is straightforward and broadly applicable, it will further understanding of the plasmid establishment processes in new host cell, not only gene expression but also ss to double-stranded DNA conversion and plasmid circularization.

Project description:The type VI secretion system (T6SS) is a highly sophisticated nanomachine widely used by bacteria to achieve competitive advantage and to potentiate horizontal gene transfer. Plasmid conjugation plays crucial roles in bacterial evolution by driving adaptation to environmental stimuli and pathogenicity. The lethal effect mediated by T6SS is detrimental to horizontal gene transfer by conjugation, while bacteria have evolved T6SS repression mechanisms regulated by plasmid to accomplish conjugative transfer. Two TetR family regulators encoded by large conjugative plasmid (LCP) in Acinetobacter baumannii have been proved similar in T6SS restriction, which seems redundant in function. Here, the global regulation roles and multiple DNA binding sites of two plasmid-sourced TetRs were identified. The two TetRs showed distinct preferences in similar roles of T6SS inhibition and binding with DNA probes. Crystal structures of TetRs were solved for illuminating the regulatory mechanism and possible reasons for difference in functions. In addition, plasmid-sourced TetRs also significantly downregulated biofilm formation and bacterial colonization, as well as influenced bacterial virulence in cultured cells and murine pneumonia infection models. Taken together, this work comprehensively elucidates the roles and regulatory mechanisms of TetRs and clarifies their similarity and difference in function, providing insights into plasmid encoded chromosome regulation pathways.

Project description:Sensory circuit activation can induce neurotransmitter respecification. To understand the consequences and mechanisms of this neuroplasticity we investigated the effects of olfactory system activation on transmitter expression in interneurons of the accessory olfactory bulb (AOB) during development. Frog larvae use olfactory-mediated kin recognition to distinguish siblings from non-siblings. Prolonged exposure to kin (sibling) or non-kin (non-sibling) odorants changed the number of neurons expressing dopamine or GABA compared to odorant deprivation (orphan condition). To identify signaling molecules mediating this behavior we performed mass spectrometry of kin-conditioned water samples. Vitellogenin-derived peptides, uniquely present in kin-conditioned samples of one genotype, were sufficient to elicit aversion behavior in non-kin larvae. RNA profiling identified AOB microRNAs (miRs) differentially regulated across conditions. Inhibition of miR-375 and miR-200b revealed that they regulate the dopaminergic and GABAergic phenotypes by targeting Pax6 and Bcl11b. Altering the ratio of dopamine/GABA AOB interneurons or locally introducing receptor blockers reversed kinship preference.

Secondary to this, the effect of regular post-exercise CWI on strength gains and post-exercise soreness was investigated. We used microarrays to detail the global effects of CWI on DNA methylation in vastus lateralis muscle tissue.
