Project description:Periodontal pathogens (Porphyromonas gingivalis, Fusobacterium nucleatum) are known as one of several important bacterial pathogens associated with sporadic Alzheimer's disease (AD). It is suggested that the entry of Periodontal pathogens into the brain is due to the disruption of the tight junctions of endothelial cells by Periodontal pathogens. However, genomic alterations that occur as a result of Periodontal pathogens infection in endothelial cells are not well understood. Our goal was to identify genes associated with Periodontal pathogens infection-induced endothelial dysfunction by integrating gene expression data.

Project description:The objectives of this investigation were to examine changes in the host transcriptional profiles during a polymicrobial periodontal pathogens Porphyromonas gingivalis, Treponema denticola, and Tannerella forsythia infection using a murine calvarial model of inflammation and bone resorption. P. gingivalis FDC 381, T. denticola ATCC 35404, and T. forsythia ATCC 43037 was injected into the subcutaneous soft tissue over the calvaria of BALB/c mice for 3 days, after which the soft tissues and calvarial bones were excised. RNA was isolated from infected soft tissues and calvarial bones and analyzed for transcript profiles using Murine GeneChip® MG-MOE430A Affymetrix arrays to provide a molecular profile of the events that occur following infection of these tissues. We used mouse microarrays to detail the molecular profile of the events that occur following infection of calvarial and bone tissues and identified distinct classes of up- and down-regulated genes during this process.

Project description:Periodontal infections have been associated with systemic inflammation and risk for atherosclerosis and vascular disease. We investigated the effects of comprehensive periodontal therapy on gene expression of peripheral blood monocytes. Approximately 1/3 of the patients showed substantial changes in expression in genes relevant to innate immunity, apoptosis, and cell signaling. We concluded that periodontal therapy may alter monocytic gene expression in a manner consistent with a systemic anti-inflammatory effect. Experiment Overall Design: Fifteen patients with periodontitis contributed with blood samples at four time points: 1 week prior to periodontal treatment (#1), at treatment initiation (baseline, #2), 6 weeks (#3) and 10 weeks post-baseline (#4). At baseline and 10 weeks, periodontal status was recorded and subgingival plaque samples were collected and processed by checkerboard DNA-DNA hybridization. Periodontal therapy, including periodontal surgery and extractions but no adjunctive antibiotics, was completed within 6 weeks. At each of the four time points, serum concentrations of 19 biomarkers were determined using multiplex assays for Luminex technology. Peripheral blood monocytes were purified, RNA was extracted, reverse-transcribed, labeled, and hybridized with Affymetrix U133 Plus 2.0 chips. Expression profiles were analyzed using linear random effects models. Further analysis of Gene Ontology (GO) terms summarized the expression patterns into biologically relevant categories. Treatment resulted in substantial improvement in clinical periodontal status and reduction in levels of several periodontal pathogens. Expression profiling over time revealed more than 11,000 probes sets differentially expressed at a false discovery rate of <0.05.

The calvarial bones and overlying soft tissues from 5 mice in each group were excised, snap frozen in liquid nitrogen, and stored at M-bM-^@M-^S80M-BM-0C until RNA isolation. Total RNA was isolated from the frozen calvarial tissue and calvarial bone from each mouse (Polymicrobial bacteria infected and control animals, N = 5 in each group) with Trizol reagent (Invitrogen, CA). Equal amounts of RNA from samples were labeled and hybridized on a mouse GeneChip following the protocol described in the GeneChip Expression Analysis Technical manual (Affymetrix, Santa Clara, CA). After hybridization, the GeneChip arrays were stained and scanned in an Affymetrix GCS 3000 7G Scanner.

Project description:Periodontitis can impair the osteogenic differentiation of human periodontal mesenchymal stem cells, but the underlying molecular mechanisms are still poorly understood. Long noncoding RNAs (lncRNAs) have been demonstrated to play significant roles under both physiologic and pathological conditions. We performed comprehensive lncRNAs profiling by lncRNA microarray to identify differentially expressed long noncoding RNA expression between Periodontal ligament stem cells from healthy Periodontal tissue and periodontal ligament stem cells from inflammatory periodontal tissue. Our analysis identified 233 lncRNAs and 423 mRNAs that were differently expressed (fold change >2.0, p-value < 0.05) between the two groups of cells. The GO analysis revealed that the significantly down-regulated biological processes included multicellular organismal process, developmental process and multicellular organismal development and the significantly up-regulated biological processes included cellular process, biological regulation and response to stimulus in periodontal ligament stem cells from inflammatory periodontal tissue. The Pathway analysis revealed that the differentially expressed mRNAs may involved in Focal adhesion, ECM-receptor interaction, Bacterial invasion of epithelial cells, Long-term depression, Circadian entrainment and HIF-1 signaling pathway. Two-condition experiment, periodontal ligament stem cells from healthy periodontal tissue (hPDLSCs) vs. periodontal ligament stem cells from inflammatory periodontal tissue (pPDLSCs), Biological replicates: 3 control replicates (hPDLSCs), 3 testing replicates (pPDLSCs).

The EA.hy926 cells were treated with normal glucose, high glucose, or high glucose plus Mathurameha for 24 h. Proteins were extracted and resolved on 12% SDS-PAGE. Coomassie blue-stained gel were excised in the whole lane and subjected to in-gel tryptic digestion and SWATH-MS.

Project description:Increasing evidences support that endothelial dysfunction initiates and drives pulmonary vascular remodeling. Targeting endothelial dysfunction is a promising treatment strategy for pulmonary artery hypertension (PAH). Therefore, it is urgent to elucidate the pathological mechanism of endothelial dysfunction. SIX1 is a critical transcription factor that regulates pulmonary vascular development. Lack of SIX1 impairs the differentiation of pulmonary vascular endothelial cells into smooth muscle cells. However, the role of SIX1 in endothelial dysfunction and PAH is unknown. In this study, we confimed that SIX1 was a critical transicription factor to induced endothelial dysfunction and PAH, and uncoverd that MAST4 was the target of SIX1-mediating endothelial dysfunction. MAST4 is a serine/threonine kinase, however the underlying mechanism by which MAST4 mediated endothelial dysfunction was unknown. Therefore, we planed to perform phosphoproteomics to explore the underlying mechanism.

Project description:Doxorubicin (DOX) has been demonstrated to induce cardiovascular toxicity in cancer survivors. Endothelial cell dysfunction is recoginized to play a critical role in the onset and severity of cardiotoxicity associated with DOX. Endothelial TFEB protects against EC damage and cardiac dysfunction. Cardiac single cells isolated from vehicle and doxorubicin treated wild type and EC-Tfeb Tg mice were collected for scRNA-seq analysis.
